This research reveals the existence of velvet painting in Bukittinggi. In addition, this research wants to know the potential of velvet painting and its contribution to tourism in Bukittinggi. Research on velvet painting is reviewed from the potential for the future less guarantee of its existence, considering the longer the regeneration that will preserve velvet painting, in addition to the existence of stalls where the artist displays his work is increasingly knocked out. The contribution of velvet painting is very much an influence on tourism in Bukittinggi considering the interest of tourists who buy their work as a piece or a hand from his visit to Bukittinggi. The method used in this study refers to the qualitative research methodology, which is research that is intended to understand the existence of velvet painting and its contribution to tourism in Bukittinggi and then described in accordance with the natural and scientific context. In qualitative research methods commonly used in collecting data are observation, interview and utilization of documents in the field. Then for research reports, the data collected is presented based on excerpts of data in the form of words/ sentences or data in the form of interview scripts, field notes, photos, videos, personal documents or other official documents. Keywords: velvet painting, contribution, Bukittinggi tourism. AbstrakPenelitian ini mengungkap keberadaan seni lukis beludru di Bukittinggi. Penelitian ini untuk mengetahui potensi seni lukis beludru dan kontribusinya terhadap pariwisata di Bukittinggi. Penelitian tentang seni lukis beludru ditinjau dari potensi ke depannya kurang menjamin keberadaannya, mengingat makin lama makin sedikit regenerasi yang mau melestarikan melukis beludru, di samping itu keberadaan kios tempat seniman memajang karyanya semakin tersingkir. Kontribusi karya lukis beludru sangat besar pengaruhnya terhadap pariwisata di Bukittinggi mengingat animo wisatawan yang membeli karya mereka sebagai souvenir dari kunjungannya ke Bukittinggi. Metode yang dipakai dalam penelitian ini merujuk pada metodologi penelitian kualitatif, yaitu penelitian yang dimaksudkan untuk memahami keberadaan karya lukis beludru dan kontribusinya terhadap pariwisata di Bukittinggi dan kemudian dideskripsikan   sesuai dengan   konteks   yang   alamiah dan ilmiah. Dalam penelitian kualitatif metode yang biasa digunakan dalam mengumpulkan data adalah pengamatan, wawancara dan pemanfaatan dokumen di lapangan. We developed a staining protocol for demonstration of androgen receptor (AR) in formalin-fixed, paraffin-embedded tissue sections. The method is based on the antigen retrieval microwave (MW) heating technique. Results are compared with different types of enzyme digestion pre-treatments. The strongest immunostaining signal and clearest background were obtained by MW heating of dewaxed paraffin sections in 5% urea or citrate buffer solution (pH 6); pure distilled water gave less consistent results. Enzymatic digestion with pepsin (0.05% in 2 N HCl) for 30 min at room temperature, or trypsin followed by pronase, or pronase digestion alone, also produced enhanced staining of AR in some cases, but there was more nonspecific background, and specific reactivity was less intense. The antigen retrieval MW method can be used to demonstrate AR epitope in prostate tissue after fixation in formalin for as long as 7 days. AR immunolocalization was also compared in frozen and paraffin sections processed from the same specimen of prostate carcinoma tissue and was found to be qualitatively and quantitatively similar. This study also provided new information concerning the basic principles of the antigen retrieval MW method that may be helpful in further development of this technique.

AimBreast carcinoma proliferative activity, histological grade and commercial molecular tests are all important in prognostication and treatment. There is a particular need for improved, standardised techniques for subclassification of grade 2 breast cancers into low-risk and high-risk prognostic groups. In this study we investigated whether gene expression profiling of five proliferation genes was feasible using breast cancer tissue in a clinical setting and whether these profiles could enhance pathological assessment.MethodsExpression of five proliferation gene mRNAs; Ki-67, STK 15, CCNB1, CCND1 and MYBL2, was quantified in 27 breast carcinomas and compared with Ki-67 proliferation index (PI) and Nottingham mitotic score.ResultsExpression of Ki-67, STK15 and MYBL2 mRNA showed moderate Spearman's correlation with Ki-67 PI (p<0.01), but CCND1 and CCNB1 showed weak, non-significant correlation. Individual gene expression did not associate with mitotic score but combined mRNA expression correlated with both Ki-67 PI (p=0.018) and mitotic score (p=0.03; 0.007).ConclusionsThis study confirms mRNA analysis in breast carcinoma formalin-fixed, paraffin-embedded samples is feasible and suggests gene expression profiling, using a small set of five proliferation genes, has potential in aiding histological grading or assessment of proliferative activity of breast cancers. To fully evaluate the clinical applicability of this approach, a larger cohort study with long-term follow-up data is required.
