Abstract 1. One of the two major protein components of fresh Hevea-latex serum has been isolated by an isoelectric precipitation method and the optimum conditions for the precipitation have been determined. 2. Attempts to isolate this component by fractional precipitation of the total serum proteins with ammonium sulfate gave markedly heterogeneous products. 3. A preliminary examination has been made of the purified protein. Analytical and solubility data indicate that it is a globulin lacking non-protein prosthetic groups.

Abstract 1. The serum from unpreserved rubber (Hevea brasiliensis) latex contains seven electrophoretically distinct protein components. The proteins from whole serum originating in Sumatra and Florida give very similar results in electrophoresis experiments. 2. The relationship between electrophoretic mobility and pH has been determined for five of the seven protein components of unpreserved total latex serum. The results are considerably different from those reported by workers with ammonia-preserved latex, and tend to clarify observed differences in the stability behavior of unpreserved and preserved latex. 3. Ammonia-preservation treatment rapidly alters the electrophoretic behavior of the native protein components of latex serum and reduces the number of resolvable components from seven to two. 4. The preparation of dry latex protein from rubber-free latex serum can be accomplished by the vacuum sublimation of frozen serum. This process does not appear to produce important changes in the electrophoretic properties of the total serum proteins. 5. Minor modifications of the electrodes and of the standard illumination system in the electrophoresis apparatus are described.

Rainbow trout serum was shown to be made up of three major protein components. These three major components did not coincide by electrophoretic mobility or by relative salt solubility with the classic albumin, α-, β-, and γ-globulin separation of the sera of higher animals. A component equivalent to γ-globulin of higher animals was found to be completely lacking in the sera of both immunized and non-immunized rainbow trout.Specific humoral antibodies in rainbow trout immune sera were found to be contained in the electrophoretically least mobile proteins. These proteins coincided most nearly to the β-globulins and possibly the least electrophoretically mobile α-globulins of higher animals. Salt solubility of the serum proteins containing the specific antibody assisted in this conclusion. The electrophoretically least mobile major serum protein components were found to contain all of the antibody activity. These proteins could be removed from immune serum by 20% sodium sulfate. The assumption was made that Aeromonas hydrophila antibodies in immunized rainbow trout blood serum may be made up of a complex of serum proteins with a broad range of salt solubility and electrophoretic mobilities.Serum proteins of rainbow trout could also be classified as euglobulins or pseudoglobulins. The serum proteins which contained the A. hydrophila antibodies were found to be euglobulins.

(1) Background: Daily caloric intake should aim to reduce the risk of obesity or poor anthropometric development. Our study objective was to analyze the association between food consumption, inflammatory status and anthropometric development; (2) Methods: We performed a prospective observational analytical research during September 2020 and April 2021 on a group of 160 healthy subjects, aged between 6 and 12 years old, by analyzing food ingestion, the basal metabolic rate, anthropometric development and the inflammatory status; (3) Results: IL-6 was significantly correlated to the sum of skinfolds, along with both serum proteins and triglycerides. The skin folds were significantly correlated with the caloric intake and with total fat intake, next to saturated and trans fats. Unlike the skin folds, the body weight was significantly correlated with the caloric intake along with some vitamins, such as Vitamin A and Vitamin B12. Inactive mass increased with excessive folic acid, Vitamin E, Vitamin K and saturated fat intake; (4) Conclusions: The inflammatory status was influenced by the ingestion of micronutrients, total serum lipids and proteins. The anthropometric development was associated with the ingestion of carbohydrates, energy balance and energy intake. We can conclude that daily menu and nutrition imbalances can influence both the risk of obesity and the inflammatory status.

This study describes, for the first time, the characterization of excretory–secretory antigens (ES-Ag) from Echinococcus granulosus protoscoleces, evaluating their usefulness in the immunodiagnosis of human cystic echinococcosis. ES-Ag were obtained from the first 50 h maintenance of protoscoleces in vitro. This preparation contained over 20 major protein components which could be distinguished by 1-dimensional SDS–PAGE with apparent masses between 9 and 300 kDa. The culture of of protoscoleces from liver produced a greater variety of excretory–secretory protein components than those from lung. Determination of enzymatic activities of secreted proteins revealed the presence of phosphatases, lipases and glucosidases, but no proteases. These findings were compared to those obtained from somatic extracts of protoscoleces and hydatid cyst fluid products. Immunochemical characterization was performed by immunoblotting with sera from individuals infected by cystic echinococcosis (n=15), non-hydatidic parasitoses (n=19), various liver diseases (n=24), lung neoplasia (n=16), and healthy donors (n=18). Antigens with apparent masses of 89, 74, 47/50, 32, and 20 kDa showed specificity for immunodiagnosis of human hydatidosis. The 89 and 74 kDa components corresponded to antigens not yet described in E. granulosus, whereas proteins of 41–43 kDa and 91–95 kDa were recognized by the majority of the non-hydatid sera studied.

Total serum protein values, hemocyte numbers, and muscle weights were determined for 216 intermolt lobsters immediately after their capture, and for 230 others held captive under a variety of dietary and environmental conditions. Average muscle values ranged from approximately 13% to the more normal 20–25% of the live animals' weight, depending upon experimental conditions. The total serum protein up to a level of 55 mg/ml was shown to be a reliable indicator of muscle weights, although the relationship was not identical for all lobster groups. It appeared to be modified chiefly by the areas from which the different groups were taken. Diet was more important than the temperatures (5 to 14 C) in affecting changes in muscle and serum protein values. Starvation caused a greater reduction (50 to 70%) in the size of the hepatopancreas than in the muscle. Histological examination of the hepatopancreatic tissue showed that the lipid content was markedly reduced upon starvation and that a degeneration of this organ was apparent for lobsters fed a beef liver and herring diet. Measurement of serum proteins would appear to be a useful technique in experiments on lobster nutrition and have value, within specified limits, for assessing the physiological condition of wild lobsters.

Erythropoietin (Epo) is produced primarily in the kidneys upon low blood oxygen availability and stimulates erythropoiesis in the bone marrow. Recombinant human Epo (rHuEpo), a drug developed to increase arterial oxygen content in patients, is also illicitly used by athletes to improve their endurance performance. Therefore, a robust and sensitive test to detect its abuse is needed. The aim of the present study was to investigate potential human serum biomarkers of Epo abuse employing a proteomic approach. Eight healthy male subjects were injected subcutaneously with rHuEpo (5,000 IU) every second day for a 16-day period. Serum was collected before starting the treatment regime and again at days 8 and 16 during the treatment period. Samples were homogenized and proteins separated by two-dimensional gel electrophoresis (2DE). Spots that changed significantly in response to rHuEpo treatment were identified by mass spectrometry. Both the number of reticulocytes and erythrocytes increased throughout the study, leading to a significant increase in hematocrit and hemoglobin content. In addition, transferrin levels increased but the percentage of iron bound to transferrin and ferritin levels decreased. Out of 97 serum proteins, seven were found to decrease significantly at day 16 compared with pre-Epo administration, and were identified as four isoforms of haptoglobin, two isoforms of transferrin, and a mixture of hemopexin and albumin. In support, total serum haptoglobin levels were found to be significantly decreased at both days 8 and 16. Thus a 2DE proteomic approach for discovery of novel markers of Epo action appears feasible.

Collagens are the major protein components of the Caenorhabditis elegans cuticle and are encoded by a large family of 40 to 150 closely related but nonidentical genes. We have determined temporal patterns of mRNA accumulation for a large number of collagen genes by screening recombinant phages and plasmids containing cloned collagen genes under high stringency conditions with 32P-labeled cDNA preparations specific for eggs or three postembryonic molts. We find that collagen mRNA levels are regulated both temporally and quantitatively during C. elegans development. Most genes studied exhibit one of four patterns of mRNA accumulation which correlate with changes in cuticle morphology and collagen protein composition during development. Our results suggest that, in general, there is a progressive activation of new collagen genes during normal development.

Abstract In the opinion of Pummerer, there is considerable doubt about the homogeneity of the fractions obtained by fractional precipitation of rubber from dilute benzene solutions, both with respect to the size of the molecules and to the state of aggregation. Based on the results of x-ray measurements of fibrous substances, to which type stretched rubber belongs, Meyer and Mark have advanced the hypothesis of the existence of chains of primary valences and of a micellar structure. PURIFICATION OF RUBBER There are four important and at the same time essentially different ways of carrying out the difficult process of purifying rubber. According to Harries, acetone extraction gives a colored product and leads to profound changes in the elastic properties. Staudinger adopted the method described by Wildmann in 1911, which involves the use of a mixture of acetone and chloroform. On the other hand, Pummerer and Koch used the method of fractional precipitation; the rubber after severe mastication was extracted with acetone, was dissolved in benzene, the liquid was allowed to stand for several weeks, the solution was decanted from the insoluble residue and was fractionally precipitated by alcohol and acetone. More recently this same method has been perfected by Pummerer and Meidel, and by this means a fraction of crystallized rubber was isolated from the mother liquors of the fractional precipitation. Finally Pummerer and Koch have purified rubber by treatment with an alkali, combining the precipitation method with the use of a solution of potassium hydroxide in methanol. Later this method was modified by Pummerer and Pahl. The use of latex in place of crude rubber is the most important development in obtaining a satisfactory product. De Vries and Beumée-Nieuwland have described in detail some results obtained with fresh latex. The total-rubber obtained by the methods just described contains, according to the quality and the age of the sample of latex, from 0.1 to 0.4 per cent of nitrogen which cannot be removed by washing, even when this is exhaustive.

Leukemia or cancer of the blood is the most common childhood cancer, Acute lymphoblastic leukemia (ALL), is the most common form of leukemia that occurs in children. It is characterized by the presence of too many immature white blood cells in the child’s blood and bone marrow, Acute lymphoblastic leukemia can occur in adults too, treatment is different for children. Children with ALL develop symptoms related to infiltration of blasts in the bone marrow, lymphoid system, and extramedullary sites, such as the central nervous system (CNS). Common constitutional indications consist of fatigue (50%), pallor (25%), fever (60%), and weight loss (26%). Infiltration of blast cells in the marrow cavity and periosteum often lead to bone pain (23%) and disturbance of normal hematopoiesis. Thrombocytopenia with platelet counts less than 100,000 are seen in approximately 75% of patients. About 40% of patients with childhood ALL present with hemoglobin levels less than 7 g/dL. Although leukocyte counts greater than 50,000/mm3 occur in 20% of cases, neutropenia defined as an absolute neutrophil count less than 500 is common at presentation and is associated with an increased risk of infection. The aim of this study was to investigate the differentiations in some biochemical parameters (Hb, PCV, total serum proteins Aspartate amino transferase(AST), Alanin amino transferase (ALT), and Malondialdehyde (MDA) in blood which can be conceder as a marker of ALL. Samples were collected from 50 patients (between 1-16 years old) diagnosed with ALL after one month treatment with induction therapy, compared with 30 control samples taken from healthy persons at the same age .
