A progeny of 130 F1 individuals of an interspecific cross between Populus tremula L. and Populus tremuloides Michx. was used for genetic mapping of linkage group XIX with SSR markers based on the Populus trichocarpa Torr. & A. Gray genome sequence. Several fully sex-linked SSR markers were identified and mapped to a central position on the male P. tremuloides map of linkage group XIX. For the SSR markers tested here, the position on the assembled P. trichocarpa genome sequence is known, allowing sex-linked markers to be assigned to the central region of scaffold/chromosome 19 of P. trichocarpa. The sex linkage of the SSR markers was validated in other P. tremula × P. tremuloides crosses and also tested in Populus alba L. and Populus nigra L.

Chickpea (Cicer arietinum L.) is an economically important food legume grown in arid and semi-arid regions of the world. Chickpea is cultivated mainly in the rainfed, residual moisture, and restricted irrigation condition. The crop is always prone to drought stress which is resulting in flower drop, unfilled pods, and is a major yield reducer in many parts of the world. The present study elucidates the association between candidate gene and morpho-physiological traits for the screening of drought tolerance in chickpea. Abiotic stress-responsive gene Dehydrin (DHN) was identified in some of the chickpea genotypes based on the sequence similarity approach to play a major role in drought tolerance. Analysis of variance revealed a significant effect of drought on relative water content, membrane stability index, plant height, and yield traits. The genotypes Pusa1103, Pusa362, and ICC4958 were found most promising genotypes for drought tolerance as they maintained the higher value of osmotic regulations and yield characters. The results were further supported by a sequence similarity approach for the dehydrin gene when analyzed for the presence of single nucleotide polymorphisms (SNPs) and indels. Homozygous indels and single nucleotide polymorphisms were found after the sequencing in some of the selected genotypes.

The Populus L. genus includes tree species that are botanically grouped into several sections. This species successfully hybridizes both in the same section and among other sections. Poplar hybridization widely occurs in nature and in variety breeding. Therefore, the development of poplar species’ specific molecular markers is very important. The effective markers for trees of the Aigeiros Duby section have recently been developed using the polymorphism of 5S rDNA non-transcribed spacers (NTSs). In this article, 5S rDNA NTS-based markers were designed for several species of the Leuce Duby section. The alb9 marker amplifies one fragment with the DNA matrix of P. alba and P. × canescens (natural hybrid P. alba × P. tremula). The alb2 marker works the same way, except for the case with Populus bolleana. In this case, the amplification of three fragments was observed. The tremu1 marker amplification was detected with the DNA matrix of P. tremula and P. × canescens. Thus, the developed markers may be applied as a useful tool for P. alba, P. tremula, P. × canescens, and P. bolleana identification in various areas of plant science such as botany, dendrology, genetics of populations, variety breeding, etc.

Dehydrins are major plant proteins that increase with cold or drought. Using a previously identified 2.0-Kb blueberry dehydrin cDNA, additional dehydrin clones were isolated and characterized. Experiments were conducted to determine effect of drought and cold treatment on dehydrin gene expression in several blueberry cultivars. Besides previously characterized 65, 60, and 14 kDa dehydrins in floral buds additional less abundant dehydrins were observed in stem and root tissues. The same major dehydrins were induced by both drought and cold stress but at different levels. Dehydrin expression was positively correlated with cold and drought tolerance. In addition, photoperiod was observed to affect dehydrin expression. Dehydrins were induced to higher level in cold treated plants kept in total dark than in plants kept at a short photoperiod of 10 hours of light/14 hours dark.
