Condensation with glyoxylic acid vapor, recently introduced as a highly sensitive method for fluorescence histochemical visualization of biogenic monamines, has been found to allow also the demonstration of certain tryptophan- or dopa-containing peptides. Thus, in model experiments, treatment with glyoxylic acid vapor induced fluorescence from di- and tetrapeptides with tryptophan or dopa in NH2-terminal or COOH-terminal position. Peptides without tryptophan or dopa in terminal positions gave no observable fluorescence. Differences were recorded between peptides with NH2-terminal and COOH-terminal tryptophan with respect to their fluorescence yields under different reaction conditions and the spectral characteristics of the fluorophores (excitation/emission maxima: 375-380/500-520 nm and 340-370/435 nm, respectively). In addition, the tryptophan-containing peptides could be distinguished microspectrofluorometrically from the dopa-containing peptides. Thus, the fluorophore of the NH2-terminal dopa peptide had excitation/emission maxima at 330 and 380/495 nm, and the corresponding values for the fluorophore of the COOH-terminal dopa peptide were 330 and 370/460 nm. The glyoxylic acid treatment produced intense fluorescence in certain endocrine cell systems in pituitary, thyroid and gastric mucosa, previously suggested to store peptides with NH2-terminal tryptophan. The present findings support this hypothesis.

Films of soluble ribonucleic acid (sRNA) and polyadenylic acid (poly-A) were used to investigate the distribution of nuclease activity in normal rat tissues. The reactions obtained with films of sRNA were similar to those previously observed with standard RNA and both substrates apparently reveal the same group of nucleases. On the other hand, the distribution of enzyme activity shown by films of poly-A differed markedly from that observed with RNA films, and it appears that films of poly-A demonstrate a different group of nucleases.

Vascular permeability factor (VPF)/vascular endothelial growth factor (VEGF) is a approximately 43-kDa secreted protein that has been shown in bioassays to induce endothelial proliferation, angiogenesis, and capillary hyperpermeability. VPF has been suggested to play an important role in the physiology of normal vasculature. To further elucidate the natural functions of VPF in vivo, the expression of VPF in normal tissues was examined using Northern blot analysis and in situ hybridization histochemistry. VPF mRNA is expressed in the brain, kidney, liver, lung, and spleen of the healthy adult rat. On Northern blots, the relative abundance of VPF mRNA observed in these tissues was highest in the lung and lowest in the spleen. As determined by in situ hybridization, the patterns of VPF expression are organ specific. Hybridization of an antisense VPF probe was concentrated in the cerebellar granule cell layer of the brain and in the glomeruli and tubules of the kidney. In the liver and lung, intense hybridization was observed homogeneously throughout both tissues, demonstrating that VPF mRNA is present in virtually every hepatocyte and pulmonary alveolar cell. Hybridization to the spleen was weaker and more diffuse. The widespread expression and organ-specific distribution of VPF mRNA in normal rat tissues supports the suggestion of an extensive role for this factor in the physiology of normal vasculature.

Capecitabine is a 'pro-drug' to the cytotoxic agent 5-fluorouracil (5-FU) intended to administered orally. Capecitabine is generally used as first line monotherapy for advanced colon cancer. Simple, rapid, accurate UV spectrophotometric methods were developed in the present study and validated for the estimation of Capecitabine in bulk and its formulations as per ICH guidelines. Three solvent systems viz., 0.1N NaOH, 0.1N HCl and Methanol: Water (1:3) was tried. The results suggest that the developed method shows linearity over the range of concentration 2-24μg/ml and a correlation coefficient of 0.9999. Accuracy, precision, linearity, robustness, and ruggedness were statistically validated as per ICH guidelines for all the developed methods. The % RSD values for validated methods were found to be less than 1.5 and methods will find application in routine analysis of drug formulations containing Capecitabine.
