The anatomical changes occurring in the strawberry fruit in the course of its growth were investigated. Flowers or fruits were collected from the apexes of the inflorescence of plants of the cultivar 'Talisman' cultivated in the glasshouse under long day. A method was developed for embedding the receptacle in a mixture of paraffin, rubber and wax, because the classical paraffin method did not give good results. Three different vascular bundle systems were distinguished in the strawberry, supplying the particular parts of the flower. Most intensive growth of the mean number of cells occurs in the cortex. The number of pith cells, beginning with the 5th day after pollination does not practically increase. Neither does between the 5th and 10th day after pollination, the number of cortical cells increase in the apical part of the fruit.

Mutations in the Arabidopsis TETRATRICOPEPTIDE THIOREDOXIN-LIKE 1 (TTL1) gene cause reduced tolerance to osmotic stress evidenced by an arrest in root growth and root swelling, which makes it an interesting model to explore how root growth is controlled under stress conditions. We found that osmotic stress reduced the growth rate of the primary root by inhibiting the cell elongation in the elongation zone followed by a reduction in the number of cortical cells in the proximal meristem. We then studied the stiffness of epidermal cell walls in the root elongation zone of ttl1 mutants under osmotic stress using atomic force microscopy. In plants grown in control conditions, the mean apparent elastic modulus was 448% higher for live Col-0 cell walls than for ttl1 (88.1 ± 2.8 vs. 16.08 ± 6.9 kPa). Seven days of osmotic stress caused an increase in the stiffness in the cell wall of the cells from the elongation zone of 87% and 84% for Col-0 and ttl1, respectively. These findings suggest that TTL1 may play a role controlling cell expansion orientation during root growth, necessary for osmotic stress adaptation.

First-order laterals of Pinus banksiana seedlings were inoculated with variant strains of ectomycorrhizal Laccaria bicolor in an aseptic culture system. Macroscopic observations of 10 fungal strains indicated that 6 are mycorrhizal and 4 are apparently nonmycorrhizal. Furthermore, light microscopic examinations revealed significant intraspecific variation in mycorrhizal structures. The mean mantle thickness, mean mantle density, and mean Hartig net penetration of the six mycorrhizal strains ranged from 2.5 to 13.4 hyphae, 278 to 411 hyphae/mm and 2 to 2.8 root cell layers, respectively. Three of these strains formed fewer macroscopically observable mycorrhizae and developed significantly thinner mantles but their Hartig nets usually separated cortical cells more extensively. Three of the four apparently nonmycorrhizal strains showed infrequent and poor Hartig net development (mean penetration of 0.3 to 0.8 root cell layer), poor surface colonization, and no mantle development. These three strains were better able to colonize long roots. Only one strain could be considered truly nonmycorrhizal because it only colonized root surfaces poorly and never showed mantle or Hartig net formation. The observed intraspecific variability raises questions concerning the determinants of mycorrhiza development and structure.

Purpose. To evaluate the effect of phacoemulsification and intraocular lens (IOLs) implantation in eyes with medically uncontrolled primary angle-closure glaucoma (PACG) previously treated with trabeculectomy and to quantify the anatomical changes in the anterior chamber angle by ultrasound biomicroscopy (UBM). Methods. Forty-four eyes of 37 consecutive patients with medically uncontrolled PACG coexisting cataracts with a surgical history of trabeculectomy were included in this study. Each patient underwent phacoemulsification and IOL implantation. Indentation gonioscopy and UBM were performed preoperatively and then again 3 months after surgery. The main outcome measures were best-corrected visual acuity (BCVA), intraocular pressure (IOP), number of antiglaucoma medications and anatomical changes in the anterior chamber angle. Results. The mean logarithm of the minimum angle of resolution BCVA significantly improved from 0.52 ± 0.30 preoperatively to 0.26 ± 0.23 postoperatively (p<0.001). The mean IOP significantly decreased from 24.33 ± 9.65 mmHg preoperatively to 18.04 ± 7.86 mmHg postoperatively (p<0.05). 001). The median number of antiglaucoma medications decreased from 2 preoperatively to 1 postoperatively (p<0.001). There was no significant difference in the extent of peripheral anterior synechia after the surgery (p>0.05). Some parameters, including anterior central chamber depth, angle opening distance at 500 μm, trabecular-iris angle, and scleral ciliary process angle, were significantly higher after than before surgery (p<0.001). However, the crystalline lens rise was significantly smaller following the surgery (p<0.001). Conclusions. Phacoemulsification and IOL implantation reduced the IOP and improved vision in eyes with medically uncontrolled filtered PACG. The mechanism underlying the outcomes observed following surgery might be related to the anterior chamber deepening, widened drainage angle, and improved aqueous fluid flow to the trabecular meshwork.

Two multitype models are considered: a Galton-Watson process in discrete time, and its analogous birth-death process in continuous time. The mean number of cells with parity 0, 1, 2, … is obtained in both cases; some simple results are also derived for the second moments of the two processes.

Dog granulocytes were labeled in vitro with radioactive diisopropylfluorophosphate (DFP32) and then returned to the circulation of the donor. Granulocytes were separated from whole blood by utilizing hexadimethrine bromide as the sedimenting agent and saponin as a lysing agent. The labeled granulocytes disappeared from the circulation in an exponential fashion with a mean (±1 sd) half-time disappearance of 5.6 ± 0.95 hr. The size of the total blood granulocyte ( TBGP), circulating granulocyte ( CGP), and marginal granulocyte ( MGP) pools, and the granulocyte turnover rate ( GTR) were measured in 31 normal, unanesthetized dogs. The mean values ± 1 sd, expressed as number of cells x107/kg body wt., were as follows: TBGP, 102 ± 34.8; CGP, 54 ± 20.7; MGP, 48 ± 23.4; and GTR, 305 ± 111.5 cells/kg day. The values observed in anesthetized and in unanesthetized, splenectomized dogs were not significantly different from the above values.

Delivery of cells into tubular tissue constructs with large diameters poses significant spatial and temporal challenges. This study describes preliminary findings for a novel process for rapid and uniform seeding of cells onto the luminal surface of large tubular constructs. Fibroblasts, tagged with superparamagnetic iron oxide nanoparticles (SPION), were directed onto the luminal surface of tubular constructs by a magnetic field generated by a k4-type Halbach cylinder device. The spatial distribution of attached cells, as measured by the mean number of cells, was compared with a conventional, dynamic, rotational cell-delivery technique. Cell loading onto the constructs was measured by microscopy and magnetic resonance imaging. The different seeding techniques employed had a significant effect on the spatial distribution of the cells ( p < 0.0001). The number of attached cells at defined positions within the same construct was significantly different for the dynamic rotation technique ( p < 0.05). In contrast, no significant differences in the number of cells attached to the luminal surface were found between the defined positions on the construct loaded with the Halbach cylinder. The technique described overcomes limitations associated with existing cell-delivery techniques and is amenable to a variety of tubular organs where rapid loading and uniform distribution of cells for therapeutic applications are required.

Staminate and hermaphroditic flowers show an hypogynous disc which is composed of two separate parts, an apical and a basal part. These histologic zones should be considered as nectaries. They differ by their position, their shape, and the content and state of differentiation of their cells. Basal nectaries are not vascularized but phloem bundles reach the tip of the disc. Initiation of nectariferous tissue occurs after that of the floral parts. The basal nectaries appear first but the number of cells still increases during the formation of the apical part of the disc. Both basal and apical nectaries have a superficial origin. Numerous anomocytic stomata are present especially on the surface of the apical nectaries; they are rounded and keep their stomatal pores open. On basal nectaries, however, they occur much less frequently.

The aluminium distribution in transverse sections of fractured and freeze-dried roots of cabbage, lettuce and kikuyu grass was studied by energy-dispersive X-ray analysis. The mean ratios of the integrated counts corresponding to Kα emission spectra for aluminium, phosphorus and silicon were recorded between aluminium and control treatments over three experiments. Peak-to-back- ground ratios (PA/B) were calculated and the aluminium ratios corrected for variations in the corresponding total silicon ratios. Aluminium was found to be uniformly distributed along roots of the three species. The highest peaks (PA) and peak-to-background ratios, suggesting higher concentrations, were recorded in the epidermis followed by the cortex. Aluminium was recorded in the stele of all species and in the protoplasm of cortical cells, with smaller amounts in the protoplasm of xylem parenchyma cells for lettuce and kikuyu grass. The distribution of aluminium supports the hypothesis that its entry to the stele can be achieved by transport both into meristematic cells and the symplasm via the cortex and hence bypassing the barrier at the endodermis. The latter evidence was supported by the presence of aluminium in the radial wall (and cytoplasm) of the endodermis for each species. There was a poor correlation between the distribution of aluminium and phosphorus.

We performed a pathologic study with further using an immunohistochemical technique (using anti-p63 and anti-CK5) on tissues obtained by open lung biopsy from 18 patients with previous exposure to sulphur mustard (SM) as case group and 8 unexposed patients (control group). The most frequent pathologic diagnosis was constrictive bronchiolitis (44.4%), followed by respiratory (22.2%) and chronic cellular bronchiolitis (16.7%) in the case group, and hypersensitivity bronchiolitis (50%) in the control group. The pathologic diagnoses were significantly different in the case and control groups (P=0.042). In slides stained by anti-p63 and anti-CK5, the percent of stained cells and the mean number of epithelial cells were lower in the case group in comparison to the control group. This difference was significant for the mean number of cells stained by anti-CK5 (P=0.042). Furthermore, there was a significant correlation between pathologic diagnosis and total number of cells and mean number of cells stained with anti-p63 and anti-CK5 (P  value = 0.002, <0.001, 0.044). These results suggest that constrictive bronchiolitis may be the major pathologic consequence of exposure to SM. Moreover, decrease of p63 in respiratory tissues affected by SM may suggest the lack of regenerative capacity in these patients.

The effect of oxygen concentration and the source of protein in culture medium on the development of 2- to 4-cell goat embryos in vitro was investigated. Embryos were collected from superovulated Angora-Cashmere-cross goats 48 h after ovulation and cultured for 6 days in synthetic oviduct fluid (SOF) medium under one of two oxygen concentrations (20% or 7%) and in the presence of one of five protein sources; Miles bovine serum albumin (Miles BSA), Commonwealth Serum Laboratory bovine serum albumin (CSL BSA), goat serum (GS), fetal calf serum (FCS) and human serum (HS). In the presence of 20% oxygen the percentage of embryos reaching the expanded and/or hatched blastocyst stage in SOF medium containing Miles BSA was 29%, with a mean cell number per embryo of 28.1 +/- 6.0 (+/- s.e.m.). Use of an oxygen concentration of 7% significantly increased the percentage of embryos reaching this stage (80%, P less than 0.01) and the mean number of cells per embryo (65.3 +/- 8.2, P less than 0.01). The mean number of cells of the early-cleavage-stage embryos was significantly lower when the medium contained CSL BSA, GS or FCS (42.7 +/- 5.6, 29.0 +/- 6.1 and 21.3 +/- 3.2, respectively) than with Miles BSA (92.8 +/- 6.4) or HS (104.8 +/- 17.2) (P less than 0.01). Under 7% oxygen and with Miles BSA or HS, embryos were morphologically comparable to those developed in vivo, but the mean cell numbers in vitro were only approximately half those obtained in vivo.
