The solubility product extends the buffering concept to heterotypic biomolecular condensates.

Biomolecular condensates are formed by liquid-liquid phase separation (LLPS) of multivalent molecules. LLPS from a single ('homotypic') constituent is governed by buffering: above a threshold, free monomer concentration is clamped, with all added molecules entering the condensed phase. However, both experiment and theory demonstrate that buffering fails for the concentration dependence of multi-component ('heterotypic') LLPS. Using network-free stochastic modeling, we demonstrate that LLPS can be described by the solubility product constant (Ksp): the product of free monomer concentrations, accounting for the ideal stoichiometries governed by the valencies, displays a threshold above which additional monomers are funneled into large clusters; this reduces to simple buffering for homotypic systems. The Ksp regulates the composition of the dilute phase for a wide range of valencies and stoichiometries. The role of Ksp is further supported by coarse-grained spatial particle simulations. Thus, the solubility product offers a general formulation for the concentration dependence of LLPS.

JUSTIFICATION OF THE APPLICATION OF CONSTRUCTION MATERIALS IN THE PHENOMENA OF INTOLERANCE TO ACRYLATES

Introduction. Today, for the manufacture of prostheses with a plastic base, hot polymerization acrylic plastic is most often used, which contains a free monomer — methyl methacrylate. However, in some patients, after their placement in the oral cavity, as a result of orthopedic treatment, various clinical pathological manifestations may occur, which in general are defined as individual intolerance to dental materials. The aim is to carry out a systematic analysis of domestic and foreign literary sources to determine the most suitable base material in patients with intolerance to structural materials. Methodology. The research was carried out on the basis of the search for scientific papers, in the electronic scientific library elibrary.ru for the last 15 years, as well as in the search engines PubMed, Scopus and Wiley Inter Science for 2009–2020. The search was carried out by keywords. Results. The article analyzes the literature data indicating the importance of the choice of the base material in the pathogenesis of intolerance to dental materials. During the analysis of the literature, the positive properties of a new, alternative light-curing base material, which can be used in patients with acrylate intolerance, have been substantiated. Conclusions. The results obtained, in the course of the literature analysis, allow us to highlight the advantages of using a light-curing base material in comparison with acrylic plastics and thermoplastic resins. The results of the use of the light-curing base material in clinical practice will be communicated by us to the dental community in subsequent publications.

Surface Roughness of Nanohybrid Composites with Different Monomers after Finishing and Polishing with Different Polishing Systems

Background: The most frequently used monomer in commercial composite resins is bisphenol A glycidyl dimethacrylate. Bisphenol A glycidyl dimethacrylate free monomer and the long chain dimer acid based monomer were considered as an alternative method to improve mechanical and physical properties of composite resins. Objective: To study the surface roughness of different nanohybrid composites with different monomer compositions after finishing and polishing with different polishing systems. Materials and Methods: 72 composite disk specimens were prepared and divided into 3 groups (n=24) according to the resin composite that were used (harmonizekerr, venus diamondheraeuskulzer and n'duranceseptodont). Each group was subdivided into 4 subgroups (n=6) according to the polishing technique that was used (Jiffy natural universal wheelsultradent, sof-lex spiral wheels 3m espe, hilusterkerr or enhance system dentsply). Atomic force microscope was used to measure surface roughness. Results: There was statistical significant difference in surface roughness measuring among different n'durance, venus diamond and harmonize groups (Kruskal Wallis Test p<0.05). Post hock pairwise comparison revealed that venus diamond showed statistically significant higher surface roughness when compared with either n'durance and harmonize. Among the polishing systems, jiffy natural universal wheels produce the least surface roughness. Conclusion: Different monomer compositions may have direct effect on the final surface polish of the restorative materials. Clinical significance: The use of jiffy natural universal wheels may be found to result in the smoothest surface finish with the least clinical steps.

Complex-Radical Terpolymerization of Maleic Anhydride (Styrene), Allyl Propionate and Methyl Methacrylate

<p>The radical terpolymerization reactions of the acceptor-donor-acceptor and donor-acceptor-donor systems, maleic anhydride (MA)-allyl propionate (AP)-methyl methacrylate (MMA) and styrene (St)-MMA-AP, had been studied. The terpolymerizations were carried out in methyl ethyl ketone at 60-75 °C in the presence of 2,2’-azoisobutyronitrile (ABIN) used as the initiator. Some kinetic parameters and copolymerization constants - for both, system were determined by dilatometric and Kelen-Tudos or Seiner-Lift methods. The obtained results are discussed in terms of the free monomer and complex chain growth models. It is shown that terpolymerization was carried out at a stage close to binary copolymerization of MA...AP complex with free MMA and St...MMA complex with AP in the both studied system, respectively. These systems are also used as model for interpretation of cyclocopolymerization mechanism in allyl methactylate-MA (or St) system. DTA and TGA analyses indicated the relatively high thermal stability of St-MMA-AP terpolymer. It is shown that this terpolymer decomposes through a one-step reaction at 310 °C, however MA-AP-MMA terpolymer decomposes through a multi-step reactions at 150, 260 and 310 °C.</p>

The structure and properties of septin 3: a possible missing link in septin filament formation

The human genome codes for 13 members of a family of filament-forming GTP-binding proteins known as septins. These have been divided into four different subgroups on the basis of sequence similarity. The differences between the subgroups are believed to control their correct assembly into heterofilaments which have specific roles in membrane remodelling events. Many different combinations of the 13 proteins are theoretically possible and it is therefore important to understand the structural basis of specific filament assembly. However, three-dimensional structures are currently available for only three of the four subgroups. In the present study we describe the crystal structure of a construct of human SEPT3 which belongs to the outstanding subgroup. This construct (SEPT3-GC), which includes the GTP-binding and C-terminal domains, purifies as a nucleotide-free monomer, allowing for its characterization in terms of GTP-binding and hydrolysis. In the crystal structure, SEPT3-GC forms foreshortened filaments which employ the same NC and G interfaces observed in the heterotrimeric complex of human septins 2, 6 and 7, reinforcing the notion of ‘promiscuous’ interactions described previously. In the present study we describe these two interfaces and relate the structure to its tendency to form monomers and its efficiency in the hydrolysis of GTP. The relevance of these results is emphasized by the fact that septins from the SEPT3 subgroup may be important determinants of polymerization by occupying the terminal position in octameric units which themselves form the building blocks of at least some heterofilaments.
