Abstract Preparing actions to achieve goals, overriding habitual responses, and substituting actions that are no longer relevant are aspects of motor control often assumed to be driven by deliberate top-down processes. In the present study, we investigated whether motor control could come under involuntary control of environmental cues that have been associated with specific actions in the past. We used transcranial magnetic stimulation (TMS) to probe corticospinal excitability as an index of motor preparation, while participants performed a Go/No-Go task (i.e., an action outcome or no action outcome task) and rated what trial was expected to appear next (Go or No-Go). We found that corticospinal excitability during a warning cue for the upcoming trial closely matched recent experience (i.e., cue–outcome pairings), despite conflicting with what participants expected would appear. The results reveal that in an action–outcome task, neurophysiological indices of motor preparation show changes that are consistent with participants learning to associate a preparatory warning cue with a specific action, and are not consistent with the action that participants explicitly anticipate making. This dissociation with conscious expectancy ratings reveals that conditioned responding and motor preparation can operate independently of conscious expectancies about having to act.

We assessed the effects of anodal/cathodal direct current stimulation (DCS) applied epidurally over the cerebellum. We studied the excitability of both the motor cortex and the anterior horn of the spinal cord in adult rats under continuous anesthesia. We also investigated the effects on the spatial representation of a couple of agonist/antagonist muscles on primary motor cortex. Moreover, we evaluated the effects on the afferent inhibition in a paradigm of conditioned corticomotor responses. Anodal DCS of the cerebellum (1) decreased the excitability of the motor cortex, (2) reduced the excitability ofFwaves, as shown by the decrease of both meanF/meanMratios and persistence ofFwaves, (3) exerted a “smoothing effect” on corticomotor maps, reshaping the representation of muscles on the motor cortex, and (4) enhanced the afferent inhibition of conditioned motor evoked responses. Cathodal DCS of the cerebellum exerted partially reverse effects. DCS of the cerebellum modulates the excitability of both motor cortex and spinal cord at the level of the anterior horn. This is the first demonstration that cerebellar DCS tunes the shape of corticomotor maps. Our findings provide a novel mechanism by which DCS of the cerebellum exerts a remote neuromodulatory effect upon motor cortex.

This article reviews the physiology and application of the currently available paired-pulse protocols. Paired-pulse transcranial magnetic stimulation (TMS) techniques study the modulation of human motor cortical excitability. Paired-pulse experiments are designed to give insight into the nature of the cortical circuitry activated by TMS. Changes in motor cortical excitability produced by the conditioning pulse are estimated by changes in the size of the conditioned motor-evoked potential (MEP). It is possible to identify specific abnormalities in the balance between inhibitory and facilitatory processes, even if the pathology lies in abnormal afferent signalling to the motor cortex rather than in the motor cortex itself. The conclusion that emerges from the studies on interhemispheric interactions is that it is now possible by means of TMS protocols to chart long-range functional interhemispheric connectivity of remote areas of the human brain.
