<p><strong>Objective: </strong>The present study deals with the cytotoxic activity of n-hexane and ethyl acetate extracts of <em>Calotropis gigantea</em> L. stem bark and its fractions such as A, B, C, D and E fractions on murine leukemia cell line P388.</p><p><strong>Methods: </strong>The crude extracts of <em>C. gigantea</em> stem bark were prepared using n-hexane and ethyl acetate solvents. The plant extracts were subjected to vacuum liquid chromatography followed by TLC. According to the similarity of stain patterns, the fractions were combined. The extracts and its combined fractions were then subjected for the phytochemical test. Cytotoxic activity of those extracts and its combined fractions were tested using MTT assay. Fraction D was subjected to gravity column chromatography followed by TLC. Then, fractions A, B, and D2 were crystallized and subjected to GC-MS.</p><p><strong>Results: </strong>The qualitative screening of n-hexane extract of <em>Calotropis gigantea</em> L. stem bark for secondary metabolites showed the presence of terpenoid, flavonoids, phenolics and coumarins. While the ethyl acetate extract contained phenolics, steroids, flavonoids, saponins and coumarins compounds. IC<sub>50 </sub>values for n-hexane extract and E fraction are 76.29 µg/ml and 18.48 µg/ml, respectively. In the ethyl acetate extract and C fraction obtained IC<sub>50</sub> values 57.05 µg/ml and 52.58 µg/ml.</p><p><strong>Conclusion: </strong>Cytotoxic activity from E fraction of n-hexane extract of <em>C. gigantea</em> stem bark is the most potent and containing flavonoids, phenolics and coumarins. The main components from several compounds of n-hexane extract of <em>C. gigantea</em> are germacrane-A, (-)-globulol, urs-12-ene and veridiflorol. </p>

In this study, the antimicrobial compounds present in leaves of Alphonsea madraspatana was identified by using RP-HPLC, LC/MS and GC/MS and found to be active against multiple drug resistant (MDR) bacteria e.g. Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa. Extraction of dried plant leaves was performed by solvent gradient technique. Antimicrobial study was performed with each extract. n-Hexane and methanol extracts were further subjected to analysis for identification of potent antimicrobial compounds due to their better antimicrobial activity as compared to remaining extracts. The methanolic extract was more active than n-hexane extract. As n-hexane extract contains non-polar compounds, it was subjected to GC-MS analysis and methanol extract was subjected to RPHPLC followed by LC/MS due to presence of polar components. Among the isolated compounds, four compounds were identified as potent antimicrobial. RP-HPLC and LC/MS analysis of methanol extract also ensured the presence of four potent antimicrobial compounds as follows: (i) kaempferol-3-O-rotinoside-orhamnoside (m/z 748), (ii) 3-hydroxypropylglucosinolate (m/z 377), (iii) luteolin-7-O-glucoside (m/z 448) and (iv) genistein-7-O-glycoside (m/z 432). Leaves of this plant may be used as potent antibacterial agents due to presence of antimicrobial compounds.

Although Acanthospermum hispidum has long been considered as a competitive invasive weed of agricultural plantations, it has recently become a sought after raw material to manufacture syrups in the treatment of asthma, bronchitis and fever. In the present study, the bioactive components of Acanthospermum hispidium entire plant have been accessed via GC-MS analysis. Six compounds from the hexane extract of the whole plant were identified. The most prevalent are glycerine (100.00%), 1-benzoypiperidine (40.26%) and erythritol (15.79%). Phytochemical screening investigation carried out using standard procedures showed the presence of Alkaloids, Steriods, Saponnins, Tannin, Flavonoids, Phenol, Terpenoids.

Antioksidan adalah suatu senyawa yang dapat menangkap radikal bebas. Salah satu sumber anti oksidan alami adalah dari tumbuh-tumbuhan. Dalam usaha pencarian antioksidan baru, telah dilakukan identifikasi senyawa antioksidan dari ekstrak n-heksana kulit batang Garcinia bancana Miq dengan metode 1,1-difenil-2-pikrilhidrazil (DPPH). Metode isolasi dilakukan dengan cara  kombinasi kromatografi (Kromatografi kolom vacum, kromatografi kolom gravitasi, kromatografi lapis tipis  sentrifugal dan kromatografi lempeng tipis)  Penentuan struktur molekul berdasarkan analisis data spektroskopi UV-VIS, IR, LC-MS, NMR proton dan karbon. Dari  hasil isolasi didapatkan suatu senyawa isoprenil bezofenon dengan bobot molekul 466,22 dan  rumus molekul C28H34O6 dengan nama IUPAC 3-(3,4-dihidroksibenzoil)-4-hidroksi-8-8-dimetil-1,7-bis(3-metilbut-2enil) bisiklo (3.3.1) non-3-ene-2,9-dione atau disebut bacanone yang diduga senyawa baru. Dari proses isolasi juga ditemukan senyawa atsiri b- caryophyllene, α-humulene dan b-cadinene serta stigmasterol. Hasil uji aktivitas antioksidan ekstrak n-heksana dan hasil isolat murni menunjukkan IC50 berturut-turut 17,78 ppm dan 12,79 ppm dimana pembanding kuersetin adalah 9,90 ppm.Kata kunci:  Garcinia bancana, antioksidan, DPPH, 3-(3,4-dihidroksibenzoil)-4-hidroksi-8-8-dimetil-1,7-bis(3-metilbut-2enil)bisiklo(3.3.1)no-3-ene-2,9-dione, bacanone. Antioxidants are compounds that can capture free radicals. One source of natural antioxidant is from plants. On searching  for new antioxidant, identification of antioxidant compound of the n-hexane extract of the stem bark of Garciniabancana Miq was done by  1.1-diphenyl-2-pikrilhidrazil (DPPH) method. Isolation active compound were done by combination of chromatographic methods (Flash column chromatography, gravitation column chromatography, cetrifugal thin layer chromatography and thin layer chromatography) .Determination of molecular structure by analysis spectroscopic data of UV-VIS, IR, LC-MS, NMR proton and carbon.Isolation results were      isophrenyl bezophenon with molecular weight 466.22 and the molecular formula is C28H34O6 IUPAC name 3 - (3,4-dihydroxybenzoil)-4-hydroxy-8-8-dimethyl-1,7-bis (3-methylbut-2enyl) bicyclo (3.3.1) non-3-ene-2,9-dione or named bacanone wich is suspected as a new compound. From the isolation were also found of known volatile compounds b - caryophyllene, α-humulene and b-cadinene and stigmasterol. The test results of antioxidant activity of  n-hexane extract and pure compound  showed IC50 respectively 17.78 ppm and 12.79 ppm which comparison with  quercetin is 9.90 ppm. St. John Wort (Hypercum perforatum L., Hypericaceae) has been used as a medicinal plant for a long period of time as this plant is characterized by a diversity of bioactive constituents which possess well documented pharmacological activities including antiviral, antimicrobial, anti-inflammatory, antioxidant, hepatoprotective and anti-tumoral activity. Nowadays, special interest is put on its essential oil as some experimental studies showed great biological and pharmacological potential. According this, the main goal of this study was GC/MS analysis of the essential oil, aroma components as well as n-hexane extracts of Hypericum perforatum that grows in Western region in R. Macedonia. GC/FID/MS analyses of the isolated essential oils from leaf, flower and herb resulted in the identification of 84 compounds. The fraction of sesquiterpenes was dominated in all examined oils and the main constituents were germacrene D (17.77-39.03%), E-caryophyllene (11.37-25.71%) and β-selinene (0.69-4.77%). GC/HS/MS analyses of the aroma components resulted in the identification of 23 compounds. Among them, isononane was identified as main aroma component (up to 75%). GC/FID/MS analyses of the n-hexane extracts resulted in the identification of 60 compounds which were characterized by the presence of terpenoid (mono- and sesquiterpene) components and non-terpenoid constituents mainly consisted of hydrocarbons and their oxygenate derivatives and related components. The non-terpenoid fraction represented the largest part of the analysed extracts. The most abundant were nonacosane (15.45-49.28%), octacosane (1.33-40.05%) and pentacosane (1.68-9.04%). The aerial parts of H. perforatum collected from Western part of R. Macedonia could be considered as a good source of essential oil with specific chemical profile as well as aroma components and high lipophilic compounds, but further investigation should be done in accordance to their possible commercial or medicinal use.  Hexane, dichloromethane, ethyl acetate, methanol and 50 % aqueous methanol extracts of leaf, flower and fruit of Melastoma malabathricum were screened for the presence of different classes of phytochemicals. In phytochemical screening, ethyl acetate, methanol and 50 % aqueous methanol extracts showed the presence of phenolics and flavonoids. Glycosides were found only in the 50 % methanol extract of fruit. Free radical scavenging activity of methanol extract of leaf, flower and fruit was determined using 2,2-Diphenyl-1-picrylhydrazyl (DPPH) free radical. Total phenolic content was measured spectrophotometrically using Folin-Ciocalteu, total flavonoids content by using aluminum chloride and total sugar content by using anthrone reagent. Gallic acid was used as a standard for the calibration of phenolics, catechin for flavonoids and glucose for carbohydrates. The highest amount of phenolic was found in the 50 % methanol extract of leaf (242.88 ± 3.52 mg GAE/g extract), the highest amount of flavonoids was found in the ethyl acetate extract of leaf (127.41 ± 6.33 mg CE/g extract). Total sugar in 50 % methanol extract of fruit was found to be 151.12 ± 8.79 mg GE/g extract. Methanol extracts of flower showed the strongest antioxidant activity with IC50 value of 17.23 μg/ml. Silyl derivative of hexane extracts were prepared by using N-methyl-N-(trimetylsilyl) trifluoroacetamide for gas chromatography-mass spectroscopic (GC-MS) analysis. The GC-MS analysis of hexane extract allowed the identification of 17 phyto-constituents by comparing the fragmentation pattern of each component in mass spectrum with the standard NIST mass spectral database.
