Scalable in situ single-cell profiling by electrophoretic capture of mRNA

Methods to spatially profile the transcriptome are dominated by a trade-off between resolution and throughput. Here, we developed a method named EEL FISH that can rapidly process large tissue samples without compromising spatial resolution. By electrophoretically transferring RNA from a tissue section onto a capture surface, EEL speeds up data acquisition by reducing the amount of imaging needed, while ensuring that RNA molecules move straight down towards the surface, preserving single-cell resolution. We applied EEL on eight entire sagittal sections of the mouse brain and measured the expression patterns of up to 440 genes to reveal complex tissue organisation. Moreover, EEL enabled the study of challenging human samples by removing autofluorescent lipofuscin, so that we could study the spatial transcriptome of the human visual cortex. We provide full hardware specification, all protocols and complete software for instrument control, image processing, data analysis and visualization.

Aesthetic Preference in the Transverse Orientation of the Occlusal Plane in Rehabilitation: Perspective of Laypeople and Dentists

The present study had a convenience sample with 236 laypeople and 242 dentists who completed an online questionnaire to choose the most attractive image among six pairs for comparison. Control image: symmetric (parallelism between occlusal plane (OP), commissural line (CL), and interpupillary line (IL)). Change of Control, obtaining three images with a 3-degree inclination of the labial commissures. Image A: OP parallel to IL; Image B: OP parallel to CL; Image C: OP at 1.5 degree mean angulation between IL and CL. Non-parametric comparison (IBM© SPSS Statistics vs. 27.0, p < 0.05). The “Dentists” group’s decreasing order of preference (attractiveness) of the images is: Control > A > C > B (p < 0.05). In the “Lay” group, it is: Control > A > (C not ≠ B). Dentists significantly prefer more the Control and Image A than laypeople (p < 0.001). Sex (single exception in laypeople), age, and dentist’s area of activity did not interfere in the perception of attractiveness. Dentists and laypeople preferred the Control when compared to images with CL canted. In the existence of CL inclination, the preference of the groups was the IL as a reference for OP orientation, with the mean angulation or coincident with the CL being considered less aesthetic.

Research on Intelligent Control and Integration Technology for Visual Inspection of TEC Components

Technical schemes such as uniform TEC component feeding, posture adjustment, multi-station positioning, and communication between upper and lower machines were designed. The opto-electromechanical integration mechanism was integrated by software to realize the detection of TEC component surface defects, and qualified TEC components were separated from unqualified TEC components by blow sieving. Adopt ARM MCU for real-time control; Image recognition adopts the machine vision deep learning platform based on linux to learn and recognize the TEC components, and sends the results to the lower computer for sieving. The accuracy of triggering the camera to take pictures is 99.9%, which provides a guarantee for capturing and processing images accurately. The accuracy of blow sieving is over 99.9%, which can meet the sieving requirements. The electrical control system of the whole machine can meet the motion control requirements of detection.

Neural correlates of the improvement of cognitive performance resulting from enhanced sense of competence: A magnetoencephalography study

The alterations in neural activity related to the improvement of cognitive performance, which would be leading to better academic performance, remain poorly understood. In the present study, we assessed neural activity related to the improvement of task performance resulting from academic rewards. Twenty healthy male volunteers participated in this study. All participants performed four sessions of a 1-back-Stroop task under both target and control conditions. An image indicating that the task performance of each participant was above average and categorized as being at almost the highest level was presented immediately after each session under the target condition, whereas a control image did not indicate task performance. Neural activity during the 1-back-Stroop task was recorded by magnetoencephalography. The correction rate of the 1-back-Stroop task in the final session relative to that in the first under the target condition was increased compared with the control condition. Correlation analysis revealed that the decreases in alpha band power in right Brodmann’s area (BA) 47 and left BA 7 were positively associated with the increased correction rate caused by the target condition. These findings are expected to contribute to a better understanding of the neural mechanisms underlying the improvement of cognitive performance.

User engagement analysis of visual vs text-based summaries of new medical research in the general medical community (Preprint)

BACKGROUND In the digital age of publication and era of social media, there is increased need for precise and concise reporting of medical manuscripts to educate students, physicians, and other health professionals. Both visual abstracts (VAs) and textual reports (TRs) have been utilized online, but there is a paucity of study about whether VAs really boost user engagement through visual representation of key study findings. Furthermore, prior studies have focused on specialty journals content as opposed to the general medical community. OBJECTIVE The current study was designed to analyze the effectiveness of standardized VAs across the general medical community. METHODS In this prospective case-control study, we used Twitter to publish 18 pairs of VAs and TRs from an open-access, physician-run medical news organization with healthcare professional readership across all medical specialties in order to investigate the effect of VAs on user engagement. Each VA/TR pair, which generally covered new research applicable to the general internist, was published on the same day, covered the same content, and had the same title on Twitter; posts for TRs had a nonspecific control image and link to a text abstract, while VA posts had a partial preview and link to the full VA. The outcomes studied were views, engagement rate, and percent change in engagement rate. RESULTS Results showed that while there was no difference in number of views on Twitter (P=.83), VAs had significantly higher engagement rates (P=.002), with an average fold change of 2.75 (95% CI 1.83 to 3.67). CONCLUSIONS Our results show that Twitter posts containing previews and links to VAs had more user engagement than posts with no visual content and links to TRs, which suggests that VAs are more effective tools for promoting engagement with medical content.
