SYNERGISTIC IMMUNOMODULATORY ACTIVITY OF AQUEOUS ROOT EXTRACT OF ASPARAGUS RACEMOSUS WILLD AND ETHANOL WHOLE PLANT EXTRACT OF BOERHAVIA DIFFUSA LINN

Objective: On the basis of traditional use and Ethno pharmacological evidences Boerhavia diffusa whole plant and root part of Asparagus racemosus (Shatavari) both are widely used to enhance the immunity. But in combination there is no scientific evidence so current study was designed. Methods: Laboratory based study, namely, carbon clearance, cyclophosphamide induced immune suppression and neutrophil adhesion was designed using mice as an experimental animals in different combination of aqueous whole plant extract of B. diffusa and methanol extract of root part of A. racemosus were used as a test drug in the ratio 1:1, 1:2, and 2:1 (100 mg/kg) against the established standard drug Ashwagandha. Results: The results revealed that animals treated with combined extract (1:1, 1:2, and 2:1) at a dose of 100 mg/kg increase rate of carbon clearance from blood, there is significance alternation in blood parameter in cyclophosphamide group and also improve the Neutrophil adhesion when treated with different combination of polyherbal formulation treated groups. Conclusion: The polyherbal formulation in different ratio showing good significant immunomodulatory activity as compare to standard.

Uji Efek Imunumodulator Vco (Virgin Coconut Oil) Pada Tikus Jantan

(Article History: Received August 25, 2021; Revised Sept 17, 2021; Accepted Sept 20, 2021) ABSTRAKVirgin coconut oil merupakan minyak kelapa murni.VCO mengandung Medium Chain Fatty Acid (MCFA) yang memiliki pengaruh dalam meningkatkan sistem imunitas. Penelitian ini bertujuan untuk menguji efek imunomodulator VCO terhadap aktivitas fagositosis dan nilai titer antibodi pada tikus jantan. Kelompok uji bersihan karbon dibagi menjadi 5 kelompok dengan masing-masing terdiri dari 5 tikus yaitu kelompok suspensi CMC Na 0,5%, Imboost dosis 30 mg/kgBB, virgin coconut oil 5 ml/KgBB, 10 ml/KgBB,15 ml/KgBB. Perlakuan diberikan sehari sekali selama tujuh hari dan hari ke delapan dilakukan uji imunomodulator dengan metode bersihan karbon. Kelompok perlakuan uji titer antibodi sama seperti di atas selain kontrol positif menggunakan levamisole dosis 25 mg/kgBB diberikan secara oral selama 14 hari. Hasil penelitian menunjukkan bahwa VCO 5 ml/KgBB, 10 ml/KgBB, 15 ml/KgBB meningkatkan aktivitas fagositosis sel makrofag dibandingkan dengan Na-CMC 0,5% (P<0,05) dan berpengaruh secara signifikan meningkatkan pembentukan antibodi sel imun tikus jantan.Kata Kunci: Virgin coconut oil; imunomodulator; fagositosis; titer antibodi ABSTRACTVirgin coconut oil (VCO) is pure coconut oil. VCO contains Medium Chain Fatty Acid (MCFA) which has an influence increased immune system. This study was to determine the immunomodulatory effect of VCO on phagocytic activity and antibody titer values in male rats.The carbon clearance test group was divided into 5 groups with 5 rats each of 0.5% CMC Na suspension, Imboost dose of 30 mg/kgBW, Virgin coconut oil 5 ml/KgBW, 10 ml/KgBW, 15 ml/KgBW. The treatment was given once a day for seven days, on the eight day immunomodulatory test was performed use carbon clearance method. The antibody titer test treatment group was same, except for the positive control, levamisole at a dose of 25 mg/kgBW orally for 14 days. The results showed that VCO 5 ml/KgBB, 10 ml/KgBB, 15 ml/KgBB significantly increased macrophage cell phagocytic activity compared to 0.5% Na-CMC (P<0.05) and could increase cell antibody formation immunity of male mice significantly.Keywords: Virgin coconut oil; immunomodulator; phagocytosis; antibody titer

Immunomodulatory activity of Alcoholic extracts of Tinospora cordifolia Stem

Tinospora cordifolia is a plant well known for its medicinal value in Indian ayurveda and Indian traditional medicine system. However, to prove its efficiency for the clinical utilization, more experimental data will be beneficial. In the present investigation, evaluated the immunomodulatory activity of the alcoholic extracts of Tinospora cordifolia stem on various in-vivo experimental models such as neutrophil adhesion test, phagocytic index by carbon clearance test, Hemagglutinating antibody (HA) titre and delayed type hypersensitivity (DTH) responses. The evaluation of immunomodulatory potential by oral administration of alcoholic stem extracts (50, 100, 200 and 300mg/kg b.w, p.o) evoked a significant increase in percent neutrophil adhesion to nylon fibers as well as a dose dependent increased in antibody titre values, and potentiated delayed type hypersensitivity reaction induced by sheep red blood cells and significant response towards phagocytosis in carbon clearance assay (*p<0.05, *p<0.01, ***p<0.001). This may be due to concentrations of active phytochemicals present in particular plant extract. Hence, it was concluded that the plant extracts increased humoral as well as cell mediated immunity.

Immunomodulator Activity of Puguntano (Picria fel-terrae Lour.) Extract in White Male Mice By Carbon Clearance Method

Poguntano Herb Plants (Picria fel-terrae Lour.) contains flavonoids compounds that have been potentially to developed into immunomodulators. The purpose of this study was to determine the immunomodulatory activity of Poguntano herb extract (Picria fel-terrae Lour.), with the method of carbon clearance. n-hexane extract puguntano herb(ENHHP), extract of ethyl acetate of Poguntano herb (EEAHP), and ethanol extract of puguntano herb(EEHP) Poguntano multistage results in the test of immunomodulatory activity with a method of carbon clearance. Results showed that the EEHP 200 mg/kg BB showed a strong immunostimulation effect, EEAHP 200 mg/kg BB showed strong immunostimulation effect, ENHP 200 mg/kg BB demonstrated strong immunostimulation effect. It is thought that extract N-hexane herb Poguntano (ENHHP), extracts of ethyl herb acetate Poguntano (EEAHP), and herb ethanol extract Poguntano (EEHP) Poguntano herb may increase phagocytosis activity. Extract of ethyl acetate Poguntano herb 200 mg/kg bw (EEAHP) has the best phagocytosis activity with a value of 2.376 which indicates there is a significant difference (P < 0.05) with another treatment group.

EVALUATION OF IN VIVO IMMUNOMODULATORY ACTIVITY OF AQUEOUS AND ETHANOLIC EXTRACT OF CAPPARIS MOONII

Objective: The present study was intended to evaluate the in vivo immunomodulatory activity of aqueous and ethanolic extract of dried fruits of Capparis moonii Wight. Methods: The effect of both the extracts was evaluated by carbon clearance assay in mice, and they showed a significant increase in the phagocytic index at 50, 100, and 200 mg/kg dose. The effect of the extracts on delayed-type hypersensitivity (DTH) and antibody titer assay was evaluated in ovalbumin immunized mice. Results: Both extracts showed a significant effect on DTH response and stimulation of antibody titer at 200 mg/kg dose. The effects of the extracts in cyclophosphamide-induced myelosuppression mice were also statistically significant. Conclusion: The studies demonstrated specific and non-specific immunostimulating properties of both the ethanol and aqueous extract of C. moonii fruits in various in vivo experimental methods suggesting its therapeutic usefulness in immunocompromised conditions.

Effects of Polysaccharide from Malus halliana Koehne Flowers in Cyclophosphamide-Induced Immunosuppression and Oxidative Stress on Mice

The immunomodulatory effects of Malus halliana flower polysaccharide (MHFP) were investigated in this paper. The model of immunosuppressive mice was established by cyclophosphamide, which was treated with different dosages of MHFP (600, 400, and 200 mg/kg·d-1). The results showed that MHFP significantly increased the index of the spleen and thymus and improved the atrophy of immune organs. MHFP enhanced the ability of carbon clearance and phagocytosis of mononuclear phagocytes in mice. Meanwhile, MHFP promoted the proliferation of splenic lymphocytes. MHFP could enhance the content of serum hemolysin and improve the decrease of hemolysin induced by cyclophosphamide. The contents of ACP and LDH in the serum and spleen were determined, indicating that MHFP could enhance the activity of macrophages. MHFP promoted the content of cytokines (IL-2, IL-6, TNF-α, and IFN-γ) and mRNA expression. At the same time, the pathological changes of the spleen tissue also showed that MHFP could improve the immunosuppression induced by cyclophosphamide. In addition, MHFP increased the content of SOD, T-AOC, and CAT in the serum and spleen tissue, decreased the level of MDA, and improved the oxidative stress caused by cyclophosphamide. In conclusion, MHFP could effectively improve the immunosuppression and oxidative stress induced by cyclophosphamide and enhance the immune capacity of mice.

IMMUNOMODULATORY ACTIVITY OF MUNTINGIA CALABURA L. FRUITS USING CARBON CLEARANCE ASSAY AND THEIR TOTAL FLAVONOID AND PHENOLIC CONTENTS

Objective: Muntingia calabura L fruits have several advantages for health and can be consumed as alternative medicine. This study aims to evaluate the immunomodulatory activity of methanol extract (ME) and fractions of M. calabura L fruits against non-specific immune response in mice and its total phenolic and flavonoid contents. Methods: This research using carbon clearance assay or phagocytosis method which was done on mice. Seventy male Swiss mice were divided into 14 groups. Mice were treated with ME, hexane fraction (HF), dichloromethane fraction (DF), and ethyl acetate fraction (EAF) of M. calabura L. orally at a dose of 50, 100, and 200 mg/kg daily for 7 days. Zymosan with a dose of 15 mg/kg was used as a positive control and given intraperitoneally, and CMC Na was used as a negative control. On the 8th day, mice were injected with carbon ink suspension intravenously and blood samples were taken at the minutes of 0, 5, 10, 15, 20, and 30. Furthermore, the transmittance was measured at λ 675 nm. The total phenolic content was measured using the Folin–Ciocalteu method, and flavonoid content was measured using the AlCl3 method. Results: Zymosan, ME, and EAF of M. calabura L. at a dose of 200 mg/kg showed a strong immunostimulatory effect (phagocytic index >1.5) while the HF and DF groups showed that the higher doses decreased the phagocytic index. The total phenolic contents of ME, HF, DF, and EAF were 27.90, 11.11, 16.72, and 30.11%, respectively, while the flavonoid contents were 4.07, 0.17, 3.07, and 1.86%, respectively. Conclusion: ME and EAF of M. calabura L. fruits have immunostimulatory activity.

Uji Aktivitas Immunomodulator Berbagai Tanaman Famili Piperaceae pada Mencit Galur Balb/C dengan Metode Carbon Clearance

Tanaman keluarga Piperaceae tersebar luas di daerah tropis dan telah banyak digunakan sebagai tanaman obat, diantaranya adalah kemukus (Piper cubeba), kiseureuh (Piper aduncum), dan cabe jawa (Piper retrofractum). Tujuan penelitian adalah untuk mengevaluasi aktivitas imunomodulator ekstrak etanol beberapa tanaman dari keluarga Piperaceae pada mencit jantan galur Balb/c dengan metode carbon clearance. Penelitian dilakukan dengan membagi hewan coba menjadi 12 kelompok, yaitu kelompok kontrol normal, pembanding imunosupresan, pembanding imunostimulan, serta masing-masing tiga kelompok variasi dosis kemukus, kiseureuh, dan cabe jawa. Parameter aktivitas imunomodulator ditinjau dari indeks fagositosis, kadar leukosit darah tepi, dan kadar leukosit limpa. Hasil penelitian menunjukkan Ekstrak etanol kemukus (Piper cubeba) memiliki aktivitas imunomodulator sebagai imunosupresan, peningkatan dosis meningkatkan efek imunosupresan. Ekstrak etanol kiseureh (Piper aduncum) dosis rendah memiliki aktivitas imunomodulator sebagai imunosupresan, sedangkan pada dosis sedang dan tinggi memiliki kecenderungan sebagai imunostimulan. Ekstrak etanol cabe jawa (Piper retrofractum) memiliki aktivitas imunomodulator sebagai imunostimulan, dengan aktivitas tertinggi dicapai pada dosis sedang. Ekstrak etanol kemukus (Piper cubeba), kiseureh (Piper aduncum), dan cabe jawa (Piper retrofractum) mempengaruhi jumlah komponen granulosit darah tepi, dimana peningkatan neutrofil mempengaruhi peningkatan efek fagositosis. Ekstrak etanol kemukus (Piper cubeba), kiseureh (Piper aduncum), dan cabe jawa (Piper retrofractum) tidak mempengaruhi jumlah komponen granulosit limpa.

A Comparative Study on the Effects of Different Parts of Panax ginseng on the Immune Activity of Cyclophosphamide-Induced Immunosuppressed Mice

The objective of the present study was to compare the effects of the immunological activity of various parts (root/stem/leaf/flower/seed) of five-year-old ginseng on the immune system of immunosuppressive mice. Immunosuppression was induced by cyclophosphamide (CTX) in the mouse model, whereas levamisole hydrochloride tablet (LTH) was used for the positive control group. We found that ginseng root (GRT), ginseng leaf (GLF), and ginseng flower (GFR) could relieve immunosuppression by increased viability of NK cells, enhanced immune organ index, improved cell-mediated immune response, increased content of CD4+ and ratio of CD4+/CD8+, and recovery of macrophage function, including carbon clearance, phagocytic rate, and phagocytic index, in immunodeficient mice. However, ginseng stem (GSM) and ginseng seed (GSD) could only enhance the thymus indices, carbon clearance, splenocyte proliferation, NK cell activities, and the level of IL-4 in immunosuppressed mice. In CTX-injected mice, GRT and GFR remarkably increased the protein expression of Nrf2, HO-1, NQO1, SOD1, SOD2, and CAT in the spleen. As expected, oral administration of GRT and GFR markedly enhanced the production of cytokines, such as IL-1β, IL-4, IL-6, IFN-γ, and TNF-α, compared with the CTX-induced immunosuppressed mice, and GRT and GFR did this relatively better than GSM, GLF, and GSD. This study provides a theoretical basis for further study on different parts of ginseng.
