Effects of Salvianolic Acid B/ginsenoside Rg1 Combination Against Chronic Pulmonary Embolism Induced by Polystyrene Microspheres

Pulmonary embolism (PE) is the most life-threatening complication of venous thromboembolism, but few effective treatments have been discovered to attenuate chronic PE currently. In this study, we investigated the protective effects of salvianolic acid B (SalB) and ginsenoside Rg1 (Rg1) combination (SalB/Rg1) on chronic PE and explored the potential mechanisms. The PE model was induced by 45 μm polystyrene microspheres and 20 mg/kg of SalB/Rg1 was administered to PE rats intraperitoneally. A histopathological analysis of the lungs and heart was performed through hematoxylin and eosin staining and immunohistochemical analysis. The pulmonary index and right ventricular cardiomyocyte cross-sectional area were evaluated. SalB/Rg1 markedly downregulated pulmonary index, attenuated pulmonary interstitial changes, suppressed neutrophil infiltration, prevented collagen deposition, and inhibited MMP-9 activities in the lung. We also found that SalB/Rg1 improved right ventricular hypertrophy accompanied by reducing the cardiomyocyte cross-sectional area. These data suggest that SalB/Rg1 played a protective role against microsphere-induced PE and holds a high potential for the treatment of PE in the future.

Method to determine particle release during long-term loading for assessment of coating durability of cardiovascular stents

Many catheters and vascular implants are coated to increase biocompatibility or to reduce friction during catheter based implantation. Several regulations require assessment of coating durability over the implant’s life time. An in vitro method for stent testing is presented to measure released particulate matter at defined inspection intervals. The method was validated using polystyrene microspheres with a size of 10, 25 and 50 μm to check for particle recovery (n=6). Two cleaning steps followed. Particle counting was performed by light obscuration method. The recovery rate was 103±5% (10μm), 94±4% (25 μm) and 78±12% (50 μm), respectively, meeting the requirements of FDA guidance documents (i.e. FDA 1545). Less than 3% of the particles were found in the cleaning solutions. The method using a fixed volume during stent loading can be adapted to all durability testers where tubes are used to fix the stents (radial pulsatile, bending, axial compression, torsion).

Serum Adsorption Study to Validate the Specificity of a Rapid Test to Detect Strongyloides stercoralis Infection

A lateral flow rapid test for strongyloidiasis will greatly facilitate the control and elimination of the disease. Previously SsRapid™ prototype rapid test showed high diagnostic specificity to detect Strongyloides infection, determined using non-Strongyloides sera negative by IgG-ELISAs. Since high specificity is crucial before a test is used for public health control activities, further validation of its specificity is needed. Also, it needs to be ascertained whether non-Strongyloides sera positive by IgG-ELISAs and SsRapid are truly positive for Strongyloides or are cases of cross-reactivity. We performed 84 rapid tests (two types of dipsticks and cassettes) using 34 serum samples. They were divided into four groups based on Strongyloides infection and coinfection with other parasites and the availability of recombinant proteins and rapid tests for the latter. Sera was adsorbed using polystyrene microspheres beads separately coated with four recombinant parasite proteins. The small sample size is a limitation of this study; however, the overall results showed that the sera adsorption procedure was successful, and the SsRapid test is specific.

Preparation, Structure, and Properties of Polystyrene-Microsphere-Reinforced PEG-Based Hydrogels

To improve the mechanical strength and practicability of hydrogels, polystyrene microspheres with core–shell structure were prepared by the soap-free emulsion polymerization, polyethylene glycol hydrogels with polystyrene microspheres by the in-situ polymerization. The structure, morphology, roughness, swelling property, surface energy, and mechanical properties of the microspheres and hydrogels were investigated by Fourier transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, confocal laser microscopy, swelling test, contact angle measurement, and compression test. The results showed that they have certain swelling capacity and excellent mechanical properties, and can change from hydrophobic to hydrophilic surface. The reason is that the hydrophilic chain segment can migrate, enrich, and form a hydration layer on the surface after soaking for a certain time. Introducing proper content of polystyrene microspheres into the hydrogel, the compressive strength and swelling degree improved obviously. Increasing the content of polystyrene microspheres, the surface energy of the hydrogels decreased gradually.
