Introduction: Bacterial isolates and control strains stocking is an integral part of clinical microbiology laboratories. This is an essential step in maintaining quality. Preserving the strains without altering the character is an essentiality. There are different stock culture preparations studied in past showing varied level of performance. Aim: To evaluate the performance in terms of longevity and phenotypic character preservation of Peptone Glycerol Broth (PGB) in comparison to Brucella Glycerol Broth (BGB) and Skim Milk (SKM). Materials and Methods: The present study was a prospective analytical study. Three quality control strains and seven clinical isolates with different types of resistance were stocked in triplicates with cryobead based peptone broth with 15% glycerol, Brucella Broth (BB) with 15% glycerol and 10% SKM and stored at -80°C. Isolates were revived in monthly pattern, quarterly pattern and once after 10 months to assess the variations in viability and loss of phenotypic properties arising out of repeated freeze thaw and contaminations. Viability was assessed by time taken to produce observable confluent growth on revival. Metabolic characters and antibiotic susceptibility testing were compared before and after stock revival at intervals. Results: The phenotypic characters like metabolic features and antibiotic susceptibility were preserved with all three preparations both with repeated freeze thaw and single freeze thaw at the end of 10 months. PGB and BGB had a 100% revival rate of stored isolates with a confluent growth at 24 hours in comparison to 93.56% with SKM. Conclusion: Cryobead preparation of peptone broth-15% glycerol can be used as an effective preparation for stock culture maintenance of non-fastidious bacteria and yeast.

AbstrakPenatalaksanaan utama pada masalah akne vulgaris adalah penggunaaan antibotik baik topikal maupun oral. Akan tetapi penggunaan antibiotik dinilai telah menimbulkan dugaan resistensi terhadap P. acnes sebagai agent penyebab akne sehingga mendorong berbagai pihak untuk mengembangkan preparat antiinflamasi yang dapat diberikan topical ataupun sistemik. Curcuma xanthorrhiza Roxb. memiliki senyawa utama xanthorrizol yang dinilai potensial untuk dikembangan sebagai antibakteri. Tujuan penelitian ini adalah untuk mengetahui kadar hambat minimum dan kadar bunuh minimum serta perubahan struktur dinding sel dari Curcuma xanthorrhiza Roxb. terhadap pertumbuhan Propionibacterium acnes. Desain penelitian yang di gunakan adalah eksperimen dengan sampel P. acnes berupa isolate stock culture (ATCC® 11827™) yang selanjutnya ditumbuhkan pada media MHA. Jumlah replikasi yang digunakan sebanyak 4 ulangan. Konsentrasi ekstrak Curcuma xanthorrhiza Roxb. masing-masing 6,25 µg/ml, 12,5 µg/ml, 25 µg/ml, 50 µg/ml dan 100 µg/ml. Pengukuran aktivitas antibakteri didasrkan pada KHM, KBM dan pengamatan struktur dinding sel bakteri melalui metode Microscop Electron Screening (MES). Pemberian ekstrak Curcuma xanthorrhiza Roxb. memiliki efek antibakteri terhadap bakteri P. acnes secara in vitro. Konsentrasi ekstrak 25 µg/ml merupakan kadar minimum yang mampu menghambat pertumbuhan P.acnes melalui dilusi cair, sedangkan konsentrasi minimal yang mampu membunuh P.acnes adalah 50 µg/ml. Bakteri P. acnes yang dipapar dengan ekstrak etanol Curcuma xanthorrhiza Roxb. mengalami perubahan morfologi berupa timbulnya dinding sel kasar kasar akibat penyusutan serta adanya dinding sel yang hancur sehingga sitoplasma keluar dan tampak seperti meleleh. Respon daya hambat pertumbuhan bakteri yang dihasilkan Curcuma xanthorrhiza Roxb. dipengaruhi oleh senyawa aktif yang terkandung didalamnya seperti minyak atsiri, alkaloid, flavonoid, tanin, kurkuminoid dan terpenoid. Kandungan xanthorrizol yang dimiliki mampu menghambat pertumbuhan P.acnes mampu merusak aktivitas enzim sel, selain itu kandungan Curcuminoid turut berperan dalam menghambat pertumbuhan bakteri dengan cara mendenaturasi dan merusak membran sel sehingga proses metabolisme sel terganggu  Kata Kunci: Antibakteri, Akne vulgaris, Curcuma xanthorriza Roxb., MES

We report the draft genome sequence of Porphyromonas gingivalis strain 381 Okayama (381OKJP). The strain, obtained from the Socransky collection, has been used for experimentation since 1987.

Sphingopyxis macrogoltabida strain 203, the type strain of the species, grew on polyethylene glycol (PEG) and has been deposited to the stock culture at the Biological Resource Center, National Institute of Technology and Evaluation (NITE), under the number NBRC 15033. Here, we report the complete genome sequence of strain NBRC 15033. Unfortunately, genes for PEG degradation were missing.

Prevotella intermedia , a Gram-negative black-pigmented anaerobic rod, is frequently isolated from not only periodontal pockets but also purulent infections. We report here the complete genome sequence of P. intermedia strain 17-2, which is a non-exopolysaccharide-producing variant obtained from exopolysaccharide (EPS)-producing P. intermedia strain 17 stock culture.
