Try or Die: Dynamics of Plant Respiration and How to Survive Low Oxygen Conditions

Fluctuations in oxygen (O2) availability occur as a result of flooding, which is periodically encountered by terrestrial plants. Plant respiration and mitochondrial energy generation rely on O2 availability. Therefore, decreased O2 concentrations severely affect mitochondrial function. Low O2 concentrations (hypoxia) induce cellular stress due to decreased ATP production, depletion of energy reserves and accumulation of metabolic intermediates. In addition, the transition from low to high O2 in combination with light changes—as experienced during re-oxygenation—leads to the excess formation of reactive oxygen species (ROS). In this review, we will update our current knowledge about the mechanisms enabling plants to adapt to low-O2 environments, and how to survive re-oxygenation. New insights into the role of mitochondrial retrograde signaling, chromatin modification, as well as moonlighting proteins and mitochondrial alternative electron transport pathways (and their contribution to low O2 tolerance and survival of re-oxygenation), are presented.

Characterization of Oxygen Levels in an Uninfected and Infected Human Blood-Cerebrospinal-Fluid-Barrier Model

The host–pathogen interaction during meningitis can be investigated with blood-cerebrospinal-fluid-barrier (BCSFB) cell culture models. They are commonly handled under atmospheric oxygen conditions (19–21% O2), although the physiological oxygen conditions are significantly lower in cerebrospinal fluid (CSF) (7–8% O2). We aimed to characterize oxygen levels in a Streptococcus (S.) suis-infected BCSFB model with transmigrating neutrophils. A BCSFB model with human choroid plexus epithelial cells growing on transwell-filters was used. The upper “blood”-compartment was infected and blood-derived neutrophils were added. S. suis and neutrophils transmigrated through the BCSFB into the “CSF”-compartment. Here, oxygen and pH values were determined with the non-invasive SensorDish® reader. Slight orbital shaking improved the luminescence-based measurement technique for detecting free oxygen. In the non-infected BCSFB model, an oxygen value of 7% O2 was determined. However, with S. suis and transmigrating neutrophils, the oxygen value significantly decreased to 2% O2. The pH level decreased slightly in all groups. In conclusion, we characterized oxygen levels in the BCSFB model and demonstrated the oxygen consumption by cells and bacteria. Oxygen values in the non-infected BCSFB model are comparable to in vivo values determined in pigs in the CSF. Infection and transmigrating neutrophils decrease the oxygen value to lower values.

Effect of Artificial Regime Shifts and Biotic Factors on the Intensity of Foraging of Planktivorous Fish

It is still to be confirmed whether global warming with its predicted elevated water temperature will cause an increase in predation and alter phenological and physiological processes leading to changes in the size of aquatic organisms. In an experimental system of water column stratification simulating a natural combination of field conditions, we created artificial abiotic factors that mimicked the natural environment, i.e., light intensity, oxygen conditions, and thermal stratification. Subsequently, we added biotic factors such as algae, Daphnia, and planktivorous fish. We studied the intensity of foraging of planktivorous fish on individuals of Daphnia per min in different conditions of biotic and abiotic gradients. We demonstrated a possible scenario involving the risk of elimination of large prey within macrocladocera communities by predatory pressure as a result of climate change. A higher intensity of foraging of planktivorous fish caused or increased the occurrence of larger groups of planktonic animals with a smaller body size. The mechanisms of a future scenario were discovered at a higher trophic level in the aquatic environment.

Is Oxygenation Related to the Decomposition of Organic Matter in Cryoconite Holes?

Cryoconite is a sediment occurring on glacier surfaces worldwide which reduces ice albedo and concentrates glacier surface meltwater into small reservoirs called cryoconite holes. It consists of mineral and biogenic matter, including active microorganisms. This study presents an experimental insight into the influence of sediment oxygenation on the cryoconite ability to produce and decomposition of organic matter. Samples were collected from five glaciers in the Arctic and the European mainland. Cryoconite from three glaciers was incubated in stagnant and mechanically mixed conditions to imitate inter-hole water–sediment mixing by meltwater occurring on glaciers in Northern Hemisphere, and its effect on oxygen profiles and organic matter content. Moreover, we investigated short-term changes of oxygen conditions in cryoconite from four glaciers in illuminated and dark conditions. An anaerobic zone was present or approaching zero oxygen in all illuminated cryoconite samples, varying in depth depending on the origin of cryoconite: from 1500 µm from Steindalsbreen (Scandinavian Peninsula) and Forni Glacier (The Alps) to 3100 µm from Russell Glacier and Longyearbreen (Arctic) after incubation. Organic matter content varied between glaciers from 6.11% on Longyearbreen to 16.36% on Russell Glacier. The mixed sediment from the Forni Glacier had less organic matter than stagnant, the sediment from Longyearbreen followed this trend, but the difference was not statistically significant, while the sediment from Ebenferner did not differ between groups. Our results have implications for the understanding of biogeochemical processes on glacier surfaces, the adaptation of organisms to changing physical conditions due to abrupt sediment mixing, but also on the estimation of productivity of supraglacial systems.

INNOVATIVE METHOD OF NITRIFICATION AND DENITRIFICATION ON THE EXAMPLE OF WASTEWATER TREATMENT PLANT OF KOLOMYIA

The article is devoted to the unsatisfactory operation of biological wastewater treatment plants. At present in Ukraine there are no developments of equipment and devices to ensure the operation of treatment facilities using the technology of nitridenitrification of deep wastewater treatment from nitrogen and phosphorus compounds. The analysis of the equipment of biological treatment systems - nitri treatment plant - denitrifier, clarifier tanks is carried out. At the same time, the standards of the degree of purification of both Ukraine and the European Union were taken into account. On the example of treatment facilities in Kolomyia, Ivano-Frankovsk region, modern schemes of biological wastewater treatment for the removal of nitrogen and phosphorus have been proposed, namely, the re-equipment of single-corridor aerotanks into bioreactors of nitridenitrifiers by creating zones with different oxygen conditions in their volume.

Omics-Inferred Partitioning and Expression of Diverse Biogeochemical Functions in a Low-O 2 Cyanobacterial Mat Community

Cyanobacterial mats are dense communities of microorganisms that contain photosynthetic cyanobacteria along with a host of other bacterial species that play important yet still poorly understood roles in this ecosystem. Although such cyanobacterial mats were critical agents of Earth’s biological and chemical evolution through geological time, little is known about how they function under the low-oxygen conditions that characterized most of their natural history.

Oxygen-Dependent Changes in the N-Glycome of Murine Pulmonary Endothelial Cells

Supplemental oxygen is frequently used together with mechanical ventilation to achieve sufficient blood oxygenation. Despite the undoubted benefits, it is vigorously debated whether too much oxygen can also have unpredicted side-effects. Uncertainty is also due to the fact that the molecular mechanisms are still insufficiently understood. The lung endothelium is covered with an exceptionally broad glycocalyx, carrying N- and O-glycans, proteoglycans, glycolipids and glycosaminoglycans. Glycan structures are not genetically determined but depend on the metabolic state and the expression level and activity of biosynthetic and glycan remodeling enzymes, which can be influenced by oxygen and the redox status of the cell. Altered glycan structures can affect cell interactions and signaling. In this study, we investigated the effect of different oxygen conditions on aspects of the glycobiology of the pulmonary endothelium with an emphasis on N-glycans and terminal sialylation using an in vitro cell culture system. We combined a proteomic approach with N-glycan structure analysis by LC-MS, qRT-PCR, sialic acid analysis and lectin binding to show that constant and intermittent hyperoxia induced time dependent changes in global and surface glycosylation. An siRNA approach identified St6gal1 as being primarily responsible for the early transient increase of α2-6 sialylated structures in response to hyperoxia.

The microbial metabolic activity on carbohydrates and polymers impact the biodegradability of landfilled solid waste

Biological waste degradation is the main driving factor for landfill emissions. In a 2-year laboratory experiment simulating different landfill in-situ aeration scenarios, the microbial degradation of solid waste under different oxygen conditions (treatments) was investigated. Nine landfill simulation reactors were operated in triplicates under three distinct treatments. Three were kept anaerobic, three were aerated for 706 days after an initial anaerobic phase and three were aerated for 244 days in between two anaerobic phases. In total, 36 solid and 36 leachate samples were taken. Biolog® EcoPlates™ were used to assess the functional diversity of the microbial community. It was possible to directly relate the functional diversity to the biodegradability of MSW (municipal solid waste), measured as RI4 (respiration index after 4 days). The differences between the treatments in RI4 as well as in carbon and polymer degradation potential were small. Initially, a RI4 of about 6.5 to 8 mg O2 kg−1 DW was reduced to less than 1 mg O2 kg−1 DW within 114 days of treatment. After the termination of aeration, an increase 3 mg O2 kg−1 DW was observed. By calculating the integral of the Gompertz equation based on spline interpolation of the Biolog® EcoPlates™ results after 96 h two substrate groups mainly contributing to the biodegradability were identified: carbohydrates and polymers. The microbial activity of the respective microbial consortium could thus be related to the biodegradability with a multilinear regression model.

The Reactivity of Polyethylene Microplastics in Water under Low Oxygen Conditions Using Radiation Chemistry

Polyethylene (PE) is an intensely utilized polymer, which has consequently led to it becoming a common environmental contaminant. PE and other plastic waste are known to be highly persistent in surface waters; however, chemical and physical changes do take place over time, dependent mostly on highly variable natural conditions, such as oxygen (O2) availability. Gamma radiation was used to generate reactive oxygen species, namely hydroxyl radicals, in initially aerated aqueous solutions to simulate the natural weathering of microplastics in waters where there are fluctuations and often depletions in dissolved O2. The headspace of the irradiated PE-containing solutions was probed for the formation of degradation products using solid-phase microextraction (SPME) fibers in combination with gas chromatography mass spectrometry (GCMS). The major species detected were n-dodecane, with trace levels of tridecane, 2-dodecanone, and hexadecane, which were believed to be predominately adsorbed in the PE microplastics in excess of their aqueous solubility limits. Surface characterization by Raman spectroscopy and light and dark field microscopy indicated no change in the chemical composition of the irradiated PE microplastics under low O2 to anaerobic conditions. However, morphological changes were observed, indicating radical combination reactions.
