AbstractL-lactate is an important metabolite, energy source, and signaling molecule in health and disease. In mammals, its transport across biological membranes is mediated by monocarboxylate transporters (MCTs) of the solute carrier 16 (SLC16) family. Malfunction, overexpression or absence of transporters of this family are associated with diseases such as cancer and type 2 diabetes. Moreover, lactate acts as a signaling molecule and virulence factor in certain bacterial infections. Here, we report the rational, structure-guided identification of potent, nanomolar affinity inhibitors acting on an L-lactate-specific SLC16 homologue from the bacterium Syntrophobacter fumaroxidans (SfMCT). High-resolution crystal structures of SfMCT with bound inhibitors uncovered their interaction mechanism on an atomic level and the role of water molecules in inhibitor binding. The presented systematic approach is a valuable procedure for the identification of L-lactate transport inhibitors. Furthermore, identified inhibitors represent potential tool compounds to interfere with monocarboxylate transport across biological membranes mediated by MCTs.

Sepsis is the number one cause of death in intensive care units. This life-threatening condition is caused by bacterial infections and triggered by endotoxins of Gram-negative bacteria that leads to an overreaction of the immune system. The synthetic anti-lipopolysaccharide peptide Pep19-2.5 is a promising candidate for the treatment of sepsis as it binds sepsis-inducing lipopolysaccharides and thus prevents initiation of septic shock. For clinical evaluation precise quantification of the peptide in blood and tissue is required. As the peptide is not extractable from biological samples by commonly used methods there is a need for a new analysis method that does not rely on extraction of the peptide. In order to quantify the peptide by mass spectrometry, the peptide was synthesized containing 13C9,15N1-labeled phenylalanine residues. This modification offers high stability during acidic hydrolysis. Following acidic hydrolysis of the samples, the concentration of 13C9,15N1-labeled phenylalanine determined by LC-MS could be unambiguously correlated to the content of Pep19-2.5. Further experiments validated the accuracy of the data. Moreover, the quantification of Pep19-2.5 in different tissues (as studied in Wistar rats) was shown to provide comparable results to the results obtained with radioactively-labeled (14C) Pep19-2.5- Radioactive labeling is considered as the gold standard for quantification of compounds that refrain from reliable extraction methods. This novel method represents a valuable procedure for the determination of Pep19-2.5 and sticky peptides with unpredictable extraction properties in general.

Cervical laminoplasty is a valuable procedure for myelopathy but it is associated with complications such as increased kyphosis. The effect of ligament damage during cervical laminoplasty on biomechanics is not well understood. We developed the C2–C7 cervical spine finite element model and simulated C3–C6 double-door laminoplasty. Three models were created (a) intact, (b) laminoplasty-pre (model assuming that the ligamentum flavum (LF) between C3–C6 was preserved during surgery), and (c) laminoplasty-res (model assuming that the LF between C3–C6 was resected during surgery). The models were subjected to physiological loading, and the range of motion (ROM), intervertebral nucleus stress, and facet contact forces were analyzed under flexion/extension, lateral bending, and axial rotation. The maximum change in ROM was observed under flexion motion. Under flexion, ROM in the laminoplasty-pre model increased by 100.2%, 111.8%, and 98.6% compared to the intact model at C3–C4, C4–C5, and C5–C6, respectively. The ROM in laminoplasty-res further increased by 105.2%, 116.8%, and 101.8% compared to the intact model at C3–C4, C4–C5, and C5–C6, respectively. The maximum stress in the annulus/nucleus was observed under left bending at the C4–C5 segment where an increase of 139.5% and 229.6% compared to the intact model was observed for laminoplasty-pre and laminoplasty-res model, respectively. The highest facet contact forces were observed at C4–C5 under axial rotation, where an increase of 500.7% and 500.7% was observed compared to the intact model for laminoplasty-pre and laminoplasty-res, respectively. The posterior ligaments of the cervical spine play a vital role in restoring/stabilizing the cervical spine. When laminoplasty is performed, the surgeon needs to be careful not to injure the posterior soft tissue, including ligaments such as LF.

In a conventional interpretation of decision-making based on ambiguity, a decision-maker must prefer the best possible opportunity including various feasible possibilities. However, the dilemma of picking the best possible alternative has continued to be a substantial task to resolve. In this manuscript, we improve the existing complex intuitionistic fuzzy soft set (CIFSS), which includes the grade of truth and falsity with the rule that the sum of the real and imaginary parts of both grades is confined to [0, 1]. CIFS is a valuable procedure to determine the authenticity and consistency of the elaborated approaches. The fundamental laws and their related examples are also determined. Moreover, by using these laws, we investigated the complex intuitionistic fuzzy soft prioritized weighted averaging operator (CIFSPWAO), the complex intuitionistic fuzzy soft prioritized ordered weighted averaging operator (CIFSPOWAO), the complex intuitionistic fuzzy soft prioritized weighted geometric operator (CIFSPWGO), complex intuitionistic fuzzy soft prioritized ordered weighted geometric operator (CIFSPOWGO), and their related properties are also developed. Based on the developed operators, the multiattribute decision-making (MADM) tool is developed by using the explored operators based on CIFSS. Some numerical examples are also illustrated by using the investigated operators to determine the feasibility and consistency of the developed approaches. Finally, the comparative analysis and their geometrical manifestations are also determined to enhance the excellence of the performed explorations.

Objective: To determine the outcome of surgical removal of oral pyogenic granuloma in terms of post-operative pain (POP), post-operative healing and post-operative infection. Materials and methods: This observational study was conducted at Oral and Maxillofacial Surgery department, Liaquat University of Medical and Health Sciences Jamshoro/Hyderabad, from October 2018 to March 2019. Patients of oral pyogenic granuloma, 18-45 years old and either of gender were included. Surgical process of oral pyogenic granuloma were done under local anesthetic by giving infiltration surrounding deep oral pyogenic granuloma by 2% Xylocaine along with adrenaline. Patients were followed for three weeks on weekly basis to access post-operative complications including post-operative healing. Data was collected via study proforma. Results: Total 37 patients took part in the study; their mean age was 47.86+7.61 years. On 1st week assessment all patients were noted with history of mild pain, moderate pain and severe pain (19, 10 and 8 cases respectively), while healing was achieved in 19 patients. On second week assessment 25 patients exhibited mild pain and 6 exhibited moderate pain and healing was achieved by 16 patients. On 3rd week assessment, only 09 patients exhibited mild pain and mostly were healed few cases showed minor infection. Conclusion: It was concluded that surgical removal is valuable procedure for the management of pyogenic granuloma of oral cavity with lower rates of post-operative infection and pain, and highest rates of post-operative healing.

The principle of a complex interval-valued Pythagorean fuzzy set (CIVPFS) is a valuable procedure to manage inconsistent and awkward information genuine life troubles. The principle of CIVPFS is a mixture of the two separated theories such as complex fuzzy set and interval-valued Pythagorean fuzzy set which covers the truth grade (TG) and falsity grade (FG) in the form of the complex number whose real and unreal parts are the sub-interval of the unit interval. The superiority of the CIVPFS is that the sum of the square of the upper grade of the real part (also for an unreal part) of the duplet is restricted to the unit interval. The goal of this article is to explore the new principle of CIVPFS and its algebraic operational laws. By using the CIVPFSs, certain Einstein operational laws by using the t-norm and t-conorm are also developed. Additionally, we explore the complex interval-valued Pythagorean fuzzy Einstein weighted geometric (CIVPFEWG), complex interval-valued Pythagorean fuzzy Einstein ordered weighted geometric (CIVPFEOWG) operators and utilized their special cases. Moreover, a multicriteria decision-making (MCDM) technique is explored based on the elaborated operators by using the complex interval-valued Pythagorean fuzzy (CIVPF) information. To determine the consistency and reliability of the elaborated operators, we illustrated certain examples by using the explored principles. Finally, to determine the supremacy and dominance of the explored theories, the comparative analysis and graphical expressions of the developed principles are also discussed.

Abstract Background: combination treatment was identified as a valuable procedure to decrease breast tumorigenesis. It has been confirmed that microRNAs (miRs) can be considered as a therapeutic agent in breast cancer (BC) combination treatment. Our aim was the examination of potential action and underlying molecular activity of miR-424-5p and miR-142-3p combination in the human BC cells. Methods and results: MiR-424-5p and miR-142-3p mimics were transfected into MCF-7 cells, in a combination or separate manner. The proliferation of the cells was assessed through the MTT test. Also, apoptosis, autophagy, and cell cycle were analyzed by flow cytometry. Western blot and qRT-PCR determined the expression levels of downstream molecular factors of miR-424-5p and miR-142-3p. The results exhibited that miR-424-5p and miR-142-3p synergetically inhibited MCF-7 cell proliferation via apoptosis and autophagy induction and regulation of apoptotic factors, Bax and Bcl-2, and autophagy-associated gene, Beclin1 as well as proliferative factor c-Myc. Also, miR-424-5p and miR-142-3p combination treatment reduced MCF-7 cell colony numbers via modulating Oct-3 and resulted in G2 arrest through controlling STAT3 expression. Conclusions: The simultaneous restoration of miR-424-5p and miR-142-3p showed more cytotoxic effects on BC cells as compared to mono-treatment. Thus, the combination of two miRs (miR-424 and miR-142) could be used us as a new method for breast carcinoma treatment.

AbstractAvailable analytical methods cannot detect nanoplastics at environmentally realistic concentrations in complex matrices such as biological tissues. Here, we describe a one-step polymerization method, allowing direct radiolabeling of a sulfonate end-capped nano-sized polystyrene (nPS; proposed as a model nanoplastic particle representing negatively charged nanoplastics). The method, which produces nanoplastics trackable in simulated environmental settings which have already been used to investigate the behavior of a nanoplastic in vivo in a bivalve mollusc, was developed, optimized and successfully applied to synthesis of 14C-labeled nPS of different sizes. In addition to a description of the method of synthesis, we describe the details for quantification, mass balance and recovery of the labelled particles from complex matrices offered by the radiolabelling approach. The radiolabeling approach described here, coupled to use of a highly sensitive autoradiographic method for monitoring nanoplastic body burden and distributions, may provide a valuable procedure for investigating the environmental pathways followed by negatively charged nanoplastics at low predicted environmental concentrations. Whether the behaviour of the synthetic nPS manufactured here, synthesised using a very common inititator, represents that of manufactured nPS found in the environment, remains to be seen.

Abstract Background Colorectal cancer (CRC) is the most common tumor of the gastrointestinal tract. Anastomotic leak (AL) and prolonged postoperative ileus (PPOI) are two important complications of colorectal surgery. In this observational retrospective study, we evaluated the positive effects of transanal tube No Coil® in patients with CRC undergoing low anterior resection (LAR) and left hemicolectomy (LC). Methods Thirty-eight cases and forty controls resulted eligible for the final sample. No Coil® placement (SapiMed Spa, Alessandria, Italy) was considered an inclusion criteria for the case group. No Coil® was placed immediately after the end of surgical treatment. Results PPOI was significantly more frequent in the control group. AL was evident in 1 patient (2.6%) of cases and 3 patients (7.5%) of controls. No statistical difference was found in AL occurrence between groups. POI days and AL resulted associated with hospital stay. POI days were negatively associated with No Coil placement and positively with AL. Conclusion With our preliminary data, we suggest that No Coil® placement can be considered as a valuable procedure assisting colorectal surgery, but further studies are required to confirm and enlarge actual evidence.
