Does Pharmacodynamics of Drugs Change After Presenting them as Nanoparticles Like their Pharmacokinetics?

: Nowadays, the breakthrough in different medical branches makes it feasible to designate new methods of drug delivery to achieve the most cost-effective and the least unpleasant consequenceimposing solutions to overcome a wide range of diseases. : Nanoparticle (NP) drugs entered the therapeutic system, especially in cancer chemotherapy. These drugs are quite well-known for two traits of being long-acting and less toxic. For a long time, it has been investigated how NPs will change the kinetics of drugs. However, there are a few studies that inclined their attention to how NPs affect the dynamics of drugs. In this review, the latter point will mainly be discussed in an example-based manner. Besides, other particular features of NPs will be briefly noted. : NPs are capable of affecting the biologic system as much as a drug. Moreover, NPs could arise a wide variety of effects by triggering their own receptors. NPs are able to change a receptor function and manipulate its downstream signaling cascade.

Conception of Biologic System: Basis Functional Elements and Metric Properties

A notion of biologic system or just a system implies a functional wholeness of comprising system components. Positive and negative feedback are the examples of how the idea to unite anatomical elements in the whole functional structure was successfully used in practice to explain regulatory mechanisms in biology and medicine. There are numerous examples of functional and metabolic pathways which are not regulated by feedback loops and have a structure of reciprocal relationships. Expressed in the matrix form positive feedback, negative feedback, and reciprocal links represent three basis elements of a Lie algebra sl(2,ℝ) of a special linear group SL(2,ℝ). It is proposed that the mathematical group structure can be realized through the three regulatory elements playing a role of a functional basis of biologic systems. The structure of the basis elements endows the space of biological variables with indefinite metric. Metric structure resembles Minkowski's space-time (+, −, −) making the carrier spaces of biologic variables and the space of transformations inhomogeneous. It endows biologic systems with a rich functional structure, giving the regulatory elements special differentiating features to form steady autonomous subsystems reducible to one-dimensional components.

Na delivery and ENaC mediate flow regulation of collecting duct endothelin-1 production

Collecting duct (CD) endothelin-1 (ET-1) is an important autocrine inhibitor of Na and water transport. CD ET-1 production is stimulated by extracellular fluid volume expansion and tubule fluid flow, suggesting a mechanism coupling CD Na delivery and ET-1 synthesis. A mouse cortical CD cell line, mpkCCDc14, was subjected to static or flow conditions for 2 h at 2 dyn/cm2, followed by determination of ET-1 mRNA content. Flow with 300 mosmol/l NaCl increased ET-1 mRNA to 65% above that observed under static conditions. Increasing perfusate osmolarity to 450 mosmol/l with NaCl or Na acetate increased ET-1 mRNA to ∼184% compared with no flow, which was not observed when osmolarity was increased using mannitol or urea. Reducing Na concentration to 150 mosmol/l while maintaining total osmolarity at 300 mosmol/l with urea or mannitol decreased the flow response. Inhibition of epithelial Na channel (ENaC) with amiloride or benzamil abolished the flow response, suggesting involvement of ENaC in flow-regulated ET-1 synthesis. Aldosterone almost doubled the flow response. Since Ca2+ enhances CD ET-1 production, the involvement of plasma membrane and mitochondrial Na/Ca2+ exchangers (NCX) was assessed. SEA0400 and KB-R7943, plasma membrane NCX inhibitors, did not affect the flow response. However, CGP37157, a mitochondrial NCX inhibitor, abolished the response. In summary, the current study indicates that increased Na delivery, leading to ENaC-mediated Na entry and mitochondrial NCX activity, is involved in flow-stimulated CD ET-1 synthesis. This constitutes the first report of either ENaC or mitochondrial NCX regulation of an autocrine factor in any biologic system.

Out with the Old, In with the New

Temporal resolution has long been a challenge to microscopists. Certainly, spatial resolution has occupied center stage, but we're all concerned about what happens over time in a biologic system, for example, a cell. Tags such as green fluorescent protein (GFP) have been used with confocal microscopy and other light microscopic techniques to achieve outstanding temporal resolution, but good spatial and temporal resolution have proven to be difficult to achieve simultaneously. This has been accomplished in a remarkable study by Guido Gaietta, Thomas Deerinck, Stephen Adams, James Bouwer, Oded Tour, Dale Laird, Gina Sosinsky, Roger Tsien, and Mark Ellisman, who demonstrated a pulse-chase technique that correlates with both fluorescence and electron microscopy.
