Optimization of Picrosirius red staining protocol to determine collagen fiber orientations in vaginal and uterine cervical tissues by Mueller polarized microscopy

We use the well established method of polarized microscopy on picrosirius red stained tissue samples and a dispersion model to quantify over 37000 measured layer-specific 3D fiber orientations of 11 human thoracic and abdominal aortas and common iliac arteries. Our results yield mean fiber directions, quantitative dispersion data and the existence of two distinct and prominent fiber families for all layers of the aortic wall.

Collagenofibrotic glomerulopathy, a collagen type III kidney disease, is associated with proteinuria and accumulation ofcollagen type III in the glomerulus specifically the mesangium and/or capillary walls. The puromcyin aminonucleoside (PAN) nephropathy model was evaluated to examine the relation between COL3A1 mRNA and proteinuria. In Wistar rats administered PAN, a robust increase in proteinuria was accompanied by glomerular hypertrophy and expansion of both the Bowman’s capsule and Bowman’s space. An ~4-fold increase in renal COL3A1 mRNA was observed in the PAN cohort with urine protein exhibiting a direct (r = 0.8) and significant correlation with kidney COL3A1 mRNA level. Both Picrosirius red polarized microscopy and immunohistochemical analysis showed localization of collagen type III to the glomerular mesangium. Gene ontology-driven transcriptomic analysis reveals a robust COL3A1 network in the glomerular compartment.

The arrangement of collagen fibers has previously been studied with picrosirius red (PSR) staining and brightfield microscopy. We discovered that PSR staining can also be visualized by fluorescence microscopy. PSR-stained collagen was strongly fluorescent using excitation and barrier filters for rhodamine, and distracting background cytoplasmic fluorescence was drastically reduced with phosphomolybdic acid (PMA) treatment before PSR staining. The PMA-PSR fluorescence method was more sensitive than the brightfield PSR or PMA-PSR method, and permitted confocal microscopic study. We applied the method to the study of collagen fiber three-dimensional arrangement in perimysial and endomysial septa of the heart, showing the three-dimensional course of the fibers in stereo views generated by confocal microscopy. The PMA-PSR fluorescence method should be generally useful for accurately determining collagen fiber three-dimensional arrangement, a necessary prelude to mechanical modeling of collagen-reinforced tissues.

Background: Dermatosparaxis is an autosomal recessive genetic disease that affects the connective tissue of animals. Collagen proteins form fibrillar structures that provide strength and structure to the extracellular matrix of tissues and organs in the body. Therefore, changes in collagen synthesis result in hyperextensibility and skin fragility. Similar to dermatosparaxis in animals, some cases of Ehlers-Danlos syndrome type VIIC have been reported in humans. The objective of this study was to describe the clinical, anatomopathological, and molecular findings of a case of dermatosparaxis in a sheep bred by crossing the Santa Inês and White Dorper breeds.Case: A case of dermatosparaxis was reported in a 20-day-old female sheep, a cross between the Santa Inês and White Dorper breeds. The sheep exhibited hyperextensibility of the skin with minimal tension and increased joint volume shortly after birth. Because of clinical worsening and the poor prognosis of the disease, the owners opted for euthanasia. The necropsy revealed large ulcerated areas in several parts of the body, including the cervical, sternal, scapular, and thoracic regions, and the inner and outer thighs. The skin was extremely easy to be removed during the necropsy and the use of a scalpel was not necessary. Extensive areas of hemorrhage were observed in the subcutaneous tissue and small intestine. The histopathological findings of the skin lesions evidenced the presence of dermatosparaxis, particularly regarding the disorganization of collagen fibers. The tests involving Masson’s trichrome staining, picrosirius red staining, and Gomori’s trichrome staining under polarized light evidence of collagen fiber dysplasia.Discussion: The diagnosis of dermatosparaxis in this study was based on clinical, anatomopathological, and molecular analysis. Molecular diagnosis was confirmed by identification of SNP c.421G>T on the ADAMTS2 gene in both the mother and the sheep under study. In humans, in addition to hyperextensibility and skin fragility, other rare clinical signs include ligamentous laxity, bone abnormalities, vascular abnormalities, uterine rupture, ocular abnormalities, and extensive areas of hemorrhage in the intestine. In this case, the humeral-radial-ulnar joints of both thoracic limbs were substantially enlarged and, when sectioned, a moderate amount of dark yellow fluid was released. Dermatosparaxis is well documented in the White Dorper sheep breed. In the present study, the genetic test revealed that the mother (Santa Inês breed) was heterozygous and phenotypically normal. All the rams in the animal herd (White Dorper breed) were phenotypically normal. Extensive hemorrhage in organs of the abdominal cavity and the high likelihood of skin rupture have been reported in other species that are affected by the disease. The histopathological findings of cutaneous lesions in this sheep agree with the descriptions in the literature on the disorganization of collagen fibers. The tests involving Masson’s trichrome staining, Gomori’s trichrome, and picrosirius red staining under polarized light were crucial for providing evidence of collagen fiber dysplasia. Even when phenotypically normal, the heterozygous sheep in the herd can produce recessive homozygous animals, which may be clinically affected, mainly, if there is a history of uncontrolled crossing in the herd.

Neurons labeled with horseradish peroxidase and reacted with diaminobenzidine (DAB) can be imaged using a confocal scanning laser microscope (CSLM) in the reflection mode. In contrast to fluorescent markers, the DAB reaction product is thought to be stable and can be observed by both light and electron microscopy. We have investigated the sensitivity of the DAB reaction product to laser irradiation, and present the spectrophotometric properties of DAB before and after exposure in the CSLM.Pyramidal neurons in slices of rat hippocampus were injected with biocytin (a biotin-lysine complex), fixed overnight in 4% paraformaldehyde, and vibratome sectioned at 75 μm. Biocytin was detected with avidin-HRP (1:200) in 0.5% Triton X-100, incubated in DAB (0.5 mg/ml) with or without 0.04% nickel ammonium sulfate (Ni), dehydrated, and imaged in a Bio Rad MRC-500 CSLM with an argon ion laser (488 and 514 nm). Spectrophotometric measurements of the soma were made on a Zeiss microspectrophotometer, as a function of laser exposure (100-1000 scans) and staining protocol.

AbstractThe cervix is responsible for maintaining pregnancy, and its timely remodeling is essential for the proper delivery of a baby. Cervical insufficiency, or “weakness”, may lead to preterm birth, which causes infant morbidities and mortalities worldwide. We used a mouse model of pregnancy and term labor, to examine the cervical structure by histology (Masson Trichome and Picrosirius Red staining), immunohistochemistry (Hyaluronic Acid Binding Protein/HABP), and ex-vivo MRI (T2-weighted and diffusion tensor imaging), focusing on two regions of the cervix (i.e., endocervix and ectocervix). Our results show that mouse endocervix has a higher proportion of smooth muscle cells and collagen fibers per area, with more compact tissue structure, than the ectocervix. With advanced gestation, endocervical changes, indicative of impending delivery, are manifested in fewer smooth muscle cells, expansion of the extracellular space, and lower presence of collagen fibers. MRI detected three distinctive zones in pregnant mouse endocervix: (1) inner collagenous layer, (2) middle circular muscular layer, and (3) outer longitudinal muscular layer. Diffusion MRI images detected changes in tissue organization as gestation progressed suggesting the potential application of this technique to non-invasively monitor cervical changes that precede the onset of labor in women at risk for preterm delivery.
