The mitotic apparatus is a structure of obvious biological and medical interest, but it has proved to be a difficult cellular machine to understand. The chemical composition of the spindle is only slightly elucidated, largely because of the difficulties in preparing useful isolates of the structure. Chemical studies of the mitotic spindle have been reviewed elsewhere (Mcintosh, 1977), and will not be discussed further here. One would think that structural studies on the mitotic apparatus (MA) in situ would be straightforward, but even with this approach there is some disagreement in the results obtained with various methods and by different investigators. In this paper I will review briefly the approaches which have been used in structural studies of the MA, pointing out the strengths and problems of each approach. I will summarize the principal findings of the different methods, and identify what seem to be fruitful avenues for further work.

Titan’s haze is strongly suspected to be an HCN-derived polymer, but despite the first in situ measurements by the ESA-Huygens space probe, its chemical composition and formation process remain largely unknown. To investigate this question, we simulated the atmospheric haze formation process, experimentally. We synthesized analogues of Titan’s haze, named Titan tholins, in an irradiated N2–CH4 gas mixture, mimicking Titan’s upper atmosphere chemistry. HCN was monitored in situ in the gas phase simultaneously with the formation and evolution of the haze particles. We show that HCN is produced as long as the particles are absent, and is then progressively consumed when the particles appear and grow. This work highlights HCN as an effective precursor of Titan’s haze and confirms the HCN-derived polymer nature of the haze.

A study was carried out to evaluate the effect of single cell protein (SCP) supplement as a protein source on nutrient intake, digestibility, nitrogen balance and in situ digestion kinetics in four Nili Ravi buffalo bulls. Four iso-caloric and iso-nitrogenous concentrates containing 3, 6, 9 and 12% of Saccharomyces cerevisiae-fermented citrus pulp were formulated. All animals were fed a ration with a concentrate/forage ratio of 50:50. Diets were provided ad libitum twice a day as a total mixed ration in a 4 × 4 Latin Square Design. Each experimental period lasted 3 weeks while the overall study 12 weeks. The first 2 weeks of each experimental period were used as adaptation period while the third week as collection period. Chemical composition of fermented citrus pulp appeared as an excellent source of protein. No significant difference was observed on dry matter intake, digestibility of nutrients and SCP among all the treatments. Moreover, no significant effect was observed on ruminal pH and ammonia nitrogen at different times. Rate of disappearance and lag time of in situ dry matter digestion kinetics remained nonsignificant regardless of SCP percentage. Based on results of similar nutrients intake, nutrient digestibility, and ruminal parameters it is concluded that SCP could be used in the concentrate diet of ruminant up to 12%. Furthermore, the SCP has the potential of an alternative protein source in animal diet formulation.

In situ investigation results of aerosol optical properties (absorption and scattering) and chemical composition at an urban background site in Lithuania (Vilnius) are presented. Investigation was performed in May-June 2017 using an aerosol chemical speciation monitor (ACSM), a 7-wavelength Aethalometer and a 3-wavelength integrating Nephelometer. A positive matrix factorisation (PMF) was used for the organic aerosol mass spectra analysis to characterise the sources of ambient organic aerosol (OA). Five OA factors were identified: hydrocarbon-like OA (HOA), biomass-burning OA (BBOA), more and less oxygenated OA (LVOOA and SVOOA, respectively), and local hydrocarbon-like OA (LOA). The average absorption (at 470 nm) and scattering (at 450 nm) coefficients during the entire measurement campaign were 16.59 Mm−1 (standard deviation (SD) = 17.23 Mm−1) and 29.83 Mm−1 (SD = 20.45 Mm−1), respectively. Furthermore, the absorption and scattering Angström exponents (AAE and SAE, respectively) and single-scattering albedo (SSA) were calculated. The average AAE value at 470/660 nm was 0.97 (SD = 0.16) indicating traffic-related black carbon (BCtr) dominance. The average value of SAE (at 450/700 nm) was 1.93 (SD = 0.32) and could be determined by the submicron particle (PM1) dominance versus the supermicron ones (PM > 1 µm). The average value of SSA was 0.62 (SD = 0.13). Several aerosol types showed specific segregation in the SAE versus SSA plot, which underlines different optical properties due to various chemical compositions.

Near infrared (NIR) spectral analysis with a NIRSystems 6500 monochromator was applied to evaluate accuracy of predictive models for forage quality in clone breeding processes of the original Bulgarian varieties over different cuts and years. The varieties were the perennial grasses: Dactylis glomerata L., Festuca arundinacea Schreb. and Bromus inermis Leyss. Global calibrations for the 418 perennial grass samples and specific calibrations for each single grass species and internal cross-validations were performed by the PLS regression method. The effect of different spectral data pre-treatments was investigated on the residual standard errors of the NIR predictive models. Among 60 calibration equations, the model with the lowest SECV value was retained for each parameter in each database. No particular data pre-treatment was really better than the other ones. Generally, the best results of the global calibrations were obtained with SNVD and MSC. For the specific calibrations, SNVD and WMSC were the best treatments. In both cases, the first or second derivatives were needed after the first pre-treatment. Chemical composition and in vitro enzymatic digestibility of clones were predicted with accuracy similar to that of classical laboratory methods. For the cell wall component contents, the standard errors of cross-validation SECV(%DM) ranged from 0.49 for ADL ( Festuca) to 2.02 for NDF ( Dactylis). The digestibilities of dry and organic matter, IVDMD and IVOMD, were estimated with SECVs from 2.6 to 3.0%, the relative intake, from 0.06 to 0.09 rel% body weight and the relative feeding value, from 4.39 to 5.64 rel%. The global calibration models offer an acceptable accuracy for the estimation of the cell wall nutrient contents, the digestibility and the nutritive value. The standard errors of prediction of specific single species calibrations with smaller numbers of terms were lower in 60% of the cases than those obtained from the best global calibrations with higher numbers of terms. On average, SECVs from specific calibrations are better than those from global calibrations, but the differences are quite small, and for the prediction of totally new samples (new crops, another year), the global calibrations will detect less outlier samples. Even with very high variability between cuts and years, NIR spectroscopy is able via ANOVA GL Models to sort clones on their feeding value and to provide relevant information for the breeding programmes.
