Effect of plateau pika presence on the ecosystem services of alpine meadows

The activity of small mammalian herbivores influences grassland ecosystem services in arid and semi-arid regions. This study took plateau pika (Ochotona curzoniae) as example animal to investigate the effect of small mammalian herbivores on meadow ecosystem services in alpine regions. In this study, a home-range scale was used measure the forage availability, water conservation, carbon sequestration and soil nutrient maintenance services (total nitrogen, phosphorus, and potassium) of topsoil layer; and a quadrat scale was used to assess the biodiversity conservation service of alpine meadows. This study showed that plateau pika presence led to lower forage availability and water conservation services, and led to higher biodiversity conservation, carbon sequestration, soil nitrogen and phosphorus maintenance services of meadow ecosystem, whereas it had no impact on soil potassium maintenance service of meadow ecosystem in alpine regions. This study further found that the forage availability, biodiversity conservation, and soil nutrient maintenance services of meadow ecosystem in alpine regions firstly increased, and later decreased as the disturbance intensity of plateau pikas increased, whereas the water conservation service tended to decrease with the increasing disturbance intensity of plateau pikas. These results not only present a possible pattern of plateau pikas influencing the ecosystem services of meadow ecosystem in alpine regions, and consummate the small mammalian herbivores in relation to grassland ecosystem services.

Discovery and Evolution of a Divergent Coronavirus in the Plateau Pika From China That Extends the Host Range of Alphacoronaviruses

Although plateau pikas are the keystone species in the plateau ecosystem of the Qinghai Province of China, little is known about their role in the evolution and transmission of viral pathogens, especially coronaviruses. Here, we describe the characterization and evolution of a novel alphacoronavirus, termed plateau pika coronavirus (PPCoV) P83, which has a prevalence of 4.5% in plateau pika fecal samples. In addition to classical gene order, the complete viral genome contains a unique nonstructural protein (NS2), several variable transcription regulatory sequences and a highly divergent spike protein. Phylogenetic analysis indicates that the newly discovered PPCoV falls into the genus Alphacoronavirus and is most closely related to rodent alphacoronaviruses. The co-speciation analysis shows that the phylogenetic trees of the alphacoronaviruses and their hosts are not always matched, suggesting inter-species transmission is common in alphacoronaviruses. And, PPCoV origin was estimated by molecular clock based on membrane and RNA-dependent RNA polymerase encoding genes, respectively, which revealed an apparent discrepancy with that of co-speciation analysis. PPCoV was detected mainly in intestinal samples, indicating a potential enteric tropism for the virus. Overall, this study extends the host range of alphacoronaviruses to a new order (Lagomorpha), indicating that plateau pikas may be the natural reservoir of PPCoV and play an important and long-term role in alphacoronavirus evolution.

Sympatric Yaks and Plateau Pikas Promote Microbial Diversity and Similarity by the Mutual Utilization of Gut Microbiota

Interactions between species provide the basis for understanding coexisting mechanisms. The plateau pika (Ochotona curzoniae) and the yak (Bos grunniens) are considered competitors because they have shared habitats and consumed similar food on the Qinghai–Tibetan Plateau for more than 1 million years. Interestingly, the population density of plateau pikas increases with yak population expansion and subsequent overgrazing. To reveal the underlying mechanism, we sequenced the fecal microbial 16S rDNA from both sympatric and allopatric pikas and yaks. Our results indicated that sympatry increased both gut microbial diversity and similarity between pikas and yaks. The abundance of Firmicutes, Proteobacteria, Cyanobacteria, and Tenericutes decreased, while that of Verrucomicrobia increased in sympatric pikas. As for sympatric yaks, Firmicutes, Bacteroidetes, and Spirochaetes significantly increased, while Cyanobacteria, Euryarchaeota, and Verrucomicrobia significantly decreased. In sympatry, plateau pikas acquired 2692 OTUs from yaks, and yaks obtained 453 OTUs from pikas. The predominant horizontally transmitted bacteria were Firmicutes, Bacteroidetes, Verrucomicrobia, and Proteobacteria. These bacteria enhanced the enrichment of pathways related to prebiotics and immunity for pikas, such as heparin sulfate, heparin, chitin disaccharide, chondroitin-sulfate-ABC, and chondroitin-AC degradation pathways. In yaks, the horizontally transmitted bacteria enhanced pathways related to hepatoprotection, xenobiotic biodegradation, and detoxification. Our results suggest that horizontal transmission is a process of selection, and pikas and yaks tend to develop reciprocity through the horizontal transmission of gut microbiota.

Use of DNA metabarcoding of bird pellets in understanding raptor diet on the Qinghai-Tibetan Plateau of China

Background Diet analysis is essential to understanding the functional role of large bird species in food webs. Morphological analysis of regurgitated bird pellet contents is time intensive and may underestimate biodiversity. DNA metabarcoding has the ability to circumvent these issues, but has yet to be done. Methods We present a pilot study using DNA metabarcoding of MT-RNR1 and MT-CO1 markers to determine the species of origin and prey of 45 pellets collected in Qinghai and Gansu Provinces, China. Results We detected four raptor species [Eurasian Eagle Owl (Bubo bubo), Saker Falcon (Falco cherrug), Steppe Eagle (Aquila nipalensis), and Upland Buzzard (Buteo hemilasius)] and 11 unique prey species across 10 families and 4 classes. Mammals were the greatest detected prey class with Plateau Pika (Ochotona curzoniae) being the most frequent. Observed Shannon’s and Simpson’s diversity for Upland Buzzard were 1.089 and 0.479, respectively, while expected values were 1.312 ± 0.266 and 0.485 ± 0.086. For Eurasian Eagle Owl, observed values were 1.202 and 0.565, while expected values were 1.502 ± 0.340 and 0.580 ± 0.114. Interspecific dietary niche partitioning between the two species was not detected. Conclusions Our results demonstrate successful use of DNA metabarcoding for understanding diet via a novel noninvasive sample type to identify common and uncommon species. More work is needed to understand how raptor diets vary locally, and the mechanisms that enable exploitation of similar dietary resources. This approach has wide ranging applicability to other birds of prey, and demonstrates the power of using DNA metabarcoding to study species noninvasively.

Surviving winter on the Qinghai-Tibetan Plateau: Pikas suppress energy demands and exploit yak feces to survive winter

The Qinghai-Tibetan Plateau, with low precipitation, low oxygen partial pressure, and temperatures routinely dropping below −30 °C in winter, presents several physiological challenges to its fauna. Yet it is home to many endemic mammalian species, including the plateau pika (Ochotona curzoniae). How these small animals that are incapable of hibernation survive the winter is an enigma. Measurements of daily energy expenditure (DEE) using the doubly labeled water method show that pikas suppress their DEE during winter. At the same body weight, pikas in winter expend 29.7% less than in summer, despite ambient temperatures being approximately 25 °C lower. Combined with resting metabolic rates (RMRs), this gives them an exceptionally low metabolic scope in winter (DEE/RMRt = 1.60 ± 0.30; RMRt is resting metabolic rate at thermoneutrality). Using implanted body temperature loggers and filming in the wild, we show that this is achieved by reducing body temperature and physical activity. Thyroid hormone (T3 and T4) measurements indicate this metabolic suppression is probably mediated via the thyroid axis. Winter activity was lower at sites where domestic yak (Bos grunniens) densities were higher. Pikas supplement their food intake at these sites by eating yak feces, demonstrated by direct observation, identification of yak DNA in pika stomach contents, and greater convergence in the yak/pika microbiotas in winter. This interspecific coprophagy allows pikas to thrive where yak are abundant and partially explains why pika densities are higher where domestic yak, their supposed direct competitors for food, are more abundant.

Improved Estimation of Aboveground Biomass of Disturbed Grassland through Including Bare Ground and Grazing Intensity

Accurate approaches to aboveground biomass (AGB) estimation are required to support appraisal of the effectiveness of land use measures, which seek to protect grazing-adapted grasslands atop the Qinghai-Tibet Plateau (QTP). This methodological study assesses the effectiveness of one commonly used visible band vegetation index, Red Green Blue Vegetation Index (RGBVI), obtained from unmanned aerial vehicle (UAV), in estimating AGB timely and accurately at the local scale, seeking to improve the estimation accuracy by taking into account in situ collected information on disturbed grassland. Particular emphasis is placed upon the mapping and quantification of areas disturbed by grazing (simulated via mowing) and plateau pika (Ochotona curzoniae) that have led to the emergence of bare ground. The initial model involving only RGBVI performed poorly in AGB estimation by underestimating high AGB by around 10% and overestimating low AGB by about 10%. The estimation model was modified by the mowing intensity ratio and bare ground metrics. The former almost doubled the estimation accuracy from R2 = 0.44 to 0.81. However, this modification caused the bare ground AGB to be overestimated by about 38 and 19 g m−2 for 2018 and 2019, respectively. Although further modification of the model by bare ground metrics improved the accuracy slightly to 0.88, it markedly reduced the overestimation of low AGB values. It is recommended that grazing intensity be incorporated into the micro-scale estimation of AGB, together with the bare ground modification metrics, especially for severely disturbed meadows with a sizable portion of bare ground.
