Managing endangered species in fragmented landscapes requires estimating dispersal rates between populations over contemporary timescales. Here we develop a new method for quantifying recent dispersal using genetic pedigree data for close and distant kin. Specifically, we describe an approach that infers missing shared ancestors between pairs of kin in habitat patches across a fragmented landscape. We then apply a stepping-stone model to assign unsampled individuals in the pedigree to probable locations based on minimizing the number of movements required to produce the observed locations in sampled kin pairs. Finally, we use all pairs of reconstructed parent-offspring sets to estimate dispersal rates between habitat patches under a Bayesian model. Our approach measures connectivity over the timescale represented by the small number of generations contained within the pedigree and so is appropriate for estimating the impacts of recent habitat changes due to human activity. We used our method to estimate recent movement between newly discovered populations of threatened Eastern Massasauga Rattlesnakes (Sistrurus catenatus) using data from 2996 RAD-based genetic loci. Our pedigree analyses found no evidence for contemporary connectivity between five genetic groups, but, as validation of our approach, showed high dispersal rates between sample sites within a single genetic cluster. We conclude that these five genetic clusters of Eastern Massasauga Rattlesnakes have small numbers of resident snakes and are demographically isolated conservation units. More broadly, our methodology can be widely applied to determine contemporary connectivity rates, independent of bias from shared genetic similarity due to ancestry that impacts other approaches.

In the face of ongoing habitat loss and fragmentation, maintaining an adequate level of landscape connectivity is needed to both encourage dispersal between habitat patches and to reduce the extinction risk of fragmented wildlife populations. In a developing region of southwestern Ontario, Canada, a declining population of Eastern Massasauga rattlesnakes (Sistrurus catenatus) persists in fragmented remnants of tallgrass prairie in an urban park system. The goal of this study was to identify potential connectivity pathways between habitat patches for this species by using a GIS least-cost permeability swath model, and to evaluate the outputs with snake road mortality data. Results identified seven pathways between five core habitat blocks, a subset of which were validated with aerial imagery and mortality data. Four high-ranking pathways intersected roads through or near road mortality hotspots. This research will guide conservation interventions aimed at recovering endangered reptiles in a globally rare ecosystem, and will inform the use of permeability swaths for the identification of locations most suitable for connectivity interventions in dynamic, urbanizing landscapes.

A 0.34 kg adult female Massasauga rattlesnake (Sistrurus catenatus) was presented for evaluation of a subcutaneous mass affecting the ventral scales. The mass was diagnosed as a sarcoma via punch biopsy with no evidence of metastasis on diagnostic imaging. Surgical margins of 1-2 cm were planned to achieve complete excision of the neoplasm. A technique for tissue expansion was employed due to concerns regarding the ability to close the surgical site without excess tension or dehiscence. Two 27 mm diameter×5 mm hygroscopic self-inflating tissue expanders were placed subcutaneously under the lateral scales adjacent to the mass. Maximum skin expansion occurred over a four-week period, and no direct negative effects were noted. Excision of the primary mass was performed routinely five weeks after implant placement. Primary closure of the defect was achieved with minimal tension by incorporating the expanded skin. While the surgery was successful with no evidence of metastasis, the snake died of sepsis two weeks postoperatively. This is the first report of the use of self-inflating hygroscopic tissue expanders to help close a surgical defect in a reptile.

At the northern limit of the Eastern Massasauga Rattlesnake’s (Sistrurus catenatus (Rafinesque, 1818)) range, individuals spend up to half the year overwintering. In hummock hibernacula found in peatlands, it is likely that subsurface temperature and water table position are contributing factors dictating habitat suitability. As a step towards assessing the vulnerability of hibernacula to anthropogenic changes, we combined subsurface temperature and water table dynamics to assess the likelihood that unflooded and unfrozen conditions were present in hummock hibernacula. Our results indicate that taller hummocks are more resilient to an advancing frost line and fluctuating water table by providing a larger area and duration of unfrozen and unflooded conditions, and a critical overwintering depth that is farther from the hummock surface. In two study sites along eastern Georgian Bay, an unflooded and unfrozen zone was present for over 90% of the overwintering period for hummocks taller than 25–27 cm. Our findings highlight the vulnerability of peatland hummocks to variability of winter weather where deep freezing and (or) water table rise may nonlinearly reduce resilience. This suggests that height is not the only component affecting the suitability of hummock hibernacula and that further research should examine the structure and spatial arrangement of hummocks within a peatland.
