Антимикробная активность некоторых экстрактов лишайников

Cladonia rangiferina — кустистый лишайник из рода кладония, имеющий мультирегиональное распространение, в качестве основного биологически активного вещества содержит усниновую кислоту. Из изученных экстрактов Cladonia rangiferina биологически активными являются только глицериновые экстракты, полученные при температуре экстракции 40 °C, и только в отношении грамположительных микроорганизмов (Staphylococcus aureus) и грибов (Candida albicans). Увеличение температуры экстракции до 90 °C уменьшает антимикробную активность экстрактов. Настоящее исследование демонстрирует, что лишайник может быть богатым источником эффективных противомикробных веществ.

Transcriptome Comparison of Secondary Metabolite Biosynthesis Genes Expressed in Cultured and Lichenized Conditions of Cladonia rangiferina

Lichen secondary metabolites are natural products of high medicinal and industrial value, which are produced by the fungal symbiont (mycobiont) of lichens in response to environmental changes. It has been shown that the cultured mycobiont is capable of secondary metabolite production, specifically polyketides, and polyketide production is affected by the presence or absence of the algal or cyanobacterial symbiont (photobiont). Identification of polyketide synthases encoding genes is, in turn, key for understanding the regulation of secondary metabolite synthesis. Using a previously established method of resynthesis for Cladonia rangiferina as well as the sequenced and assembled genome of that species, we compared transcriptomes of C. rangiferina cultured alone and resynthesized with the photobiont (Asterochloris glomerata) to reveal transcriptionally active genes in secondary metabolic gene clusters, as well some of the neighbouring genes, induced by the presence of the photobiont and events of lichenization. The results identify potential candidates for PKS genes in C. rangiferina, identify potential neighbouring genes in the PKS cluster, and offer insights into further research. The study provides preliminary insights into the activity of several identified biosynthetic gene clusters (BGC) as well as interactions of genes within those clusters.

Identifikasi dan Karakteristik Habitat Jenis Lumut Kerak di Taman Wisata Alam Sicikeh-Cikeh Kabupaten Dairi Sumatera Utara

Lumut kerak adalah hasil simbiosis dari fungi dan alga. Penelitian ini dilakukan untuk mengetahui jenis-jenis lichen di Taman Wisata Alam Sicikeh-cikeh Kabupaten Dairi Sumatera Utara. Metode yang digunakan merupakan metode survey eksploratif yaitu menjelajahi sepanjang jalur lokasi penelitian. Pada penelitian ini ditemukan 19 jenis yaitu lichen dari 7 famili. 8 jenis dari famili yaitu Parmeliaceae (<em>Parmotrema arnoldii</em> (Du Rietz) Hale, <em>Usnea dasypoga</em> (Acharius.) Nyl., <em>Usnea barbata</em> (L.) Weber ex F.H.Wigg., <em>Cetrelia olivetorum</em> (S.) Sticto., <em>Hypogymnia physode</em>s (L.) Nyl., <em>Bulbothrix tabacina</em> (Mont.) Hale, <em>Parmotrema perlatum</em> (Huds.) M. Choisy and <em>Parmelia sulcata</em> (Taylor.), 4 jenis dari famili Cladoniaceae yaitu <em>Pseudevernia cladonia</em> (Tuck.) W. L. Culb., <em>Cladonia rangiferina</em> (L.) Weber ex F.H.Wigg., <em>Cladonia cristatella</em> (Tuck.), 3 jenis dari famili Lobariaceae yaitu <em>Sticta sylvatica</em> (Hudson). Ach., <em>Pseudocyphellaria anthraspis</em> (Ach.) H. Magn. dan <em>Sticta crocodia</em> (Ach). Masing-masing 1 jenis dari famili Physciaceae (<em>Heterodermia leucomalos</em> (L.) Kalb.), Pertusariaceae (<em>Ochrolichia subviridi</em> (Hoeg) Erichsen.), Trentepohliaceae (<em>Trentopohlia flava</em> (Kutzing) De Toni.), and Pyrenulaceae (<em>Pyrenula sulcata</em> Eschw.). Karakteristik habitat Lumut kerak yang di temukan memiliki karakteristik habitat dengan menempel di pohon, hidup di serasah dan tanah dengan tipe talus Crustose 2 jenis dengan ciri talus berbentuk mirip kulit yang menempel ketat pada substratnya, tipe talus Foliose 8 jenis dengan karakteristik tumbuh seperti semak dan tipe talus Fruticose 9 jenis dengan ciri berwarna hijau dengan struktur permukaan halus.

Distribution and habitat preference of protected reindeer lichen species (Cladonia arbuscula, C. mitis and C. rangiferina) in the Balaton Uplands (Hungary)

The maintenance of protected lichen species and their biodiversity in general depends on good management practices based on their distribution and habitat preferences. To date, 10 of the 17 protected lichen species of Hungary have been recorded in the Bakony Mts including the Balaton Uplands region. Habitat preferences of three protected Cladonia species (C. arbuscula, C. mitis and C. rangiferina) growing on underlying rocks of red sandstone, basalt, Pannonian sandstone and gravel were investigated by detailed sampling. We recorded aspect, underlying rock type, soil depth, pH and CaCO3 content, habitat type (as defined by the General National Habitat Classification System Á-NÉR), all species of lichen, bryophyte and vascular plants as well as percentage cover of exposed rock, total bryophytes, lichens, vascular plants and canopy, degree of disturbance and animal impacts. Sporadic populations of these species mostly exist at the top of hills and mountains in open acidofrequent oak forests, but they may occur in other habitats, such as closed acidofrequent oak forests, slope steppes on stony soils, siliceous open rocky grasslands, open sand steppes, wet and mesic pioneer scrub and dry Calluna heaths. Cladonia rangiferina was found to grow beneath higher canopy cover than either C. arbuscula or C. mitis in the Balaton Uplands. Furthermore, there were significant differences in canopy cover between occupied and unoccupied quadrats in the case of all three species. Cladonia rangiferina is a good indicator species of natural habitats in Hungary due to its restricted distribution and low ecological tolerance. These results may lead to the adoption of effective conservation methods (e.g. game exclusion, artificial dispersal) in the future.

Види рослин і лишайників Кий-острова в Гербарії Національного лісотехнічного університету України (LWFU)

Кий-острів, що розташований в Онезькій затоці Білого моря (географічні координати 64°00′ п. ш., 37°53′ с. д.) – це виступ гранітної плити, яка залягає на незначній глибині і виходить на поверхню моря у вигляді скель. Геоморфологічні та кліматичні особливості острова, близькість до Північного полярного кола та Арктичної області, зумовили формування на ньому своєрідних природних комплексів, де переважають соснові ліси. Виконані в липні 1984 р. серії описів лісових біоценозів острова дали змогу встановити як основні типи цих лісів (сосняк лишайниковый скальный, сосняк лишайниковый каменистый, сосняк лишайниковый багульниково-вересковый на вершинах сельг [Kucherov, Zverev, 2012; Rupestrian landscapes…, 2008; Rysin, 1975]), так і відповідні їм типи асоціацій (Pinetum cladinosum petraeum, Myrtillus-Calluna-Cladina-Typ, Pinetum uliginosi-vaccinioso-ledosum). Наведений перелік домінантних видів рослин і лишайників нижніх ярусів лісових біоценозів демонструє наявність у їх складі характерних представників бореальної та гіпоарктичної флори. Найбільша частина з них – це класичні бореальні види: Andromeda polifolia L., Arctostaphylos uva-ursi (L.) Spreng., Chamaedaphne calyculata (L.) Moench, Ledum palustre L., Linnaea borealis L., Nardus stricta L. та інші. На другій позиції знаходяться гіпоарктичні види, серед яких Betula nana L., Rubus arcticus L., Rubus chamaemorus L., Cornus suecica L. На поверхнях скель високу частку складають представники відділу Lichenomycota – характерні елементи північних та південних лишайникових тундр, а саме: Cladonia stellaris (Opiz) Pouzar & Vězda, Cladonia sylvatica (L.) Hoffm., Cladonia rangiferina (L.) Weber ex F.H.Wigg., Cetraria islandica (L.) Ach., Peltigera aphthosa (L.) Willd. Зібраний гербарний матеріал (45 зразків 16-ти таксонів) достатньою мірою відображає найбільш характерні риси біоти скельних сосняків Кий-острова та являє собою єдину колекцію видів з північної тайги європейської частини Росії в Гербарії Національного лісотехнічного університету України, який офіційно зареєстрований в міжнародних базах даних під акронімом LWFU.

STREPTOMYCES AVERMITILIS: COMPONENT COMPOSITION, PROPERTIES

The component composition of S. avermitilis biomass was studied, including avermectin, lipids, proteins, and a polysaccharide complex (murein). It was established that the composition of lipids of S. avermitilis contains predominantly saturated fatty acids – 86%, the content of monounsaturated (palmitic and vaccine) and polyunsaturated (linoleic and α-linolenic) fatty acids is the same and is about 7%. It should be noted the presence of a significant amount of branched chain fatty acids (isoforms) – more than 70% of the total amount of fatty acids. A technique has been developed for isolating the polysaccharide complex from S. avermitilis biomass. Using IR spectroscopy, it was found that the S. avermitilis polysaccharide complex, as well as the chitin-glucan complex isolated from the thallus of the epigeneic lichen of the species Cladonia rangiferina, is built from chitin units – β-N-acetylglucosamine. It was shown that the polysaccharide complex (murein) has a polyampholytic nature and has high sorption properties with respect to the main (methylene blue) and acid (Congo red) dyes. It was also established that the sorption capacity with respect to the dyes under study for the polysaccharide complex S. avermitilis is 2.8–7.6 times higher than for the chitin – glucan complex Cladonia rangiferina. This indicates a significantly higher content of active functional groups (sorption centers) in the murein structure, which allows us to recommend it for use as an effective sorbent (enterosorbent).
