Rumen (Calicophoron/Paramphistomum spp.) and Liver Flukes (Fasciola hepatica) in Cattle—Prevalence, Distribution, and Impact of Management Factors in Germany

This study was carried out to determine the prevalence of rumen flukes on German cattle farms via the sedimentation technique, and to identify the rumen fluke species occurring in Germany. Additionally, the prevalence of patent Fasciola hepatica infections was determined. Furthermore, a short questionnaire was answered by the farmers. A prevalence of 5.5% and 9.5% was detected for rumen flukes and liver flukes, respectively. Coinfections occurred on 2.1% of farms. In northern Germany, the rumen fluke prevalence was higher than in southern Germany, while for liver fluke the distribution was reversed. Rumen flukes were mostly identified as Calicophoron daubneyi, but in four cases, sequencing revealed Paramphistomum leydeni for the first time in Germany. Grazing and feeding of fresh grass, as well as organic farming, were significantly associated with rumen and liver fluke occurrence. In contrast, suckler cow husbandry only had an influence on the occurrence of rumen flukes, but not liver flukes. Trematode eggs could be detected in both, farms with and without deworming. Since there were only a few studies about Paramphistomidosis in Germany, more attention should be paid to these parasitic diseases for animal welfare and animal health reasons.

Paramphistomidose – eine Übersicht zu Epidemiologie und klinischer Symptomatik

ZusammenfassungDie Paramphistomidose ist eine weltweit auftretende parasitäre Erkrankung bei diversen Wiederkäuern, ausgelöst durch verschiedene Pansenegelarten (u. a. Paramphistomum cervi, Calicophoron daubneyi und Paramphistomum leydeni). In Europa sind lokale Vorkommen von Pansenegelinfektionen bei Haus- und Wildwiederkäuern seit Jahrzehnten beschrieben. Mittlerweile gibt es Hinweise, dass die Paramphistomidose an Bedeutung gewinnt, denn in Großbritannien, Irland, Frankreich, Spanien, Belgien und den Niederlanden wurden teilweise hohe Prävalenzen festgestellt. Aktuelle Prävalenzdaten aus Deutschland liegen nicht vor. In neueren Untersuchungen in Norddeutschland, Hessen und Bayern konnte Calicophoron daubneyi, die auch in Europa aktuell am häufigsten vorkommende Pansenegelart, nachgewiesen werden. Die Entwicklung des Pansenegels ist an Zwischenwirte aus der Familie der Wasserlungenschnecken gebunden. Calicophoron daubneyi und der Große Leberegel Fasciola hepatica teilen im Laufe ihrer Entwicklung den gleichen Zwischenwirt, die Zwergschlammschnecke Galba truncatula. Der Endwirt nimmt die infektiösen Metazerkarien auf. Im Dünndarm schlüpfen die jungen Pansenegel und heften sich im Duodenum an. Danach wandern sie in den Pansen ein, wo sie als Adulte anfangen, Eier auszuscheiden. Die Infektion kann während der intestinalen Phase zu schwerem Durchfall und bei hoher Befallsintensität zum Tod führen. Die ruminale Paramphistomidose verläuft in den meisten Fällen subklinisch. Aktuell steht zur Diagnostik der koproskopische Einachweis mittels Sedimentationsverfahren zur Verfügung. Aufgrund der Ähnlichkeit besteht die Gefahr der Verwechslung mit Eiern des Großen Leberegels. Die Paramphistomidose kann mit Oxyclozanid behandelt werden. Über die Wirksamkeit anderer Wirkstoffe liegen widersprüchliche Ergebnisse vor. Umso wichtiger erscheint die Prophylaxe dieser Parasitose. Bekämpfungsvorschläge richten sich aufgrund der ähnlichen Epidemiologie nach den Empfehlungen, die zur Vorbeugung der Fasziolose abgegeben werden. Ob es sich bei der Paramphistomidose auch in Deutschland um eine sich ausbreitende Infektionskrankheit handelt, kann aktuell noch nicht beurteilt werden.

Molecular identification of the rumen flukes Paramphistomum leydeni and Paramphistomum cervi in a concurrent infection of the red deer Cervus elaphus

Paramphistomosis, caused by paramphistomid flukes, is a gastrointestinal parasitic disease of domestic and wild ruminants. Originally thought to be limited to the tropics and subtropics, the disease has recently been reported in temperate regions. Here we describe the concurrent infection of a red deer doe (Cervus elaphus) with Paramphistomum leydeni and Paramphistomum cervi. This is the first report of P. leydeni in Croatia. Flukes were identified on the basis of morphological keys (tegumental papillae) and sequencing of the internal transcribed spacer region 2 in ribosomal DNA. Our results confirm that the absence of tegumental papillae allows P. cervi to be differentiated morphologically from other paramphistomid species in Europe based on incident light stereomicroscopy. Nevertheless the limitations of morphological identification and taxonomic issues suggest that previous findings on paramphistomid infection should be interpreted carefully. The possible worldwide distribution of these pathogens means that paramphistomosis may be more common and its economic impact greater than previously thought.

First report of Paramphistomum leydeni Näsmark, 1937 (Trematoda: Paramphistomidae) in Argentina, and re-examination of Cotylophoron cotylophorum sensu Racioppi et al. (1994)

Cotylophoron cotylophorum and Balanorchis anastrophus are the only amphistomes of ruminants that have been described so far in Argentina. Recent observations suggest that C. cotylophorum could have been misclassified. To confirm this, we examined specimens, collected in Corrientes Province and classified by Racioppi et al. (1994) as Cotylophoron cotylophorum, and re-classified these as Paramphistomum leydeni Näsmark, 1937. Flukes collected from Zárate (Buenos Aires) were also studied and compared against the former, resulting in the same species. The present study reports the existence of P. leydeni in Argentina, modifies the previous classification of C. cotylophorum and suggests that, based on the evidence of the amphistomes described so far, P. leydeni would be the most prevalent species in cattle of Argentina.

Notable seasonal variation observed in the morphology of the reindeer rumen fluke (Paramphistomum leydeni) in Finland

Although numerous Paramphistomum species have been described from the rumen and reticulum of domestic and wild ruminants, information about rumen flukes in reindeer is sparse and their nomenclature is somewhat conflicting. Rumen fluke of reindeer is usually referred to as P. cervi, but P. leydeni and Cotylophoron skriabini are also mentioned in the literature. Here, the surface structures and internal anatomy of rumen flukes from reindeer, as seen by scanning electron microscopy (SEM) and in histological sections under light microscopy, are presented. The aim of the study was to find morphological information to enable identification of rumen flukes in reindeer to species level. In addition, the morphology of rumen flukes collected in winter (winter flukes) was compared with that of flukes collected in summer (summer flukes). Key morphological findings were as follows: the acetabulum of the rumen flukes was of paramphistomum type, the pharynx of liorchis type, and the genital atrium of leydeni type. Both winter and summer flukes shared these morphological features. Based on these findings, it was concluded that rumen flukes of reindeer in Finland belonged to the species P. leydeni. Significant morphological variation was observed when winter and summer flukes were compared. The winter fluke was smaller in size, possessed immature gonads (testes, ovary, uterus), and immature accessory genital glands (Mehlis’ gland, vitelline follicles), and had barely discernible tegumental papillae. These data indicate that winter rumen flukes represent an immature stage of P. leydeni and summer flukes the mature stage of the same species. Further, these findings suggest that the rumen flukes of reindeer during wintertime in Finland have a slowed or inhibited lifecycle.Abstract in Finnish / Yhteenveto:Poron pötsimadon (Paramphistomum leydeni) morfologiassa esiintyy selvää vuodenaikaisvaihtelua Pötsimatoja (Paramphistomum spp.) löytyy monien villien ja kotieläiminä pidettävien märehtijöiden pötsistä tai verkkomahasta. Poron pötsimadosta on saatavilla varsin niukasti tietoa, ja lähteestä riippuen poron pötsimadon esitetään yleensä kuuluvan Paraphistomum cervi -lajiin. P. leydeni ja Cotylophoron skriabini ovat muita poron pötsimadon yhteydessä kirjallisuudessa esiintyviä lajinimiä. Tässä tutkimuksessa pyrittiin saamaan lisävalaistusta poron pötsimatojen taksonomiaan tunnistamalla Suomessa poroilta kerättyjä pötsimatoja lajitasolle. Tutkimuksessa käytettiin pötsimadoista tehtyjen kudosleikkeiden mikroskooppisen tutkimuksen sekä pyyhkäisyelektronimikroskoopin tarjoamia mahdollisuuksia. Lisäksi tutkimuksessa verrattiin poron pötsistä talviteurastuksen yhteydessä kerättyjen matojen rakennetta kesällä kuolleiden tai lopetettujen porojen pötsistä saatuihin matoihin. Sekä talvella että kesällä poimitut madot - huolimatta huomattavasta morfologisesta vaihtelusta ”talvi- ja kesämatojen” välillä - kuuluivat Paramphistomum leydeni -lajiin. Talvella kerätyt madot olivat selvästi pienikokoisempia, niiden ulkopinta oli vain niukasti nystyinen ja niiden sukuelimet apuelimineen olivat kehittymättömät verrattaessa niitä kesällä poimittujen matojen vastaaviin rakenteisiin. Tutkimuksen tulokset viittaavat siihen, että talvella poron pötsissä esiintyvät pienikokoiset pötsimadot ovat keskenkasvuisia P. leydeni -pötsimatoja ja että Suomessa pötsimadon kehittyminen on talvisaikaan pysähdyksissä tai hyvin hidasta. Uppgifter om renens vomflundror är sparsamma och de presenteras i allmänhet som Paramphistomum cervi. P. leydeni och Cotylophoron skriabini är andra i litteraturen nämnda artnamn för renens vomflundror. I denna undersökning strävade man till att få information om renens vomflundrors taxonomi genom att identifiera vomflundror som insamlats från renar i Finland. I undersökningen studerades vävnadssnitt av vomflundror med ljusmikroskop och scanningelektronmikroskopets möjligheter utnyttjades också. Därtill gjordes en morfologisk jämförelse av vomflundror som insamlats under renslakten på vintern med vomflundror som härstammade från renar som dött eller avlivats under sommaren. Trots betydande morfologiska variationer mellan de vomflundror som insamlades på vintern och de som erhölls på sommaren var det fråga om en och samma art, Paramphistomum leydeni. ”Vinter” vomflundrorna var betydligt mindre, ytan var obetydligt knottrig och könsorganen med tillhörande strukturer var outvecklade i jämförelse med motsvarande strukturer hos ”sommar” vomflundrorna. Undersökningen ger en antydan om att de på vintern i renens vom förekommande små vomflundrorna är unga P. leydeni vomflundror och att de utvecklas mycket långsamt eller rentav tillfälligt avstannar i sin utveckling på vintern i Finland.
