This research was a Classroom Action Research (CAR) which aimed to improve the students learning motivation in IAIN Bone through the application of the Two Stay Two Stray (TS-TS) learning model with Activity Window Shopping. This research was conducted in two cycles. Each cycle consists of four stages, namely planning, action, observation, and reflection. The techniques in Data collection used in this research were questionnaire and documentation. The instrument used in this research was a list of questions on the questionnaire. The technique in data analysis used was descriptive data analysis by looking at the average score percentage by describing the quantitative data obtained. Based on the results of the research, it was concluded that the Application of Two Stay Two Stray (TS - TS) Learning Model with Window Shopping Activity can increase the students learning motivation in IAIN Bone. From the result of questionnaire data analysis the research showed that the percentage of the students learning motivation was obtained by an average score of 77.10% in the first cycle and then increases to 82.07% in the second cycle or increase of 4.97%.   Abstrak Penelitian ini merupakan Penelitian Tindakan Kelas (PTK) yang bertujuan untuk meningkatkan motivasi belajar mahasiswa IAIN Bone melalui penerapan model pembelajaran Two Stay Two Stray (TS - TS) dengan Aktivitas Window Shopping. Penelitian ini dilaksanakan dalam dua siklus. Tiap siklus terdiri dari empat tahapan, yaitu perencanaan, tindakan, pengamatan, dan refleksi. Teknik pengumpulan data yang digunakan dalam penelitian ini adalah angket dan dokumentasi. Instrumen yang digunakan dalam penelitian  ini adalah daftar pertanyaan pada angket. Teknik analisis data yang digunakan adalah analisis data deskriptif dengan melihat persentase skor rata- rata dengan mendeskripsikan data kuantitatif yang diperoleh. Berdasarkan hasil penelitian disimpulkan bahwa Penerapan Model Pembelajaran Two Stay Two Stray (TS - TS) dengan Aktivitas Window Shopping dapat  Meningkatkan  Motivasi Belajar Mahasiswa IAIN Bone. Dari hasil analisis data angket penelitian menunjukkan bahwa persentase motivasi belajar mahasiswa diperoleh rata –rata  skor 77,10 % pada siklus I dan kemudian meningkat menjadi 82,07 % pada siklus II atau terjadi peningkatan 4,97%. AbstractCytidine base editors, composed of a cytidine deaminase fused to Cas9 nickase, enable efficient C-to-T conversion in various organisms. However, current base editors can induce unwanted bystander C-to-T conversions when more than one C is present in the activity window of cytidine deaminase, which negatively affects the precision. Here, we develop a new base editor with CC context-specificity using rationally engineered human APOBEC3G, thus significantly reduce unwanted bystander activities. In addition, efficient C-to-T conversion that can further recognize relaxed NG PAMs is achieved by combining an engineered SpCas9-NG variant. These novel base editors with improved precision and targeting scope will expand the toolset for precise gene modification in organisms.

Abstract The development of online social environments has changed the manner of social interaction and communication, which are driven by individual human actions. Thus temporal variations in interaction networks are deeply impacted by the temporal dimension of human activity. In this paper, we address this issue through a detailed analysis on the retweets and comments of 550,000 Twitter users. We propose a temporal network model to represent the interaction network on Twitter, in which each node contains an activity window and the emergence of the edges between nodes are dependent on it. Specifically, the activity window is defined as the backtracking length from the message flow posted by the user’s friend, which represents the user’s social ability. It complies with a power-law distribution with an exponential cut-off. The interaction network is sparser and more clustered than the followee-follower network, in which the interaction stability and burstiness fluctuate with the activity window or with the degree to which the two users are involved in the communication. Finally, the effect of activity window on the aggregating degrees of the interaction network is examined.

ABSTRACTCRISPR-mediated base editors have opened unique avenues for scar-free genome-wide mutagenesis. Here, we describe a comprehensive computational workflow called beditor that can be broadly adapted for designing guide RNA libraries with a range of CRISPR-mediated base editors, PAM recognition sequences and genomes of many species. Additionally, in order to assist users in selecting the best sets of guide RNAs for their experiments, a priori estimates, called beditor scores are calculated. These beditor scores are intended to select guide RNAs that conform to requirements for optimal base editing: the editable base falls within maximum activity window of the CRISPR-mediated base editor and produces non-confounding mutational effects with minimal predicted off-target effects. We demonstrate the utility of the software by designing guide RNAs for base-editing to create or remove thousands of clinically important human disease mutations.

The crack formed under the action of circular medium fluid around the wellbore of brittle shale has been a hot issue in recent years. This paper considers the brittle shale micro fracture formation mechanism from the perspective of inhibitive ability of drilling fluid activity. The model of activity-swelling-hydration is set up firstly through revising generalized Usher model, it states the drilling fluid activity range to keep wellbore in osmotic hydration stage with the critical expansion ratio, thus puts forward the concept of activity window and is verified by indoor test. Results show that the different activity of drilling fluid brings difference in rock strength in macro and time sensitive effect for internal structure of rock in micro. This mechanism can provide a more accurate range design for drilling fluid activity and strong guarantee for wellbore stability.
