Absolute Circulating Pericyte Progenitor Cell Counts in Mice by Flow Cytometry: comparison of 2 single-platform technologies

Background Accumulating evidence indicates that circulating pericyte progenitor cells (CPPCs) may be angiogenic biomarkers in cancer and diabetes. Their validity as biomarkers depends on the accuracy of techniques used for enumeration. In this report, absolute CPPC counts were performed by 2 single-platform technologies. The reliability of the 2 methods, including retest reliability and intraobserver and interobserver variability, was assessed according to the intraclass correlation coefficient (ICC). The linear correlation and agreement among both methods were assessed, and the stability of CPPC numbers in blood samples was analyzed. Methods The blood samples were obtained from ICR mice. The samples were processed through a no-lyse, 1-wash procedure, and Syto16+CD45-CD31-CD140b+ CPPCs were analyzed by exclusion of dead cells and by fluorescence-minus-one control. CPPCs were enumerated by 2 methods: bead-based 123count eBeads count (eBioscience) and direct volume–based Accuri C6 Flow Cytometer count (BD). The cells were measured immediately and after storage of blood samples for 24 and 48 hours. Results There were excellent retest correlations and intraobserver and interobserver agreement in both methods. The 2 methods showed a high linear correlation (R2 = 0.923) and with a high level of agreement (0.986). It was demonstrated that CPPCs are unstable in blood samples. Conclusions In this study, 2 reproducible protocols for CPPC quantification were established. These protocols should facilitate future studies to further define the role of CPPCs as cellular biomarkers.

This article revises the mission and the name of the accounting profession in the context of strengthening the economic security of enterprises under the conditions of digitalization of the economy. The authors note that in the contemporary conditions of economic management, enterprises should form and ensure the functioning of the economic security of the enterprise at the proper level. The necessity of in-depth research of economic security at microlevel was considered, the factors influencing the stability of the enterprise were highlighted. High level of economic security of the subject of management consists in guaranteeing him maximum effective and stable functioning now and in future. Subjects of economic security were individual enterprises, and objects — their economic interests. The main goals of ensuring economic security of the enterprise in the part of accounting were singled out, the tasks of accounting policy, aimed at ensuring economic security, were determined. The prospects of the accounting profession in the process of ensuring economic security and reliable safe presentation of the results of doing business in reporting are substantiated. The study contains proposals on the revision of requirements for the qualifications of accountants in order to emphasize their activities aimed at strengthening the economic security of the enterprise.

Correlation coefficients between the results of political parties in the 2016 State Duma elections in the Russian Federation as a whole and in 26 regions, as well as in the elections of regional parliaments of 35 subjects of the Russian Federation in 2012–2015 were calculated. For the 2016 State Duma elections, data was used at all levels – regions, single-member electoral districts, TEC and PEC. It is noted that the “United Russia” correlations with all major parties are generally negative. A fairly high level of correlation is observed between the liberal parties. The main focus is on correlations between parliamentary opposition parties and parties with similar names. The correlation coefficients between the results of parties and candidates in the State Duma elections of 2011 and 2016 and the Presidential elections of 2012 and 2018 were also calculated, showing the stability of the geographical distribution of the electorate of the main parties. Regional differences in the nature of correlations between the main political parties are noted. It is assumed that correlations between parties reflect not so much their ideological closeness as the social closeness of their electorate. In this regard, it is noted that a positive correlation between the results of ideologically distant parties (“Yabloko” and the Communist party or “Yabloko” and “Rodina”) is associated with their reliance on the urban electorate and, perhaps, its most educated part. The reasons for voting for spoiler parties and the role of these parties in reducing the results of the main participants in the elections are discussed.

Background: Comfort evaluation techniques are commonplace in medicine. However, measures of lower-limb comfort are infrequently used in the sporting environment. The purpose of this study was to develop an instrument for measuring lower-limb comfort, which will extend previous work in the field of injury awareness. Methods: A lower-limb comfort index (LLCI) was developed for use in the environment of elite sport. Forty professional footballers participated in development of the index. The study had three components. A critical appraisal of the literature established the need for an LLCI. The second phase involved 20 professional footballers establishing and testing the components of the comfort index as an instrument for measuring comfort. Results: Nonparametric statistics (the McNemar test) in phase 2 indicated that the LLCI demonstrated good responsiveness to suitability (P = .019) and ease of use (P < .01). After a high level of agreement for responses, the third stage required 20 players to pilot test the reliability of the LLCI in a controlled environment. Repeated measures of difference between two periods for sum comfort (intraclass correlation coefficient = 0.99) and individual anatomical segments (κ = 0.72–1) provided confidence that the comfort index was reliable. Conclusions: The LLCI showed good trait construct to provide confidence to conduct a future study to investigate interrater consistency in a wider cohort of professional footballers under different conditions, such as match-day and training-week environments. Kosovo has made significant progress in institutional building and macroeconomic stabilization, even though, the opportunity to meet needy people was very slow. The lack of economic development seems to have a negative effect on the stability that will push economic growth. The most urgent challenge for Kosovo authorities was to maintain macroeconomic stability and achieve economic growth, in pursuit of the reduction of the high level of unemployment. Reducing foreign aid and spills in the private sector will make it very difficult to achieve this goal. Mitigation of emigration is a very important issue for Kosovo's economy.


Currently, implementation of the breeding programs, including the commonly recognized areas and classic breeding methods, cannot sufficiently ensure a quick and significant increase in the productivity of sugar beet hybrids, since its gene pool is almost exhausted. Based on the achievements in the field of genetics, new approaches to and opportunities in creating highly productive agrocoenoses of sugar beet have become popular. As a result of many years of work, results have been obtained about the nature of inheriting the resistance to glyphosate in individual heterozygous apo- and syncarpous forms in case of inbreeding and pair mating with the MC tester. The expression of target genes in the generations was monitored by the survival rate of sugar beet plants after the treatment with glyphosate. During the research, individuals with a high level of gene expression were selected. Upon self-pollination of initial heterozygous original forms, deviations from Mendelian segregation were observed in most cases. The criterion for assessing the stability of expression of glyphosate resistance genes in case of seed breeding was the compliance with the laws of Mendel among the analyzed descendants. In the initial stages of the research, the level of stability gene expression had been 10 – 15 % of the total number of analyzed plants. After four self-pollinations, the stability gene expression significantly increased, and genotypes with the resistance of 91 – 100 % were selected. The first apo- and syncarpous self-pollinating lines of sugar beet with high tolerance in the role of resistance donors have been created. The positive results of preliminary tests of the first glyphosate-tolerant hybrids need confirmation. Seeds and roots of resistant forms have been obtained for further research.

Background. The index of microcirculatory resistance is an invasive measure of coronary microvascular function that has to be calculated during maximal hyperemia, classically achieved with intravenous adenosine (IV). The aim of this study was to evaluate the use of intracoronary (IC) adenosine for the calculation of IMR. Methods and Results. 31 patients with stable coronary artery disease were included in the study. Coronary pressure and thermodilution measurements were obtained at rest and during maximal hyperemia using a pressure-temperature sensor-tipped coronary guidewire. Duplicate measurements were performed using first IC and then IV adenosine. Dispersion of transit times was comparable for IC and IV adenosine. IMR values based on IC vs IV adenosine showed a high level of agreement and an intraclass correlation coefficient of 0.90. Applying an upper normal limit of 25, misclassification of IMR using IC adenosine was seen in just one patient in whom IC adenosine resulted in a lower value. A simplified procedure based on a single bolus dose of saline did not change the level of agreement or the rate of misclassification. Conclusions. We found an excellent agreement between IMR values measured during hyperemia induced by IC as compared to IV adenosine. The use of IC adenosine may facilitate invasive assessment of microvascular function and is potentially time- and cost-saving with less patient discomfort as compared to IV infusion. This study made an attempt to understand the level of South Korean librarians’ awareness of the United Nations’ Sustainable Development Goals (SDGs) and IFLA’s support strategy for the SDGs, the degree of implementation of the UN’s SDGs support strategy by individual libraries, and how much the librarians agree with IFLA’s support strategy. As a result, first, this study revealed that public librarians had little knowledge of the UN SDGs 2030 agenda or the fact that IFLA has proposed support strategies for the UN’s SDGs. Second, this study found a high level of agreement among librarians with IFLA’s support strategies set for libraries to achieve the UN SDGs . Reflecting the two results above, it can be understood that librarians, though they were not well aware of the agenda of the UN SDGs 2030 or the library’s support strategies set by IFLA for the agenda, highly agree that the library’s support strategies suggested by IFLA are the role of libraries. Therefore, widely promoting the library’s support strategies proposed by IFLA for the UN SDGs 2030 is highly likely to encourage many librarians to participate actively.

Abstract Objectives Autoanalyzers are used in clinical haematology for analysis of blood samples in clinical as well as in nonclinical studies. The results from these analyzers vary from machine to machine. In this study, we compared the lymphocyte and neutrophil count of mouse blood between ADVIA 2120i, Horiba Yumizen H2500 and CellaVision analyzers against manual counting as gold standard. Methods Blood samples from 28 female BALB/c mice were collected and analyzed. Agreement between different autoanalyzers and manual counting were determined by Bland–Altman method. Results A high level of agreement was found between CellaVision and manual technique for lymphocyte (Bias=4.75, 95% limits of agreement −14 to 24) and neutrophil count (Bias=0.68 (−17 to 19)). Agreement in lymphocyte count was also observed between ADVIA and manual counting, but to a lesser extent compared to CellaVision (Bias=13.9 (−10.45 to 38.27)). However, no agreement was observed for ADVIA (Neutrophils), Horiba (lymphocytes and neutrophils) with manual counting. Conclusions Our data suggests that CellaVision could be used for the differential counting of neutrophil and lymphocytes in mouse blood sample.

AbstractOncogenic c-Myc is a master regulator of G1/S transition. Long non-coding RNAs (lncRNAs) emerge as new regulators of various cell activities. Here, we found that lncRNA SnoRNA Host Gene 17 (SNHG17) was elevated at the early G1-phase of cell cycle. Both gain- and loss-of function studies disclosed that SNHG17 increased c-Myc protein level, accelerated G1/S transition and cell proliferation, and consequently promoted tumor cell growth in vitro and in vivo. Mechanistically, the 1-150-nt of SNHG17 physically interacted with the 1035-1369-aa of leucine rich pentatricopeptide repeat containing (LRPPRC) protein, and disrupting this interaction abrogated the promoting role of SNHG17 in c-Myc expression, G1/S transition, and cell proliferation. The effect of SNHG17 in stimulating cell proliferation was attenuated by silencing c-Myc or LRPPRC. Furthermore, silencing SNHG17 or LRPPRC increased the level of ubiquitylated c-Myc and reduced the stability of c-Myc protein. Analysis of human hepatocellular carcinoma (HCC) tissues revealed that SNHG17, LRPPRC, and c-Myc were significantly upregulated in HCC, and they showed a positive correlation with each other. High level of SNHG17 or LRPPRC was associated with worse survival of HCC patients. These data suggest that SNHG17 may inhibit c-Myc ubiquitination and thus enhance c-Myc level and facilitate proliferation by interacting with LRPPRC. Our findings identify a novel SNHG17-LRPPRC-c-Myc regulatory axis and elucidate its roles in G1/S transition and tumor growth, which may provide potential targets for cancer therapy.

ObjectiveType 2 innate lymphoid cells (ILC2s), a recently identified population of lymphoid cells lacking lineage-specific receptors, promote type 2 immunity and tissue remodelling. However, the contributive role of ILC2s in the pathogenesis of systemic sclerosis (SSc) is unknown. We aimed to evaluate the levels and correlations with fibrotic manifestations in SSc.Methods69 patients with SSc and 47 healthy controls were included. Blood samples and skin sections were analysed by flow cytometry and immunohistochemically by staining two complementary panels of markers.ResultsDermal and circulating ILC2s were significantly elevated in patients with SSc compared with controls. Dermal, but not circulating ILC2s were activated. Stratification of the SSc population in patients with limited cutaneous SSc (lcSSc) and diffuse cutaneous SSc (dcSSc) demonstrated increased levels of ILC2s in both subgroups with significantly higher frequencies in dcSSc compared with lcSSc. Moreover, dermal and circulating ILC2 counts correlated closely with the modified Rodnan skin score and with the presence of pulmonary fibrosis.ConclusionsILC2 counts are elevated in patients with SSc and correlate with the extent of skin fibrosis and the presence of interstitial lung disease providing compelling evidence for profibrotic effect of ILC2s in SSc.
