Enabling coherent BaZrO3 nanorods/YBa2Cu3O7-x interface through dynamic lattice enlargement in vertical epitaxy of BaZrO3/YBa2Cu3O7-x nanocomposites

One-dimensional c-axis-aligned BaZrO3 (BZO) nanorods are regarded as strong one-dimensional artificial pinning centers (1D-APCs) in BZO-doped YaBa2Cu3O7-x (BZO/YBCO) nanocomposite films. However, a microstructure analysis has revealed a defective, oxygen-deficient YBCO column around the BZO 1D-APCs due to the large lattice mismatch of ~7.7% between the BZO (3a=1.26 nm) and YBCO (c=1.17 nm), which has been blamed for the reduced pinning efficiency of BZO 1D-APCs. Herein, we report a dynamic lattice enlargement approach on the tensile strained YBCO lattice during the BZO 1D-APCs growth to induce c-axis elongation of the YBCO lattice up to 1.26 nm near the BZO 1D-APC/YBCO interface via Ca/Cu substitution on single Cu-O planes of YBCO, which prevents the interfacial defect formation by reducing the BZO/YBCO lattice mismatch to ~1.4%. Specifically, this is achieved by inserting thin Ca0.3Y0.7Ba2Cu3O7-x (CaY-123) spacers as the Ca reservoir in 2-6 vol.% BZO/YBCO nanocomposite multilayer (ML) films. A defect-free, coherent BZO 1D-APC/YBCO interface is confirmed in transmission electron microscopy and elemental distribution analyses. Excitingly, up to five-fold enhancement of Jc (B) at magnetic field B=9.0 T//c-axis and 65-77 K was obtained in the ML samples as compared to their BZO/YBCO single-layer (SL) counterpart’s. This has led to a record high pinning force density Fp together with significantly enhanced Bmax at which Fp reaches its maximum value Fp,max for BZO 1D-APCs at B//c-axis. At 65 K, the Fp,max ~158 GN/m3 and Bmax ~ 8.0 T for the 6% BZO/YBCO ML samples represent a significant enhancement over Fp,max ~36.1 GN/m3 and Bmax ~ 5.0 T for the 6% BZO/YBCO SL counterparts. This result not only illustrates the critical importance of a coherent BZO 1D-APC/YBCO interface in the pinning efficiency, but also provides a facile scheme to achieve such an interface to restore the pristine pinning efficiency of the BZO 1D-APCs.

Geometric control of Myosin-II orientation during axis elongation

The actomyosin cytoskeleton is a crucial driver of morphogenesis. Yet how the behavior of large scale cytoskeletal patterns in deforming tissues emerges from the interplay of geometry, genetics, and mechanics remains incompletely understood. Convergent extension flow in D. melanogaster embryos provides the opportunity to establish a quantitative understanding of the dynamics of anisotropic non-muscle myosin II. Cell-scale analysis of protein localization in fixed embryos suggests that there are complex rules governing how the control of myosin anisotropy is regulated by gene expression patterns. However, technical limitations have impeded quantitative and dynamic studies of this process at the whole embryo level, leaving the role of geometry open. Here we combine in toto live imaging with quantitative analysis of molecular dynamics to characterize the distribution of myosin anisotropy and corresponding genetic patterning. We found pair rule gene expression continuously deformed, flowing with the tissue frame. In contrast, myosin anisotropy orientation remained nearly static, aligned with the stationary dorsal-ventral axis of the embryo. We propose myosin recruitment by a geometrically defined static source, potentially related to the embryo-scale epithelial tension, and account for transient deflections by the interplay of cytoskeletal turnover with junction reorientation by flow. With only one parameter, this model quantitatively accounts for the time course of myosin anisotropy orientation in wild-type, twist, and even-skipped embryos as well as embryos with perturbed egg geometry. Geometric patterning of the cytoskeleton suggests a simple physical strategy to ensure a robust flow and formation of shape.

Optogenetic dissection of the roles of actomyosin in the mechanics underlying tissue fluidity

Rapid epithelial tissue flows are essential to building and shaping developing embryos. However, it is not well understood how the mechanical properties of tissues and the forces driving them to flow are jointly regulated to accommodate rapid tissue remodeling. To dissect the roles of actomyosin in the mechanics of epithelial tissue flows, here we use two optogenetic tools, optoGEF and optoGAP, to manipulate Rho/Rho-kinase signaling and actomyosin contractility in the germband epithelium, which flows via convergent extension during Drosophila body axis elongation. The ability to perturb actomyosin across the tissue allows us to analyze the effects of actomyosin on cell rearrangements, tissue tensions, and tissue mechanical properties. We find that either optogenetic activation or deactivation of Rho1 signaling and actomyosin contractility at the apical surface of the germband disrupts cell rearrangements and tissue-level flows. By probing mechanical tensions in the tissue using laser ablation and predicting tissue mechanical properties from cell packings, we find that actomyosin influences both the anisotropic forces driving tissue flow and the mechanical properties of the tissue resisting flow, leading to complex relationships between actomyosin activity and tissue-level flow. Moreover, our results indicate that changes in the distribution of medial and junctional myosin in the different perturbations alter tissue tension and cell packings in distinct ways, revealing how junctional and medial myosin have differential roles in promoting and orienting cell rearrangements to tune tissue flows in developing embryos.

RASSF8-mediated transport of Echinoid via the exocyst promotes Drosophila wing elongation and epithelial ordering

Cell-cell junctions are dynamic structures that maintain cell cohesion and shape in epithelial tissues. During development, junctions undergo extensive rearrangements to drive the epithelial remodelling required for morphogenesis. This is particularly evident during axis elongation, where neighbour exchanges, cell-cell rearrangements and oriented cell divisions lead to large-scale alterations in tissue shape. Polarised vesicle trafficking of junctional components by the exocyst complex has been proposed to promote junctional rearrangements during epithelial remodelling, but the receptors that allow exocyst docking to the target membranes remain poorly understood. Here, we show that the adherens junction component Ras Association domain family 8 (RASSF8) is required for the epithelial re-ordering that occurs during Drosophila pupal wing proximo-distal elongation. We identify the exocyst component Sec15 as a RASSF8 interactor. RASSF8 loss elicits cytoplasmic accumulation of Sec15 and Rab11-containing vesicles. These vesicles also contain the nectin-like homophilic adhesion molecule Echinoid, whose depletion phenocopies the wing elongation and epithelial packing defects observed in RASSF8 mutants. Thus, our results suggest that RASSF8 promotes exocyst-dependent docking of Echinoid-containing vesicles during morphogenesis.

Time Course of Root Axis Elongation and Lateral Root Formation in Perennial Ryegrass (Lolium perenne L.)

Grasses have a segmental morphology. Compared to leaf development, data on root development at the phytomer level are scarce. Leaf appearance interval was recorded over time to allow inference about the age of segmental sites that later form roots. Hydroponically grown Lolium perenne cv. Aberdart tillers were studied in both spring and autumn in increasing and decreasing day length conditions, respectively, and dissected to define the development status of roots of known age on successive phytomers basipetally on the tiller axis. Over a 90-day observation period spring and autumn tillers produced 10.4 and 18.1 root bearing phytomers (Pr), respectively. Four stages of root development were identified: (0) main axis elongation (~0–10 days), (1) primary branching (~10–18 days), (2) secondary branching (~18–25 days), and (3) tertiary and quaternary branching without further increase in root dry weight. The individual spring roots achieved significantly greater dry weight (35%) than autumn roots, and a mechanism for seasonal shift in substrate supply to roots is proposed. Our data define a root turnover pattern likely also occurring in field swards and provide insight for modelling the turnover of grass root systems for developing nutrient efficient or stress tolerant ryegrass swards.

Anterior expansion and posterior addition to the notochord mechanically coordinate embryo axis elongation

How force generated by the morphogenesis of one tissue impacts the morphogenesis of other tissues to achieve an elongated embryo axis is not well understood. The notochord runs along the length of the somitic compartment and is flanked on either side by somites. Vacuolating notochord cells undergo a constrained expansion, increasing notochord internal pressure and driving its elongation and stiffening. Therefore, the notochord is appropriately positioned to play a role in mechanically elongating the somitic compartment. We use multi-photon cell ablation to remove specific regions of the notochord and quantify the impact on axis elongation. We show that anterior expansion generates a force that displaces notochord cells posteriorly relative to adjacent axial tissues, contributing to the elongation of segmented tissue during post-tailbud stages. Unexpanded cells derived from progenitors at the posterior end of the notochord provide resistance to anterior notochord cell expansion, allowing for stress generation along the AP axis. Therefore, notochord cell expansion beginning in the anterior, and addition of cells to the posterior notochord, act as temporally coordinated morphogenetic events that shape the zebrafish embryo AP axis.
