Here we show that multiple modes of Notch signaling activation specify the complexity of spatial cellular interactions necessary for stem cell niche assembly. In particular, we studied the formation of the germline stem cell niche in Drosophila ovaries, which is a two-step process whereby terminal filaments are formed first. Then, terminal filaments signal to the adjacent cap cell precursors, resulting in Notch signaling activation, which is necessary for the lifelong acquisition of stem cell niche cell fate. The genetic data suggest that in order to initiate the process of stem cell niche assembly, Notch signaling is activated among non-equipotent cells via distant induction, where germline Delta is delivered to somatic cells located several diameters away via cellular projections generated by primordial germ cells. At the same time, to ensure the robustness of niche formation, terminal filament cell fate can also be induced by somatic Delta via cis- or trans-inhibition. This exemplifies a double security mechanism that guarantees that the germline stem cell niche is formed, since it is indispensable for the adjacent germline precursor cells to acquire and maintain stemness necessary for successful reproduction. These findings contribute to our understanding of the formation of stem cell niches in their natural environment, which is important for stem cell biology and regenerative medicine.

Abstract Campanula tommasiniana is a typical chasmophyte occupying calcareous rock crevices and cracks along a wide range of ecological gradients, demonstrating a high degree of ecological plasticity and stress tolerance with regards to abiotic factors. Generally, three ecologically and floristically distinct groups of stands were recognized and typified according to a sigmatistic approach: (a) Seslerio juncifoliae-Campanuletum tommasinianae ass. nov., with stands occupying higher elevated sites fully exposed to sun and strong winds; (b) Seslerio autumnalis-Campanuletum tommasinianae ass. nov., representing stands predominantly developed within thermophytic beech stands, semi- to fully- -shaded by the tree canopy; (c) Cystopteri fragilis-Campanuletum tommasinianae, sciophytic, stands adapted to moisture and coldwith high frequency and coverage of bryophytes. Results of DCA analyses using a unimodal model suggest that Campanula tommasiniana is primarily a plant of open and exposed sites of higher elevation despite being most frequently found in rock crevices within thermophytic and altimontane beech forests.

Ecological approaches to community assembly have emphasized the interplay between neutral processes, niche-based environmental filtering and niche-based species sorting in an interactive milieu. Recently, progress has been made in terms of aligning our vocabulary with conceptual advances, assessing how trait-based community functional parameters differ from neutral expectation and assessing how traits vary along environmental gradients. Experiments have confirmed the influence of these processes on assembly and have addressed the role of dispersal in shaping local assemblages. Community phylogenetics has forged common ground between ecologists and biogeographers, but it is not a proxy for trait-based approaches. Community assembly theory is in need of a comparative synthesis that addresses how the relative importance of niche and neutral processes varies among taxa, along environmental gradients, and across scales. Towards that goal, we suggest a set of traits that probably confer increasing community neutrality and regionality and review the influences of stress, disturbance and scale on the importance of niche assembly. We advocate increasing the complexity of experiments in order to assess the relative importance of multiple processes. As an example, we provide evidence that dispersal, niche processes and trait interdependencies have about equal influence on trait-based assembly in an experimental grassland.
