Abstract Abstract text Some studies have discussed the use of artificial intelligence and machine learning in the assessment and selection of embryos for in vitro fertilization. Complete artificial intelligence acquired using CNN’s dark box algorithm could be highly useful in assessing in embryos, though it could be difficult to perform the external validation necessary to confirm its value. But due to the inherent drawbacks in complete artificial intelligence assessing in vitro developmental embryos, such as lacking results of discard embryos, dislocations between computer scientist and embryologist, low explanatory values in dark box algorithm, here, we suggest training computers to recognize the target region (internal pellucid zone region) and the features of embryos, then continuously score the embryos starting at in vitro fertilization through the zygote to the blastocyst stage. Parameters suitable for use with various endpoints in treatment sequence could be found by AI. Further clinical studies should be performed to validate the parameters and AI needed. In fishes, gonad morphology is an important parameter to understand the reproductive biology, phylogenetic relationships and systematic studies. The main objective of this study was to make a comparative analysis of the morphology of mature ovary and testis in six fish species of the family Anostomidae. A total of 212 specimens, presenting maturing gonads, were captured from the São Francisco River, Três Marias Reservoir area, in Southeastern Brazil, between August 2008 and December 2010. The six analyzed species had mature ovaries and showed a grayish color. In vitellogenic oocytes (VO), cortical alveoli (CA) were composed by small vesicles in all Leporinus species, but in Leporellus vittatus and Schizodon knerii the CA were large vesicles. However, the CA of all species showed similar histochemical content. The pellucid zone consists of two layers to all species, but it was thicker in S. knerii (11.5 ± 1.8 µm) than in L. vittatus (9.0 ± 0.8 µm) and Leporinus species (3.5 ± 0.6 - 8.7 ± 0.9 µm). Follicular cells of the VO were respectively cubic and prismatic in animal and vegetative poles of S. knerii (22.3 ± 3.2 and 61.1 ± 9.6 µm), and squamous in the other species (1.3 ± 0.3 - 1.6 ± 0.3 µm). Females of S. knerii and males of L. vittatus showed lower values of gonadosomatic index (GSI) than the other assessed species. All evaluated species presented testicular morphology similar to most neotropical Characiformes, with unrestricted spermatogonial testis and anastomosing tubular organization of seminiferous tubules. Phenotypical differences in the vitellogenic oocyte of Anostomidae fishes, confirmed the taxonomic position of S. knerii as different genus in relation to Leporinus and L. vittatus. Despite being placed in different genera, the characteristics of the vitellogenic oocytes of L. vittatus were similar to those found in the studied Leporinus.

<p>This study aimed to evaluate the efficacy of adding sucrose in vitrification solution of ovine embryos produced <em>in vivo</em>. Forty Dorper ewes were selected and superovulated. Immediately prior to the embryo collection by laparotomy, a laparoscopy was performed to verify the superovulatory response. The recovered flushing was followed by embryo evaluation and embryos were divided in two experimental groups where embryos from Control group were submitted to a traditional vitrification protocol and embryos from Sucrose group to a modified vitrification protocol with sucrose. After warming, embryos were again divided regarding cryoprotectant removal (Indirect) or not (Direct). The embryo quality was classified as embryos of degrees I (excellent or good), II (regular), III (poor) and IV (dead or degenerate). It was also verified the homogeneity of mass, occurrence of embryonic mass retraction and rupture of pellucid zone. The results were expressed as percentages and were subjected to Chi-square test with P &lt; 0.05. The embryos vitrified in the presence of sucrose had lower proportions of lower-quality embryos after warming (22.20 vs. 44.50%), higher percentages of homogeneous embryos after warming (63.89 vs. 38.89 %) while concerning other parameters there was no difference between these groups. It can be concluded that the addition of 0.4 M sucrose during vitrification improves the embryo quality.</p>
