The relationship of bovine milk somatic cell count to neutrophil level in samples of cow's milk assessed by an automatic cell counter

This research communication describes the application of a fluorescent automatic cell counter Lactoscan SCC for simultaneous determination of somatic cell count and neutrophils in bovine milk. The obtained results were compared with results obtained by a flow cytometer and a light microscope. The Pearson correlations between the methods were calculated. A comparison between the main characteristics of the three kinds of analysis was made – the assay duration and the intra-assay precision. A relation between the SCC and neutrophil cells was observed in 55 milk samples. The obtained results confirm that the simultaneous determination of SCC and neutrophil analysis are necessary and support the early diagnosis of mastitis, the timely treatment of the animal and the avoidance of major economic losses.

Study of the red cell indices, hemogram and platelet variations in anaemic (<10gm%) patients by automatic cell counter in a tertiary care centre, Ahmednagar, Maharashtra, India

Background: Anemia is not a disease but it is the expression of underlying diseases and from the treatment point of view, it is necessary to identify the cause of anemia. The definition of anemia is as decrease in the number of red blood cells or the decreased percentage of hemoglobin in the blood. Anemia is such an important issue affecting a large population in India as well as worldwide, Hence, the present study was conducted to evaluate the red cell indices, haemogram and study the relation of platelet count with anaemia in anaemic (<10gm%) patients by automatic cell counter in Vikhe Patil Medical hospital.Methods: It was a prospective cross-sectional study. The sample size of research project was 100 patients, included according to the inclusion and exclusion criteria.Results: In present study, equal numbers of patients suffer from macrocytic normochromic anemia and hyperchromic anemia, that is, 8 patients of each. Out of 100 patients, 61% patients showed decreased platelet count. Maximum percentage of anaemic patients showed decreased platelet count in the range of 2.5-1.5 lakhs. Least percentage of patients (16%) showed platelet count less than 0.5 lakhs/cmm3.Conclusions: Screening for anaemia, treatment of anaemic women, and availability and use of food fortification (wheat flour with iron and folic acid), milk, sugar and salt with iron to build long term iron stores remains the key to reduce anemia in adolescent and pregnant women. Consumption of cheap iron-rich foodstuffs should be promoted. Effective poverty alleviation and hookworm prevention programs are also important.

The effect of a leukodepletion model on the activation stage of platelets

The preparation of thrombocyte concentrates with filtration before storage (in-line) makes it possible to avoid the presence of mononuclear cells in the concentrate and proinflammatory cytokines. Therefore, this filtration may result with decreased activation of trombocyte receptors in vitro, which may improve therapeutic efficiancy. Methods. We compared two groups, each with 30 therapeutic doses of concentrated thrombocytes. We prepared the first group using the classic model from the buffy coat and the other with concentrated thrombocyte samples filtrated during sampling, so-called in-line, with the WBC filter Imuflex (Terumo). Mononuclear cells (MNC), thrombocyte, and erythrocyte counts in the units of concentrated thrombocytes were obtained on an automatic cell counter, and we used flow cytometry to measure the expression of surface thrombocyte receptors. The results demonstrated that the trombocytes prepared with pre-storage filtration contained a very low level of mononuclear cells and markedly reduced trombocyte receptors. Conclusion. The number of MNC and expression of surface thrombocyte receptors were markedly lower in the concentrated thrombocyte units prepared with in-line filtration. The thrombocytes prepared in this way contain fewer mononuclear cells, are of higher quality, are more functional, and may produce a better therapeutic effect in vivo.
