Optical Dielectrophoretic (DEP) Manipulation of Oil-Immersed Aqueous Droplets on a Plasmonic-Enhanced Photoconductive Surface

We present a plasmonic-enhanced dielectrophoretic (DEP) phenomenon to improve optical DEP performance of a floating electrode optoelectronic tweezers (FEOET) device, where aqueous droplets can be effectively manipulated on a light-patterned photoconductive surface immersed in an oil medium. To offer device simplicity and cost-effectiveness, recent studies have utilized a polymer-based photoconductive material such as titanium oxide phthalocyanine (TiOPc). However, the TiOPc has much poorer photoconductivity than that of semiconductors like amorphous silicon (a-Si), significantly limiting optical DEP applications. The study herein focuses on the FEOET device for which optical DEP performance can be greatly enhanced by utilizing plasmonic nanoparticles as light scattering elements to improve light absorption of the low-quality TiOPc. Numerical simulation studies of both plasmonic light scattering and electric field enhancement were conducted to verify wide-angle scattering light rays and an approximately twofold increase in electric field gradient with the presence of nanoparticles. Similarly, a spectrophotometric study conducted on the absorption spectrum of the TiOPc has shown light absorption improvement (nearly twofold) of the TiOPc layer. Additionally, droplet dynamics study experimentally demonstrated a light-actuated droplet speed of 1.90 mm/s, a more than 11-fold improvement due to plasmonic light scattering. This plasmonic-enhanced FEOET technology can considerably improve optical DEP capability even with poor-quality photoconductive materials, thus providing low-cost, easy-fabrication solutions for various droplet-based microfluidic applications.

Dynamic Wetting of Photo-Responsive Arylazopyrazole Monolayers is Controlled by the Molecular Kinetics of the Monolayer

Smart surfaces that can change their wetting behavior on demand are interesting for applications such as self-cleaning surfaces or lab-on-a-chip devices. In order to functionalize aluminum oxide surfaces, we have synthesized arylazopyrazole phosphonic acids (butyl-AAP-C18PA) that represent a new class of photoswitchable molecules for these oxide surfaces. Butyl-AAP-C18PA monolayers were deposited on alpha-Al2O3(0001) and show reversible E/Z photo-switching with UV (Z) and green (E) light that can trigger contact angle changes of up to ~10°. We monitored these changes on the macroscopic level by recording the dynamic contact angle while the monolayer was switched in situ from the E to the Z state. On the molecular level, time-resolved vibrational sum-frequency generation (SFG) spectroscopy provided information on the kinetic changes within the AAP monolayer and the relevant characteristic time scales for E to Z switching and vice versa. In addition, vibrational SFG at different relative humidity indicates that the thermal stability of the Z configuration is largely influenced by the presence of water and that water can stabilize the Z state and, thus, hinder the AAP monolayer to switch into the E state when it is immersed in H2O. Having established the characteristic times for switching on the molecular scale from SFG spectroscopy, we additional measure the dynamic contact angle. Further, we reveal the time scales of the coupled substrate and droplet dynamics which we have extracted individually. For that, we report on a relaxation model, that can be solved analytically and which is verified via comparison with simulations of a Lennard Jones system and a comparison with experimental data. Indeed, our modelling of these coupled relaxation processes allows us to predict the non-trivial variation of the time-dependence of the contact angle when changing the size of the droplet. The observed slowing-down for E to Z switching upon the presence of the droplet is rationalized in terms of specific interactions of water with the exposed AAP moieties.

Lipid Droplets, Phospholipase A2, Arachidonic Acid, and Atherosclerosis

Lipid droplets, classically regarded as static storage organelles, are currently considered as dynamic structures involved in key processes of lipid metabolism, cellular homeostasis and signaling. Studies on the inflammatory state of atherosclerotic plaques suggest that circulating monocytes interact with products released by endothelial cells and may acquire a foamy phenotype before crossing the endothelial barrier and differentiating into macrophages. One such compound released in significant amounts into the bloodstream is arachidonic acid, the common precursor of eicosanoids, and a potent inducer of neutral lipid synthesis and lipid droplet formation in circulating monocytes. Members of the family of phospholipase A2, which hydrolyze the fatty acid present at the sn-2 position of phospholipids, have recently emerged as key controllers of lipid droplet homeostasis, regulating their formation and the availability of fatty acids for lipid mediator production. In this paper we discuss recent findings related to lipid droplet dynamics in immune cells and the ways these organelles are involved in regulating arachidonic acid availability and metabolism in the context of atherosclerosis.

Breaking the symmetry to suppress the Plateau–Rayleigh instability and optimize hydropower utilization

Droplet impact on solid surfaces is essential for natural and industrial processes. Particularly, controlling the instability after droplet impact, and avoiding the satellite drops generation, have aroused great interest for its significance in inkjet printing, pesticide spraying, and hydroelectric power collection. Herein, we found that breaking the symmetry of the droplet impact dynamics using patterned-wettability surfaces can suppress the Plateau–Rayleigh instability during the droplet rebounding and improve the energy collection efficiency. Systematic experimental investigation, together with mechanical modeling and numerical simulation, revealed that the asymmetric wettability patterns can regulate the internal liquid flow and reduce the vertical velocity gradient inside the droplet, thus suppressing the instability during droplet rebounding and eliminating the satellite drops. Accordingly, the droplet energy utilization was promoted, as demonstrated by the improved hydroelectric power generation efficiency by 36.5%. These findings deepen the understanding of the wettability-induced asymmetrical droplet dynamics during the liquid–solid interactions, and facilitate related applications such as hydroelectric power generation and materials transportation.

Equivalence of sessile droplet dynamics under periodic and steady electric fields

The electrohydrodynamics of a sessile droplet under the influence of periodic and steady electric fields in microgravity conditions is theoretically investigated using an inertial lubrication model. Previous studies have revealed that a freely suspended spherical droplet with unequal conductivity and permittivity ratios exhibits distinct dynamics under periodic and equivalent steady forcing in the root mean-square sense. However, it is unclear when (if at all) such distinct dynamics occur for periodic and equivalent steady forcing in the case of sessile droplets. The equivalence between periodic and steady forcing is shown to be governed by the interfacial charge buildup, which further depends on the competition between the charge relaxation and forcing timescales. A circulation-deformation map is introduced for the sessile droplet that acts as a guideline to achieve electric field-induced wetting or dewetting as the case may be. We also demonstrate that a droplet may be rendered either more or less wetting solely by tuning the forcing frequency.

193 Single Cell RNA-sequencing Reveals a Role of Lipid Metabolism in Muscle Satellite Cells

Single Cell RNA-sequencing (scRNA-seq) is a powerful technique to deconvolute gene expression of various subset of cells intermingled within a complex tissue, such as the skeletal muscle. We first used scRNA-seq to understand dynamics of cell populations and their gene expression during muscle regeneration in murine limb muscles. This leads to the identification of a subset of satellite cells (the resident stem cells of skeletal muscles) with immune gene signatures in regenerating muscles. Next, we used scRNA-seq to examine gene expression dynamics of satellite cells at various status: quiescence, activation, proliferation, differentiation and self-renewal. This analysis uncovers stage-dependent changes in expression of genes related to lipid metabolism. Further analyses lead to the discovery of previously unappreciated dynamics of lipid droplets in satellite cells; and demonstrate that the abundance of the lipid droplets in newly divided satellite daughter cells is linked to cell fate segregation into differentiation versus self-renewal. Perturbation of lipid droplet dynamics through blocking lipolysis disrupts cell fate homeostasis and impairs muscle regeneration. Finally, we show that lipid metabolism regulates the function of satellite cells through two mechanisms. On one hand, lipid metabolism functions as an energy source through fatty acid oxidation (FAO), and blockage of FAO reduces energy production that is critical for satellite cell function. On the other hand, lipid metabolism generates bioactive molecules that influence signaling transduction and gene expression. In this scenario, lipid metabolism and FAO regulate the intracellular levels of acetyl-coA and selective acetylation of PAX7, a pivotal transcriptional factor underlying function of satellite cells. These results together reveal for the first time a critical role of lipid metabolism and lipid droplet dynamics in muscle satellite cell fate determination and regenerative function; and underscore a potential role of dietary fatty acids in satellite cell-dependent muscle development, growth and regeneration.

Spatiotemporal evaporating droplet dynamics on fomites enhances long term bacterial pathogenesis

Naturally drying bacterial droplets on inanimate surfaces representing fomites are the most consequential mode for transmitting infection through oro-fecal route. We provide a multiscale holistic approach to understand flow dynamics induced bacterial pattern formation on fomites leading to pathogenesis. The most virulent gut pathogen, Salmonella Typhimurium (STM), typically found in contaminated food and water, is used as model system in the current study. Evaporation-induced flow in sessile droplets facilitates the transport of STM, forming spatio-temporally varying bacterial deposition patterns based on droplet medium’s nutrient scale. Mechanical and low moisture stress in the drying process reduced bacterial viability but interestingly induced hyper-proliferation of STM in macrophages, thereby augmenting virulence in fomites. In vivo studies of fomites in mice confirm that STM maintains enhanced virulence. This work demonstrates that stressed bacterial deposit morphologies formed over small timescale (minutes) on organic and inorganic surfaces, plays a significant role in enhancing fomite’s pathogenesis over hours and days.
