Virgin coconut oil reverses hepatic steatosis by restoring redox homeostasis and lipid metabolism in male Wistar rats

We assessed the ameliorative effect of Virgin Coconut Oil following atrazine-induced metabolic derangement in rats. Adult male wistar rats weighing 180-200g were used; randomly separated into two major groups. Thirty-five rats in the test group were randomly divided into five sub-groups of 7 rats per sub-group and treated thus: Sub-group (SG) 1, 2 and 3 received 10ml/kg of distilled water, 10ml/kg VCO, 123mg/kg of Atrazine respectively, SG4 was diabetic control; SG 5 was the diabetic group treated with 10ml/kg of VCO for 2 weeks, after which the animals were sacrificed and blood collected for analysis. 35 rats for the recovery group were also divided into 5 sub-groups of 7 rats per sub-group and were treated; SG 1, 2, 3, 4 and 5 received 10ml/kg distilled water, 10ml/kg VCO, 123mg/kg of ATZ respectively. After 2 weeks, the animals were re-treated thus: SG 1,2,3,4 and 5 received 10ml/kg of distilled water, 10ml/kg of VCO, 123mg/kg of ATZ, 10ml/kg VCO and 10ml/kg distilled water respectively. After 2 weeks, the animals were also sacrificed and blood collected for analysis. ATZ reduced serum insulin and a reduced expression of GLUT4. VCO restored GLUT4 levels but did not significantly restore the insulin to the normal levels

Sangi1), Ni Putu Purwati2) 1)Program Studi Kimia FMIPA Universitas Sam Ratulangi, Manado 95115; 2)Alumni Program Studi Kimia FMIPA Universitas Sam Ratulangi, Manado 95115 ABSTRAK Penelitian ini bertujuan untuk mengukur tingkat oksidasi lipid dalam plasma darah tikus yang diberi pakan VCO mengandung ekstrak wortel.  Tikus Wistar jantan pada penelitian ini dibagi menjadi empat kelompok, yaitu (I) pakan standar (kontrol), (II) 10% VCO tanpa ekstrak wortel, (III) 10% VCO mengandung ekstrak wortel dan (IV) 10% minyak jagung.  Setelah 4 minggu masa perlakuan, plasma darah dipisahkan dengan sentrifugasi untuk analisis tingkat oksidasi plasma dengan mengukur konsentrasi malondialdehida (MDA) menggunakan metode uji asam tiobarbiturat.  Hasil penelitian menunjukkan bahwa konsentrasi MDA tertinggi terdapat pada kelompok tikus yang diberi minyak jagung (11,253 mM), diikuti kelompok kontrol (10,167 mM), lalu kelompok tikus yang diberi VCO mengandung ekstrak wortel (8,656 mM) dan VCO (8,269 mM).  Penelitian ini menyimpulkan bahwa tingkat oksidasi lipid plasma tikus yang diberi VCO mengandung ekstrak wortel relatif sama dengan yang diberi VCO tanpa ekstrak wortel, tetapi lebih rendah daripada kontrol dan yang diberi minyak jagung. Kata kunci: malondialdehida, minyak kelapa, wortel EFFECT OF VIRGIN COCONUT OIL (VCO) CONTAINING CARROT EXTRACT ON PLASMA LIPID PEROXIDATION ABSTRACT The aim of this study was measure the level of lipid oxidation in the blood plasma of rats fed VCO containing carrot extracts. Male Wistar rats in this study were divided into four groups: (i) control, (II) 10% VCO without the carrot extract, (III) 10% VCO containing carrot extracts, and (IV) 10% corn oil. After 4 weeks of treatment, blood from each rat was taken by syringe.  Plasma is separated from whole blood by centrifugation for analysis of plasma oxidation levels by measuring the malondialdehyde (MDA) concentration using tiobarbituric acid test. The results showed that the highest concentration of MDA was found in rats fed corn oil (11.253 m M), followed by the control group (10.167 m M), then rats fed VCO containing carrot extract (8.656 m M) and VCO (8.269 m M). This study concluded that the level of plasma lipid oxidation in rats fed VCO containing carrot extracts is relatively similar to rats fed VCO without the carrot extract, but it is lower than the control and corn oil groups. <p><strong>ABSTRACT</strong></p><p><strong>Background:</strong><em> </em><em>Virgin Coconut Oil </em>(VCO) is a functional product that is highly favored and have been widely applied in the everyday lives of society. The increase of the frequencies and duration of VCO consumption is suspected that VCO could negatively impact the target organ. The kidney is a vital organ in the human body, it is responsible for clearing out the residual waste products of metabolism in the body. The evalution of the kidney’s function can be done through the measurement of creatinine and blood urea nitrogen. The purpose of this study is to determine the effect of VCO administration on kidney function.</p><p><strong>Method</strong>: 24 male wistar rats that are divided into 4 groups, which are listed as follows, K(-) or a negative-controlled group, P(1) or the first group of rats that are given VCO with a dose of 2mL/200gr of the rats’ total BW/day, P(2) or the second group of rats that are given VCO with a dose of 3mL/200gr of the rats’ total BW/day,  and P(3) or the third group of rats that are given VCO with a dose of 4mL/200gr of the rats’ total BW/day for 28 day</p><p><strong>Results</strong>: Administrating<strong> </strong>VCO with a dose of 2mL/200gr and 3mL/200gr of the rats’ total BW/day does not affect the blood’s creatinine level while with a dose of 4mL/200gr of the rats’ total BW/day decreases the blood’s creatinine level. Administrating VCO with a dose of 2 mL/200gr, 3 mL/200gr, and 4 mL/200gr of the rats’ total BW/day does not affect the level of blood urea nitrogen.</p><p><strong>Conclusion: </strong>Administration of VCO dose of 4 mL / 200 g BW rat / day significantly decreased blood creatinine levels in experimental animals, while other doses did not affect the creatinine levels or blood urea nitrogen levels<strong></strong></p><p><strong>Keywords</strong><strong>:</strong> <em>Virgin Coconout Oil </em>(VCO), cretinine, blood urea nitrogen.</p>

Background: The aim of this study was to evaluate the nutritional effects of supplementation with virgin coconut oil (VCO) in Wistar rats over a sub-chronic period (6 weeks).Methods: Twelve Wistar rats were used and randomly assigned to receive (n = 6): control - lab chow; coconut oil (CO) - lab chow with added virgin coconut oil (20%). Food and caloric intake, weight gain, food efficiency, body mass index, femur and tibia length, bone mineral composition and blood biochemistry were evaluated.Result: The CO group showed an energy intake closed to control group. Also, the supplementation with VCO generated a decrease in total blood cholesterol as compared to the control group. However, the CO group showed accumulation of fat mass, shorter femur length and anxiogenesis in relation to the control group. These results indicate few beneficial effects from the sub-chronic use of VCO and indicate that its consumption in large quantities for long periods should be questioned.
