Affinity proteomic dissection of the human nuclear cap-binding complex interactome

A 5′,7-methylguanosine cap is a quintessential feature of RNA polymerase II-transcribed RNAs, and a textbook aspect of co-transcriptional RNA processing. The cap is bound by the cap-binding complex (CBC), canonically consisting of nuclear cap-binding proteins 1 and 2 (NCBP1/2). Interest in the CBC has recently renewed due to its participation in RNA-fate decisions via interactions with RNA productive factors as well as with adapters of the degradative RNA exosome. A novel cap-binding protein, NCBP3, was recently proposed to form an alternative CBC together with NCBP1, and to interact with the canonical CBC along with the protein SRRT. The theme of post-transcriptional RNA fate, and how it relates to co-transcriptional ribonucleoprotein assembly, is abundant with complicated, ambiguous, and likely incomplete models. In an effort to clarify the compositions of NCBP1-, 2- and 3-related macromolecular assemblies, we have applied an affinity capture-based interactome screen where the experimental design and data processing have been modified to quantitatively identify interactome differences between targets under a range of experimental conditions. This study generated a comprehensive view of NCBP-protein interactions in the ribonucleoprotein context and demonstrates the potential of our approach to benefit the interpretation of complex biological pathways.

The Effects of Obstructive Factors and Rewards Related to Technological Innovation on Management Performance: Focusing on Chinese Companies

Background/Objectives: This study empirically analyzed the factors relevant to the effects of Obstructive factor and rewards on Technical performance and Image, management performance.Methods/Statistical analysis: In this study, PLS, a analytic method, was used among other structural equation models. PLS is not structurally strict and helpful in analyzing models at the early phase of developing theories despite incomplete models. This study conducted online survey among Chinese companies.Findings: Rewards gave positive effects on technological performance and images. Furthermore, technological performance showed the same effects on images and performance. Images gave the same effects on business performance.Improvements/Applications: This study demonstrated the important relationship between obstructive factors and rewards, technical performance and image, which will serve as a guideline for further management performance.

Learning under Diverse World Views: Model-Based Inference

People reason about uncertainty with deliberately incomplete models. How do people hampered by different, incomplete views of the world learn from each other? We introduce a model of “ model-based inference.” Model-based reasoners partition an otherwise hopelessly complex state space into a manageable model. Unless the differences in agents’ models are trivial, interactions will often not lead agents to have common beliefs or beliefs near the correct-model belief. If the agents’ models have enough in common, then interacting will lead agents to similar beliefs, even if their models also exhibit some bizarre idiosyncrasies and their information is widely dispersed. The CBC has come under renewed investigative interest in recent years due to its participation in RNA-fate decisions via interactions with RNA productive factors as well as with adapters of the degradative RNA exosome - including the proteins SRRT (a.k.a. ARS2) and ZC3H18, and macromolecular assemblies such as the nuclear exosome targeting (NEXT) complex and the poly(A) exosome targeting (PAXT) connection. A novel cap-binding protein, NCBP3, was recently proposed to form an alternative, non-canonical CBC together with NCBP1, and to interact with the canonical CBC along with the protein SRRT. The theme of post-transcriptional RNA fate, and how it relates to co-transcriptional ribonucleoprotein assembly is abundant with complicated, ambiguous, and likely incomplete models. In an effort to clarify the compositions of NCBP1-, 2-, and 3-related macromolecular assemblies, including their intersections and differences, we have applied an affinity capture-based interactome screening approach, where the experimental design and data processing have been modified and updated to identify interactome differences between targets under a range of experimental conditions, in the context of label-free quantitative mass spectrometry. This study generated a comprehensive view of NCBP-protein interactions in the ribonucleoprotein context and demonstrates the potential of our approach to benefit the interpretation of complex biological pathways.

Impact of re-scaled macro models for GNSS orbit determination

<p>Radiation pressure from the sun and the Earth are not negligible forces for orbit determination. Most analysis centers use empirical approaches (i.e., the classical Empirical CODE Orbit Model ECOM or the adapted version ECOM2), analytical approaches (simplified box wing models or detailed macro models), or a combination of both to account for the solar radiation pressure (SRP) and Earth radiation (albedo).</p><p>The European GNSS Agency and the Cabinet Office, Government of Japan, disclosed meta data for Galileo resp. QZSS satellites. For the other GNSS constellations either no or incomplete models are available. This is in particular problematic for SRP modeling. In case of albedo the issue is less critical as the corresponding force acting on GNSS satellites is much smaller compared to the SRP. Our approach to overcome this issue is to re-scale the existing box-wing models based on long term analysis (one year) and introduce the re-scaled macro models for the final orbit determination. The main goal is to achieve better orbit quality with fewer empirical parameters compared to our traditional solution based on pure empirical SRP models.  The key factors to determine the quality of the re-scaled macro models are, among others, the evaluation of the resulting orbits and the behavior of the long term time series of the corresponding geodynamic parameters.</p>

Optimal consumption of multiple goods in incomplete markets

We consider the problem of optimal consumption of multiple goods in incomplete semimartingale markets. We formulate the dual problem and identify conditions that allow for the existence and uniqueness of the solution, and provide a characterization of the optimal consumption strategy in terms of the dual optimizer. We illustrate our results with examples in both complete and incomplete models. In particular, we construct closed-form solutions in some incomplete models.
