The Impact of the Entomopathogenic Fungus Conidiobolus coronatus on the Free Fatty Acid Profile of the Flesh Fly Sarcophaga argyrostoma

The chemical composition of the insect cuticle varies remarkably between species and their life stages. It can affect host resistance and substrate utilization by invading entomopathogen fungi, such as the soil fungus Conidiobolus coronatus. In this study, Sarcophaga argyrostoma flies were exposed to sporulating C. coronatus colonies for 24 h; the pupae were resistant, but the adults demonstrated 60% mortality. Although the pupae demonstrated no sign of infection nor any abnormal development, our findings indicate that after 24 h of contact with the fungus, the pupae demonstrated a 25.2-fold increase in total cuticular free fatty acids (FFAs) and a 1.9-fold decrease in total internal FFAs. Also, the cuticular FFA increased from 26 to 30, while the internal FFA class increased from 13 to 23. In exposed adults, the total mass of cuticular FFAs increased 1.7-fold, while the number of FFAs stayed the same (32 FFAs). Also, the internal FFA class increased from 26 to 35 and the total FFA mass increased 1.1-fold. These considerable differences between adults and pupae associated with C. coronatus exposure indicate developmental changes in the mechanisms governing lipid metabolism and spatial distribution in the organism, and suggest that cuticular lipids play a vital role in the defence against pathogenic fungi.

The Impact of Multispecies Probiotics on Calcium and Magnesium Status in Healthy Male Rats

Although probiotics have been discovered in numerous diseases in the last decade, there is little consensus on the relationship between probiotic properties and minerals balance and their distribution in the organism. This research aimed to evaluate the calcium (Ca) and magnesium (Mg) status in rats on a diet containing multispecies probiotics. Thirty male 10-week-old Wistar rats were selected and divided into three groups (n = 10 rats)—a group fed a standard diet (C), a group fed a low-dose of multispecies probiotics with 2.5 × 109 CFU per day (LD), and a group fed high-dose of multispecies probiotics 1 × 1010 CFU per day (HD) for 6 weeks. The results revealed that HD intake significantly increased the Ca concentration in hair and Mg concentration in femur bones. A significant positive correlation was found between calcium and magnesium levels in hair. The Ca/Mg molar ratio was lower in testicles in the groups with probiotics. In conclusion, multispecies probiotics altered the Ca concentration in hair and Mg level in femur bone, and also changed the molar ratio of these elements in testicles in male rats.

Bioaccumulation of Mineral Elements in Different Biological Substrates of Athletic Horse from Messina, Italy

The objective of this study was to evaluate the levels and the potential bioaccumulation of vanadium (V), chromium (Cr), cobalt (Co), copper (Cu), zinc (Zn), cadmium (Cd), lead (Pb), and bismuth (Bi) in horses from the industrial risk area of Sicily (Italy). Different biological substrates (whole blood, serum; tail and mane) and samples of hay, concentrate and water provided to the horses were processed by means of Thermo Scientific iCAP-Q ICP–MS spectrometer for mineral concentration. One-way analysis of variance (ANOVA) was applied to show the differences in various trace elements in the biological substrates. Pearson’s test was applied to evaluate the correlation of mineral concentrations between whole blood and serum; and tail and mane. The results showed statistical differences of tested mineral elements among biological substrates; Cr whole blood concentrations were negatively correlated with serum concentrations and a positive correlation between whole blood and serum was observed for Cd and Bi. This latter also showed a positive correlation between mane and tail. The concentrations of V, Cr, and Pb in tail with serum and whole blood samples were negatively correlated, while the concentrations of Cd in tail and serum samples were positively correlated. Minerals had a non-homogenous distribution in the organism, showing different concentrations in the biological substrates.

The Distribution of Asterosaponins, Polyhydroxysteroids and Related Glycosides in Different Body Components of the Far Eastern Starfish Lethasterias fusca

Glycoconjugated and other polar steroids of starfish have unique chemical structures and show a broad spectrum of biological activities. However, their biological functions remain not well established. Possible biological roles of these metabolites might be indicated by the studies on their distribution in the organism–producer. In order to investigate the localization of polar steroids in body components of the Far Eastern starfish Lethasterias fusca, chemical constituents of body walls, gonads, stomach, pyloric caeca, and coelomic fluid were studied by nanoflow liquid chromatography/mass spectrometry with captive spray ionization (nLC/CSI–QTOF–MS). It has been shown that the levels of polar steroids in the studied body components are qualitatively and quantitatively different. Generally, the obtained data confirmed earlier made assumptions about the digestive function of polyhydroxysteroids and protective role of asterosaponins. The highest level of polar steroids was found in the stomach. Asterosaponins were found in all body components, the main portion of free polyhydroxysteroids and related glycosides were located in the pyloric caeca. In addition, a great inter-individual variability was found in the content of most polar steroids, which may be associated with the peculiarities in their individual physiologic status.

The Position of Inhaled Chemotherapy in the Care of Patients with Lung Tumors: Clinical Feasibility and Indications According to Recent Pharmaceutical Progresses

Despite new treatment modalities, including targeted therapies and checkpoint inhibitors, cytotoxic chemotherapy remains central in the care of patients with lung tumors. Use of the pulmonary route to deliver chemotherapy has been proved to be feasible and safe in phase I, Ib/IIa and II trials for lung tumors, with the administration of drug doses to the lungs without prior distribution in the organism. The severe systemic toxicities commonly observed with conventional systemic chemotherapy are consequently reduced. However, development has failed in phase II at best. This review first focuses on the causes of failure of inhaled chemotherapy. It then presents new promising technologies able to take up the current challenges. These technologies include the use of a dry powder inhaler or a smart nebulizer with advanced drug formulations such as controlled-release formulations and nanomedicine. Finally, the potential position of inhaled chemotherapy in patient care is discussed and some indications are proposed based on the literature.

Bile lipids in rats under chronic social stress

Our experiments studied changes in lipid compound of bile of non-purebred male rats under the condition of social stress while the preparation “Korvitin” was used against the stress. Using the method of thin-layer chromatography, we determined the concentrations of phospholipids, cholesterol and its esters, free fatty acids and triglycerides in the animals’ bile, which was obtained through vivesection a day and a month after the rats were first subjected to chronic social stress (model of social defeat), and also in the bile of the animals which were treated intragastrically with “Korvitin” against the stress (1 mg/kg, 7 days). In the bile of the male rats which experienced chronic social stress the concentration of free cholesterol decreased and the content of its esters increased both immediately after the initiation of stress and after a month of exposure to stress. The concentration of free fatty acids in the bile decreased after modeling chronic social stress, but increased in liver secretion, taken a month after the animals first experienced stress. In the bile of male rats immediately after the procedure of exposing the animals to stress, the content of phospholipids decreased. Using “Korvitin” during the modeling of social stress caused decrease in the content of phospholipids in the rats’ bile and caused significant increase in the concentration of free fatty acids, triglycerides and cholesterol esters in the liver secretion. The study found significant changes in the concentration of lipids in the bile and in their distribution in the organism of male rats under the conditions of experimentally induced chronic stress. The effect of stress on the bile of rats requires further study for determining its pathogenic role.

Research on the Interaction between Biomaterials of Human Serum and Dibazol by Fluorescence Spectroscopy

The interaction of dibazol to serum was investigated by fluorescence and UV-vis spectroscopy. The fluorescence experiment results show that the fluorescence of human serum can be effectively induced by light wave. The maximum fluorescence intensity of serum is excited at 280 nm and the peak wavelength is about 334 nm. It can also be concluded that the fluorescence intensity gradually increase with the increasing excitation light wavelength. The range of the fluorescence spectroscopy of serum-dibazol system induced by excited at different wavelength is about 290 - 450 nm. The fluorescence intensity of the interaction system of dibazol-serum is significantly reduced, indicating that the fluorescence quenching of serum occurred strongly caused by dibazol and there is a new compound formed between dibazol and serum. The absorption spectra show that there is a large blue in the system of dibazol and serum, which reveal that there is a kind of new complex between dibazol and serum. It is very significance to study the interaction between dibazol and serum for understanding of drug’s toxicity and its distribution in the organism.

A Family Knockout of All Four Drosophila Metallothioneins Reveals a Central Role in Copper Homeostasis and Detoxification

ABSTRACT Metallothioneins are ubiquitous, small, cysteine-rich proteins with the ability to bind heavy metals. In spite of their biochemical characterization, their in vivo function remains elusive. Here, we report the generation of a metallothionein gene family knockout in Drosophila melanogaster by targeted disruption of all four genes (MtnA to -D). These flies are viable if raised in standard laboratory food. During development, however, they are highly sensitive to copper, cadmium, and (to a lesser extent) zinc load. Metallothionein expression is particularly important for male viability; while copper load during development affects males and females equally, adult males lacking metallothioneins display a severely reduced life span, possibly due to copper-mediated oxidative stress. Using various reporter gene constructs, we find that different metallothioneins are expressed with virtually the same tissue specificity in larvae, notably in the intestinal tract at sites of metal accumulation, including the midgut's “copper cells.” The same expression pattern is observed with a synthetic minipromoter consisting only of four tandem metal response elements. From these and other experiments, we conclude that tissue specificity of metallothionein expression is a consequence, rather than a cause, of metal distribution in the organism. The bright orange luminescence of copper accumulated in copper cells of the midgut is severely reduced in the metallothionein gene family knockout, as well as in mutants of metal-responsive transcription factor 1 (MTF-1), the main regulator of metallothionein expression. This indicates that an in vivo metallothionein-copper complex forms the basis of this luminescence. Strikingly, metallothionein mutants show an increased, MTF-1-dependent induction of metallothionein promoters in response to copper, cadmium, silver, zinc, and mercury. We conclude that free metal, but not metallothionein-bound metal, triggers the activation of MTF-1 and that metallothioneins regulate their own expression by a negative feedback loop.
