Abstract Background Small RNAs (sRNAs) including microRNAs (miRNAs) and small interfering RNAs (siRNAs) serve as core players in gene silencing at transcriptional and post-transcriptional levels in plants, but their subcellular localization has not yet been well studied, thus limiting our mechanistic understanding of sRNA action. Results We investigate the cytoplasmic partitioning of sRNAs and their targets globally in maize (Zea mays, inbred line “B73”) and rice (Oryza sativa, cv. “Nipponbare”) by high-throughput sequencing of polysome-associated sRNAs and 3′ cleavage fragments, and find that both miRNAs and a subset of 21-nucleotide (nt)/22-nt siRNAs are enriched on membrane-bound polysomes (MBPs) relative to total polysomes (TPs) across different tissues. Most of the siRNAs are generated from transposable elements (TEs), and retrotransposons positively contributed to MBP overaccumulation of 22-nt TE-derived siRNAs (TE-siRNAs) as opposed to DNA transposons. Widespread occurrence of miRNA-mediated target cleavage is observed on MBPs, and a large proportion of these cleavage events are MBP-unique. Reproductive 21PHAS (21-nt phasiRNA-generating) and 24PHAS (24-nt phasiRNA-generating) precursors, which were commonly considered as noncoding RNAs, are bound by polysomes, and high-frequency cleavage of 21PHAS precursors by miR2118 and 24PHAS precursors by miR2275 is further detected on MBPs. Reproductive 21-nt phasiRNAs are enriched on MBPs as opposed to TPs, whereas 24-nt phasiRNAs are nearly completely devoid of polysome occupancy. Conclusions MBP overaccumulation is a conserved pattern for cytoplasmic partitioning of sRNAs, and endoplasmic reticulum (ER)-bound ribosomes function as an independent regulatory layer for miRNA-induced gene silencing and reproductive phasiRNA biosynthesis in maize and rice.

The world faces today a pandemic of unquestionable importance, caused by an infection with a new enveloped RNA virus that belongs to the Coronaviridae family. The new coronavirus (SARS-CoV 2) uses a glycoprotein present on its surface to bind to and infect host cells that express the angiotensin converting enzyme II (ACE-2). Although different tissues may be targeted by the virus, respiratory complications remain as the main cause of death. It has been demonstrated that Renin-Angiotensin-Aldosterone System (RAAS) inhibitors increase ACE-2 expression in animal models, raising the concern that patients under treatment with these drugs could become more susceptible to COVID-19 complications. Here, we discuss the impact of RAAS inhibitors on COVID-19 outcomes and show that no evidence so far supports that the use of these drugs could pose a risk to SARS-CoV 2-infected patients. In fact, clinical data suggest that RAAS inhibitors may even act in a protective way against COVID-19 complications and should not be discontinued.,

Enormous complications associated with diabetes contribute to the therapeutic challenge confronting most of the world, including developing countries. This study was carried out to investigate diabetes mellitus on esterases and antioxidant enzymes in different tissues compartments of rats. Animals were divided into two groups of 10 animals each. The experimental group was confirmed diabetic by a single dose of streptozotocin injection (STZ, freshly dissolved in citrate buffer, pH 4.5, 50 mg/kg) intraperitoneally. In contrast, the control group was injected with citrate buffer only. Blood glucose and weight of the animals were monitored for 7 days. Blood, liver and brain were removed, and biochemical parameters determined spectrophotometrically. Diabetes produced various degrees of alterations in antioxidant defence mechanism and esterases activities that are compartment specific. Acetylcholinesterase (AChE) activity was inhibited to various extents. While AChE was inhibited to the tune of 39% in plasma, 33% in the brain and 30% in the liver, activation of the activity was observed in the red blood cell (RBC). The same trend of significant (p < 0.001) inhibition was observed with arylesterase in the plasma, brain and liver, and activation in the RBC. Diabetes induced significant (p<0.001) inhibition in catalase, Glutathione-S-transferase, and Superoxide dismutase in the brain and liver, respectively, compared to control more than the other compartments. However, activation was also observed in the RBC of these enzymes except for catalase and nitric oxide. In conclusion, distinct compartments effects of diabetes observed in this study could suggest a new approach for effective and safer therapeutics.

The viral disease classical swine fever (CSF), caused by aPestivirus, is one of the major causes of economic losses for pig farming. The aim of this work was to validate a RT-qPCR using Taqman for detection of CSF in swine tissues. The parameters for the validation followed the specifications of the Manual of Diagnostic Tests and Vaccines for Terrestrial Animals of the World Organization for Animal Health (OIE) and the guide ABNT NBR ISO/IEC 17025:2005. The analysis of the 5′NTR region of CSF virus was performed in 145 samples from 29 infected pigs and in 240 samples from 80 pigs originated in the Brazilian CSF-free zone. The tissues tested were spleen, kidney, blood, tonsils, and lymph nodes. Sequencing of the positive samples for 5′NTR region was performed to evaluate the specificity of the RT-qPCR. Tests performed for the RT-qPCR validation demonstrated that the PCR assay was efficient in detecting RNA from CSF virus in all materials from different tissues of infected animals. Furthermore, RNA from CSF virus was not detected in samples of swine originated from the Brazilian CSF-free zone. Hence, it is concluded that RT-qPCR can be used as a complementary diagnostic for CSF.

MicroRNAs (miRNA) are endogenous tiny RNAs (about 22 nucleotides in length) that can play important regulatory roles in plants and animals by targeting mRNAs for cleavage or involved in translational suppression. Based on the sequence conservation of many miRNA genes in different plant genomes, it is possible to identify miRNAs in citrus. Identification of miRNA is the prerequisite for understanding the miRNA function in citrus. Citrus is an important fruit crop in the world and the publicly available citrus EST databases are increasing. Thirty known miRNAs from Arabidopsis were used to search the citrus EST databases for miRNA precursors. Nine possible citrus miRNA sequences were predicted to have fold-back structures. The Northern results indicated most of the 26 Arabidopsis miRNAs are expressed ubiquitously in the leaf, young shoot, flower, and root tissues of Nules Clementine mandarin (Citrus clementina Hort. Ex Tan.) and Trifoliate orange (Poncirus trifoliata [L.] Raf.). Some miRNAs accumulated preferentially in different tissues.

This is particularly striking in the study of religion and psychosis, where we and others have shown that cultural context determines both the structure and content of hallucination-like events. The cultural shaping of hallucinations may provide a rich case-study for linking cultural learning with emerging prediction-based models of perception.

I suggest calling these memory-like representations as what they are without beating around the bush. I refer to them as event memories and explain how they are different from episodic memory and how they can guide action in animal cognition.

AbstractFe XIV 530.3 nm coronal emission line observations have been used for the estimation of the green solar corona rotation. A homogeneous data set, created from measurements of the world-wide coronagraphic network, has been examined with a help of correlation analysis to reveal the averaged synodic rotation period as a function of latitude and time over the epoch from 1947 to 1991.The values of the synodic rotation period obtained for this epoch for the whole range of latitudes and a latitude band ±30° are 27.52±0.12 days and 26.95±0.21 days, resp. A differential rotation of green solar corona, with local period maxima around ±60° and minimum of the rotation period at the equator, was confirmed. No clear cyclic variation of the rotation has been found for examinated epoch but some monotonic trends for some time intervals are presented.A detailed investigation of the original data and their correlation functions has shown that an existence of sufficiently reliable tracers is not evident for the whole set of examinated data. This should be taken into account in future more precise estimations of the green corona rotation period.
