Amitraz Poisoning: A Rare Case of an Unusual Pesticide Poisoning

Amitraz is a pesticide used in animals and agriculture. It consists of triazapentadiene which is a centrally acting alpha2 adrenergic agonist. Amitraz poisoning is uncommon in human beings. It occurs via the oral, dermal or inhalational route. The clinical manifestations are mainly central nervous system depression, respiratory depression and cardiovascular system depression. How long is long enough? Decreasing effects in Aedes aegypti larval mortality by plant extracts over time

Aedes aegypti has overcome all kinds of mosquito control attempts over the last century. Strategies for population control resorts to the use of synthetic insecticides, which can lead to problems like human intoxication and environmental contamination. The effects of Bacillus thuringiensis var. israelensis (Bti), Ilex paraguariensis (yerba mate), and Ilex theezans (caúna herb) extracts against A. aegypti larvae were evaluated. The bioassays were conducted under controlled laboratory conditions of temperature (27 ± 3°C) and photoperiod (12 h). Hydroalcoholic extract of the leaves of I. theezans displayed better residual effect compared to the aqueous extract of I. paraguariensis fruits. The strongest residual effect of I. theezans was probably due to the presence of certain chemicals in its leaves, such as coumarins, hemolytic saponins, and cyanogenic glucosides, which were absent in I. paraguariensis. The results herein contributed to the prospection of natural insecticides and opened the possibility for subsequent studies on the use of plant extracts in field situations in a short-time scale.

Investigating a human pesticide intoxication incident: The importance of robust analytical approaches

A human intoxication incident attributed to pesticide abuse was investigated using cutting-edge analytical methodologies. An LC-ESI-MS/MS method, based on a hybrid solid-phase extraction protocol (hybrid-SPE), was applied for the detection and quantification of several pesticides and metabolites in human biological fluids. Concomitantly, an UHPLC-HRMS method was applied to investigate potential metabolites, assisted by a complementary GC-MS method to elucidate the presence of plausible pesticides co-formulants. The LC-ESI-MS/MS method exhibited acceptable mean recoveries at the lower limit of quantification (LLOQ) and three additional levels, varying from 85 to 106% for all analytes and matrices. In serum, urine, and gastric fluid samples, the suspect compounds, namely chlorpyrifos and myclobutanil, predominated. Gastric fluid samples contained the highest concentrations of chlorpyrifos (39,800 ng/mL) and myclobutanil (18,800 ng/mL), while the neonicotinoid imidacloprid was also quantified, below 30 ng/mL. Notwithstanding, the UHPLC-HRMS analysis unveiled several metabolites of chlorpyrifos and myclobutanil. In parallel, GC-MS analysis, corroborated the presence of several co-formulants in gastric fluid samples, exemplified by m- and o-xylene, and cyclohexanone. Overall, three analytical methods were implemented to elucidate the chemical causality of a human intoxication incident. The presence of suspected active substances, one additional, and several metabolites and co-formulants were documented.

[D-Leu1]MC-LR and MC-LR: A Small–Large Difference: Significantly Different Effects on Phaseolus vulgaris L. (Fabaceae) Growth and Phototropic Response after Single Contact during Imbibition with Each of These Microcystin Variants

[D-Leu1]MC-LR and MC-LR, two microcystins differing in one amino acid, constitute a sanitary and environmental problem owing to their frequent and concomitant presence in water bodies of the Americas and their association with human intoxication during recreational exposure to cyanobacterial bloom. Present in reservoirs used for irrigation as well, they can generate problems in the development of crops such as Phaseolus vulgaris, of nutritional and economic interest to the region. Although numerous works address the toxic effects of MC-LR, information on the toxicity of [D-Leu1]MC-LR is limited. Our objective was to study the toxic effects of [D-Leu1]MC-LR and MC-LR (3.5 µg/ml) on P. vulgaris after a single contact at the imbibition stage. Our findings indicate that 10 days post treatment, [D-Leu1]MC-LR generates morphological and physiological alterations more pronounced than those caused by MC-LR. In addition to the alterations produced by [D-Leu1]MC-LR in the development of seedlings and the structure of the leaves, roots and stems, we also found alterations in leaf stomatal density and conductivity, a longer delay in the phototropic response and a decrease in the maximum curvature angles achieved with respect to that observed for MC-LR. Our findings indicate that these alterations are linked to the greater inhibition of phosphatase activity generated by [D-Leu1]MC-LR, rather than to oxidative damage. We observed that 30 days after treatment with MC-LR, plants presented better development and recovery than those treated with [D-Leu1]MC-LR. Further studies are required on [D-Leu1]MC-LR and MC-LR toxicity and their underlying mechanisms of action.

Phytoplankton Dynamics in the Middle Adriatic Estuary, with a Focus on the Potentially Toxic Genus Pseudo-nitzschia

The Krka River estuary is a karstic, permanently stratified estuary due to the strong freshwater inflow. It is a special environment in which to study the phytoplankton community, especially because this area is an important aquaculture site. Among other potentially toxic phytoplankton species, the diatom genus Pseudo-nitzschia occurs frequently and is a potential source of domoic acid (DA), causing shellfish toxicity and human intoxication. The main objective was to examine the dynamics of the phytoplankton community and, in particular, the genus Pseudo-nitzschia in the upper part of the Krka estuary, through monthly sampling over two years. The phytoplankton community was analysed using light microscopy and scanning electron microscopy to determine the diversity of Pseudo-nitzschia species and characterise the environmental parameters associated with a high abundance of Pseudo-nitzschia species. Seven Pseudo-nitzschia species were identified in the investigation, with higher frequencies and abundances in the less variable layer, at a 7 m depth. Blooms of Pseudo-nitzschia were noted in the late summer/early autumn, dominated by P. delicatissima/arenysensis. Winter assemblages were characterised by P. pseudodelicatissima/cuspidata, P. calliantha, and P. subfraudulenta, and were associated with domoic acid occurrence in shellfish.

Survey of Tetrodotoxin in New Zealand Bivalve Molluscan Shellfish over a 16-Month Period

Tetrodotoxin (TTX) is a heat-stable neurotoxin typically associated with pufferfish intoxications. It has also been detected in shellfish from Japan, the United Kingdom, Greece, China, Italy, the Netherlands and New Zealand. A recent European Food Safety Authority (EFSA) scientific opinion concluded that a level of <0.044 mg TTX/kg in marine bivalves and gastropods, based on a 400 g portion size, does not result in adverse effects in humans. There have been no reports of human illness attributed to the consumption of New Zealand shellfish containing TTX. To obtain a greater understanding of its presence, a survey of non-commercial New Zealand shellfish was performed between December 2016 and March 2018. During this period, 766 samples were analysed from 8 different species. TTX levels were found to be low and similar to those observed in shellfish from other countries, except for pipi (Paphies australis), a clam species endemic to New Zealand. All pipi analysed as part of the survey were found to contain detectable levels of TTX, and pipi from a sampling site in Hokianga Harbour contained consistently elevated levels. In contrast, no TTX was observed in cockles from this same sampling site. No recreationally harvested shellfish species, including mussels, oysters, clams and tuatua, contained TTX levels above the recommended EFSA safe guidance level. The levels observed in shellfish were considerably lower than those reported in other marine organisms known to contain TTX and cause human intoxication (e.g., pufferfish). Despite significant effort, the source of TTX in shellfish, and indeed all animals, remains unresolved making it a difficult issue to understand and manage.

Subacute Testicular Toxicity to Cadmium Exposure Intraperitoneally and Orally

The toxic effects of cadmium (Cd) on reproductive parameters are widely described in the literature. Experimental models often make use of the intraperitoneal route (i.p.), although human intoxication occurs preferentially by the oral route and can be continuous. However, little is known about the effect of Cd administration routes on the testicular structure. Thus, this study investigated the testicular impact of Cd exposure comparing both i.p. and oral routes, both single dose (SD), in addition to the oral route in fractional doses (FD). Swiss adult male mice received CdCl2 1.5 mg/kg i.p., 30 mg/kg oral SD, and 4.28 mg/kg oral FD for 7 consecutive days. The Cd bioaccumulation was observed in all routes, mainly in the oral FD route. The concentrations of testicular Ca and Cu decreased in all animals exposed to Cd, while Zn and Mn decreased only in the i.p. route. Testicular SOD activity was reduced in both routes of oral administration, while CAT increased in the i.p. route, and GST increased in all animals exposed to Cd. Changes in the tubular parameters and cell viability were observed in both routes of Cd administration but were more intense in the oral route, mainly in the FD. Serum testosterone concentration was reduced in both routes of oral administration. Tubular damage, such as the vacuolization of the seminiferous epithelium, germ cell detachment, and seminiferous tubule degeneration, occurred in all groups exposed to Cd. Therefore, the oral Cd administration presented greater potential to promote testicular damage, mainly when the metal was given in a fractionated way.

Apprehension of illegal pesticides, agricultural productivity and food poisoning on the Brazilian state of Mato Grosso do Sul

The application of agrochemicals plays a key role in the world's food and agricultural products supply. Brazil, as one of the most relevant agricultural global players is highly dependent on the application of such products, which surely attracted not only companies that licitly commercialized the pesticides but also the so termed pesticide smugglers. Here, we conducted a retrospective survey (i.e., from 2010 to 2017) regarding the apprehension of illegal products, agricultural productivity and food poisoning in the state of Mato Grosso do Sul (MS). The statistical data were obtained by official agencies Such as Technical-Scientific Sector of the Federal Police Department of the MS State, The System of Control of Seized Goods, The National Toxic-Pharmacological Information System and The Brazilian Institute of Geography and Statistics, and by the non-official National Union of the Product Industry for Plant Protection agency. The results indicated that in 2011 the MS state had the largest number of seizures of illegal pesticides compared to other years. In addition, from 2010 to 2013, the state was among the regions with the highest number of seizures. In the same period, the agricultural productivity of the cotton, sugarcane, corn and soybean crops was lower compared to the other years. Regarding the food poisoning, the percentage of human intoxication by agrochemicals for agricultural use was low compared to all cases of intoxication by various toxic agents. It was concluded that the state MS presented a large amount of seizure of illegal pesticides.

Pinnatoxins’ Deleterious Effects on Cholinergic Networks: From Experimental Models to Human Health

Pinnatoxins (PnTXs) are emerging neurotoxins that were discovered about 30 years ago. They are solely produced by the marine dinoflagellate Vulcanodinium rugosum, and may be transferred into the food chain, as they have been found in various marine invertebrates, including bivalves. No human intoxication has been reported to date although acute toxicity was induced by PnTxs in rodents. LD50 values have been estimated for the different PnTXs through the oral route. At sublethal doses, all symptoms are reversible, and no neurological sequelae are visible. These symptoms are consistent with impairment of central and peripheral cholinergic network functions. In fact, PnTXs are high-affinity competitive antagonists of nicotinic acetylcholine receptors (nAChRs). Moreover, their lethal effects are consistent with the inhibition of muscle nAChRs, inducing respiratory distress and paralysis. Human intoxication by ingestion of PnTXs could result in various symptoms observed in episodes of poisoning with natural nAChR antagonists. This review updates the available data on PnTX toxicity with a focus on their mode of action on cholinergic networks and suggests the effects that could be extrapolated on human physiology.
