Effects of the DRD4 −521 C/T SNP on Local Neural Activity and Functional Connectivity in Children With ADHD

Objective: The present study aimed to investigate the effects of the dopamine receptor D4 (DRD4) −521 C/T single-nucleotide polymorphism on brain function among children with attention deficit hyperactivity disorder (ADHD) and to evaluate whether brain function is associated with behavioral performance among this demographic.Methods: Using regional homogeneity, fractional amplitude low-frequency fluctuation, and functional connectivity as measurement indices, we compared differences in resting-state brain function between 34 boys with ADHD in the TT homozygous group and 37 boys with ADHD in the C-allele carrier group. The Conners' Parent Rating Scale, the SNAP-IV Rating Scale, the Stroop Color Word Test, the go/no-go task, the n-back task, and the working memory index within the Wechsler Intelligence Scale for Children-Fourth Edition were selected as comparative indicators in order to test effects on behavioral performance.Results: We found that TT homozygotes had low behavioral performance as compared with C-allele carriers. The regional homogeneity for TT homozygotes decreased in the right middle occipital gyrus and increased in the right superior frontal gyrus as compared with C-allele carriers. In addition, the right middle occipital gyrus and the right superior frontal gyrus were used as the seeds of functional connectivity, and we found that the functional connectivity between the right middle occipital gyrus and the right cerebellum decreased, as did the functional connectivity between the right superior frontal gyrus and the angular gyrus. No statistically significant differences were observed in the respective brain regions when comparing the fractional amplitudes for low-frequency fluctuation between the two groups. Correlation analyses demonstrated that the fractional amplitude low-frequency fluctuation in the precentral gyrus for TT homozygotes were statistically significantly correlated with working memory.Conclusions: We found differing effects of DRD4 −521 C/T polymorphisms on brain function among boys with ADHD. These findings promote our understanding of the genetic basis for neurobiological differences observed among children with ADHD, but they must be confirmed in larger samples.

How Does Approaching a Lead Vehicle and Monitoring Request Affect Drivers’ Takeover Performance? A Simulated Driving Study with Functional MRI

With the popularization and application of conditionally automated driving systems, takeover requirements are becoming more and more frequent, and the subsequent takeover safety problems have attracted attention. The present study used functional magnetic resonance imaging (fMRI) technology, combined with driving simulation experiments, to study in depth the effects of critical degree and monitor request (MR) 30 s in advance on drivers’ visual behavior, takeover performance and brain activation. Results showed that MR can effectively improve the driver’s visual and takeover performance, including visual reaction times, fixation frequency and duration, takeover time, and takeover mode. The length of the reserved safety distance can significantly affect the distribution of longitudinal acceleration. Critical or non-critical takeover has a significant impact on the change of pupil diameter and the standard deviation of lateral displacement. Five brain regions, including the middle occipital gyrus (MOG), fusiform gyrus (FG), middle temporal gyrus (MTG), precuneus and precentral, are activated under the stimulation of a critical takeover scenario, and are related to cognitive behaviors such as visual cognition, distance perception, memory search and movement association.

Atypical Antipsychotics Mediate Dynamics of Intrinsic Brain Activity in Early-Stage Schizophrenia? A Preliminary Study

Objective Abnormalities of static brain activity have been reported in schizophrenia, but it remains to be clarified the temporal variability of intrinsic brain activities in schizophrenia and how atypical antipsychotics affect it.Methods We employed a resting-state functional magnetic resonance imaging (rs-fMRI) and a sliding-window analysis of dynamic amplitude of low-frequency fluctuation (dALFF) to evaluate the dynamic brain activities in schizophrenia (SZ) patients before and after 8-week antipsychotic treatment. Twenty-six schizophrenia individuals and 26 matched healthy controls (HC) were included in this study.Results Compared with HC, SZ showed stronger dALFF in the right inferior temporal gyrus (ITG.R) at baseline. After medication, the SZ group exhibited reduced dALFF in the right middle occipital gyrus (MOG.R) and increased dALFF in the left superior frontal gyrus (SFG.L), right middle frontal gyrus (MFG.R), and right inferior parietal lobule (IPL.R). Dynamic ALFF in IPL.R was found to significant negative correlate with the Scale for the Assessment of Negative Symptoms (SANS) scores at baseline.Conclusion Our results showed dynamic intrinsic brain activities altered in schizophrenia after short term antipsychotic treatment. The findings of this study support and expand the application of dALFF method in the study of the pathological mechanism in psychosis in the future.

Aberrant Brain Spontaneous Activity and Synchronization in Type 2 Diabetes Mellitus Subjects Without Mild Cognitive Impairment

Objective: We aimed to explore whether the percent amplitude of fluctuation (PerAF) measurement could provide supplementary information for amplitude of low-frequency fluctuation (ALFF) about spontaneous activity alteration in type 2 diabetes mellitus (T2DM) subjects without mild cognitive impairment (MCI). Then we further evaluated the synchronization through the method of functional connectivity (FC) to better demonstrate brain changes in a more comprehensive manner in T2DM.Methods: Thirty T2DM subjects without MCI and thirty well-matched healthy subjects were recruited in this study. Subjects’ clinical data, neuropsychological test results, and resting-state functional magnetic resonance imaging (rs-fMRI) data were acquired. Voxel-based group difference comparisons between PerAF and ALFF were conducted. Then, seed-based FC between the recognized brain regions based on PerAF and ALFF and the rest of the whole brain was performed.Results: Compared with healthy group, T2DM group had significantly decreased PerAF in the bilateral middle occipital gyrus and the right calcarine, increased ALFF in the right orbital inferior frontal gyrus and decreased ALFF in the right calcarine. Seed-based FC analysis showed that the right middle occipital gyrus of T2DM subjects exhibited significantly decreased FC with the right caudate nucleus and right putamen. According to the partial correlation analyses, hemoglobin A1c (HbA1c) and immediate memory scores on the auditory verbal learning test (AVLT) were negatively correlated in the T2DM group. However, we found that total cholesterol was positively correlated with symbol digit test (SDT) scores.Conclusion: PerAF and ALFF may have different sensitivities in detecting the abnormal spontaneous brain activity in T2DM subjects. We suggest PerAF values may add supplementary information and indicate additional potential neuronal spontaneous activity in T2DM subjects without MCI, which may provide new insights into the neuroimaging mechanisms underlying early diabetes-associated cognitive decline.

Brain Responses to High-Calorie Visual Food Cues in Individuals with Normal-Weight or Obesity: An Activation Likelihood Estimation Meta-Analysis

Overconsumption of high-calorie or unhealthy foods commonly leads to weight gain. Understanding people’s neural responses to high-calorie food cues might help to develop better interventions for preventing or reducing overeating and weight gain. In this review, we conducted a coordinate-based meta-analysis of functional magnetic resonance imaging studies of viewing high-calorie food cues in both normal-weight people and people with obesity. Electronic databases were searched for relevant articles, retrieving 59 eligible studies containing 2410 unique participants. The results of an activation likelihood estimation indicate large clusters in a range of structures, including the orbitofrontal cortex (OFC), amygdala, insula/frontal operculum, culmen, as well as the middle occipital gyrus, lingual gyrus, and fusiform gyrus. Conjunction analysis suggested that both normal-weight people and people with obesity activated OFC, supporting that the two groups share common neural substrates of reward processing when viewing high-calorie food cues. The contrast analyses did not show significant activations when comparing obesity with normal-weight. Together, these results provide new important evidence for the neural mechanism underlying high-calorie food cues processing, and new insights into common and distinct brain activations of viewing high-calorie food cues between people with obesity and normal-weight people.

Pictograph Portrays What It Is

As one origin of Chinese characters, pictograph has a graphical structure based on its referent. Are the aesthetic qualities of the reference objects reflected in the neural processing of pictographs? In the study reviewed in this chapter, participants were scanned while making aesthetic judgments of pictographs and their referents. The conjunction analysis revealed the common involvement of the bilateral inferior occipital gyri and inferior frontal gyri, the right superior occipital gyrus, the left middle occipital gyrus, and the inferior orbitofrontal cortex for the aesthetic judgments of pictographs and object images referring to beautiful objects. Moreover, only the beautiful judgments for pictographs but not object images activated the motor areas, implying that an approach motivation was elicited during the aesthetic perception of novel pictographs. Results indicate that not only the object images, but also the corresponding pictographs arouse a sense of beauty that relies on common neural mechanisms during aesthetic judgments.

Brain structural and functional anomalies associated with simultanagnosia in patients with posterior cortical atrophy

Simultanagnosia is a common symptom of posterior cortical atrophy, and its association with brain structural and functional changes remains unclear. In our study, 18 posterior cortical atrophy patients with simultanagnosia, 29 patients with Alzheimer’s disease and 20 cognitively normal controls were recruited and subjected to full neuropsychological evaluation, including simultanagnosia tests, and structural and resting-state functional MRI. The gray matter volume was assessed by voxel-based morphometry, while the intrinsic functional connectivity was evaluated using the reduced gray matter volume regions of interest as the seed. In contrast to the patients with Alzheimer’s disease, those with posterior cortical atrophy showed the following: (1) markedly lower simultanagnosia test scores, (2) an altered regional gray matter volume of the left middle occipital gyrus and ventral occipital areas, and (3) lowered intrinsic functional connectivity with the left middle occipital gyrus, left lingual gyrus and right middle occipital gyrus separately. Additionally, the gray matter volume of the left middle occipital gyrus and left inferior occipital gyrus were each correlated with simultanagnosia in posterior cortical atrophy patients. The intrinsic functional connectivity of the left middle occipital gyrus with the right superior occipital gyrus and that of the right middle occipital gyrus with the left superior parietal gyrus were also correlated with simultanagnosia in posterior cortical atrophy patients. In summary, this study indicated that simultanagnosia is associated with gray matter reductions and decreased functional connectivity in the left middle occipital gyrus and the left inferior occipital gyrus in patients with posterior cortical atrophy.

NEURAL CORRELATES OF SIMPLE SENSOMOTOR REACTION AS MARKERS OF INFORMATION PROCESSING SPEED IN MILITARY VETERANS

We examined the latent periods (LP) of simple sensorimo- tor reaction (SSMR) and peculiarities of brain connectiv- ity during carrying out tests on SSMR in the group of veterans of Joint Forces Operation (JFO) and in military men-patients with post-traumatic stress disorder (PTSD) and with mild traumatic brain injury (mTBI). To this aim, we used a computer test with coherent analysis of EEG. Differences in brain activation were analyzed by LORETA method. It was shown that the speed of SSMR in the group of JFO veterans was significantly lower than in the con- trol group. The LP of SSMR in the group of JFO veterans were 279 ms vs 258 ms in the control group. Comparative analysis of SSMR LP in the group of JFO veterans and in the group with PTSD as well as group with mTBI did not reveal significant differences. The EEG study and coherent analysis showed a significant decrease in the brain connectivity in the group of JFO veterans compared to control group, especially in the low frequency ranges. There were no significant differences in brain activation between the JFO veterans, PTSD and mTBI patients, but JFO veterans had significantly lower activation of cu- neus, middle occipital gyrus and lingual gyrus compared to control group. Conclusively, in order to detect hidden brain lesions, an additional medical examination may be recommended for JFO veterans.

Alterations in the Occipital Cortex of Drug-Naïve Adults With Major Depressive Disorder: A Surface-Based Analysis of Surface Area and Cortical Thickness

Objective Advances in surface-based morphometric methods have allowed researchers to separate cortical volume into cortical thickness (CTh) and surface area (SA). Although CTh alterations in major depressive disorder (MDD) have been observed in numerous studies, few studies have described significant SA alterations. Our study aimed to measure patients’ SAs and to compare it with their CTh to examine whether SA exhibits alteration patterns that differ from those of CTh in drug-naïve patients with MDD.Methods A total of 71 drug-naïve MDD patients and 111 healthy controls underwent structural magnetic resonance imaging, and SA and CTh were analyzed between the groups.Results We found a smaller SA in the left superior occipital gyrus (L-SOG) in drug-naïve patients with MDD. In the CTh analysis, the bilateral fusiform gyrus, left middle occipital gyrus, left temporal superior gyrus, and right posterior cingulate showed thinner cortices in patients with MDD, while the CTh of the bilateral SOG, right straight gyrus, right posterior cingulate, and left lingual gyrus were increased.Conclusion Compared with the bilateral occipito-temporal changes in CTh, SA alterations in patients with MDD were confined to the L-SOG. These findings may improve our understanding of the neurobiological mechanisms of SA alteration in relation to MDD.

The Value of Brain Resting-State Functional Magnetic Resonance Imaging on Image Registration Algorithm in Analyzing Abnormal Changes of Neuronal Activity in Patients with Type 2 Diabetes

The aim of this paper was to analyze the application value of resting-state functional magnetic resonance imaging (FMRI) parameters and rigid transformation algorithm in patients with type 2 diabetes (T2DM), which could provide a theoretical basis for the registration application of FMRI. 107 patients confirmed pathologically as T2DM and 51 community medical healthy volunteers were selected and divided into an experimental group and a control group, respectively. Besides, all the subjects were scanned with FMRI. Then, the rigid transformation-principal axis algorithm (RT-PAA), Levenberg–Marquardt iterative closest point (LMICP), and Demons algorithm were applied to magnetic resonance image registration. It was found that RT-PAA was superior to LMICP and Demons in image registration. The amplitude of low-frequency fluctuation (ALFF) values of the left middle temporal gyrus, right middle temporal gyrus, left fusiform gyrus, right inferior occipital gyrus, and left middle occipital gyrus in patients from the experimental group were lower than those of the control group P < 0.05 . The Montreal cognitive assessment (MoCA) score was extremely negatively correlated with the ALFF of the left middle temporal gyrus (r = −0.451 and P < 0.001 ) and highly positively associated with the ALFF of the right posterior cerebellar lobe (r = −0.484 and P < 0.001 ). In addition, the MoCA score of patients had a dramatically negative correlation with the ALFF of the left middle temporal gyrus (r = −0.602 and P < 0.001 ) and had a greatly positive correlation with the ALFF of the right posterior cerebellar lobe (r = −0.516 and P < 0.001 ). The results showed that RT-PAA based on rigid transformation in this study had a good registration effect on magnetic resonance images. Compared with healthy volunteers, the left middle temporal gyrus, right middle temporal gyrus, left fusiform gyrus, right inferior occipital gyrus, and left middle occipital gyrus in patients with T2DM showed abnormal neuronal changes and reduced cognitive function.
