The influence of age and development temperature on the temperature-related foraging risk of Formica cinerea ants

Climate change and the subsequent increase of global temperature are the most current and important threats to biodiversity. Despite the importance of temperature, our knowledge about the level of behavioural and physiological adaptations of ant species from temperate regions to cope with high temperatures is limited compared to the broad knowledge of typical thermal specialists from warmer regions. In the current study, we investigated the temperature-related foraging risk of xerothermic ant species from the temperate climate in Europe, Formica cinerea. Our aims were to check how an increase in external soil temperature affects the foraging activity of workers and how the temperature during development and worker age affects foraging activity in high temperatures. Based on our results, we can draw the following conclusions: (1) the majority of workers utilize a risk-aversive strategy in relation to foraging in high surface temperatures; (2) pupal development temperature affects the risk taken by adult workers: workers that developed in a higher temperature forage more often but for shorter intervals compared to workers that developed in a lower temperature; (3) age is an important factor in temperature-related foraging activity, as with increasing age, workers forage significantly longer at the highest temperatures. Our study is one of the first to assess the potential factors that can affect the foraging risk of ants from a temperate climate in high ambient temperatures. Significance statement Our study is the first direct test of workers' age and the development temperature of pupae on the thermal-related foraging strategy of adult F. cinerea workers. It shows that worker age and the development temperature of pupae interact to promote tolerance of thermal stress. We found that with increasing age, workers are prone to forage significantly longer at the highest and riskiest temperatures. Workers that developed in the high temperature (28°C) foraged more often but for shorter intervals compared to workers that developed in the lower temperature (20°C). Interestingly, the factor of age is more significant for ants that developed in the higher temperature of 28°C; the foraging time of these ants significantly increased with their age.

Comparative analysis of experimental testing procedures for the elicitation of rescue actions in ants

Rescue behaviour is observed when one individual provides help to another individual in danger. Most reports of rescue behaviour concern ants (Formicidae), in which workers rescue each other from various types of entrapment. Many of these entrapment situations can be simulated in the laboratory using an entrapment bioassay, in which ants confront a single endangered nest mate entrapped on a sandy arena by means of an artificial snare. Here, we compared numerous characteristics of rescue actions (contact between individuals, digging around the entrapped individual, pulling at its body parts, transport of the sand covering it and biting the snare entrapping it) in Formica cinerea ants. We performed entrapment tests in the field and in the laboratory, with the latter under varying conditions in terms of the number of ants potentially engaged in rescue actions and the arena substrate (marked or unmarked by ants’ pheromones). Rescue actions were more probable and pronounced in the field than in the laboratory, regardless of the type of test. Moreover, different test types in the laboratory yielded inconsistent results and showed noteworthy variability depending on the tested characteristic of rescue. Our results illustrate the specifics of ant rescue actions elicited in the natural setting, which is especially important considering the scarcity of field data. Furthermore, our results underline the challenges related to the comparison of results from different types of entrapment tests reported in the available literature. Additionally, our study shows how animal behaviour differs in differing experimental setups used to answer the same questions.

Heat shock proteins expression during thermal risk exposure in the temperate xerothermic ant Formica cinerea

The abiotic conditions of the desert habitat fluctuate in a circadian rhythm of hot days and cold nights. Species living in desert habitats evolved many adaptations to increase their chances of survival. However, abiotic conditions in xerothermic habitats of a temperate climate are much different. Diurnal fluctuations are not as strong, but animals have to cope with seasonal changes and hibernate during the winter, which may potentially influence their adaptations to critical temperature conditions. We attempted to assess heat resistance adaptations using the example of a widely distributed xerothermic ant Formica cinerea. Using Real-Time PCR, we measured the expression of three heat shock protein genes (Hsp60, Hsp75, Hsp90) and assessed the adaptations of F. cinerea to enable foraging in risk prone conditions. The analysis of gene expression using the Generalized Linear Model surprisingly indicated that there was no significant effect of temperature when comparing workers from the control (23ºC) with workers foraging on the surface of hot sand (47-54ºC). As a next step we tried to estimate the threshold of a thermal resistance with the use of thermal chambers. Expression of all Hsps genes increase compare to the control group, expression of Hsp60 and Hsp90 continued up to 45ºC.
