Relationship between the fouling value and the wet weight of sessile organisms attached to various test plates in Mikawa Bay, central Japan.

Biofouling of external tags can damage both the animal and the tag. It is an inherent problem to many fish-tagging studies; however, it has received little attention. Between 2005 and 2007, we investigated the taxonomic group and wet weight of sessile multicellular eukaryonts growing on 33 B-type dart tags recaptured from raggedtooth sharks (Carcharias taurus Rafinesque, 1810) along the eastern coast of South Africa. All tags, except one from a shark that had been at liberty only for 13 days, contained some degree of macro-fouling that ranged from 0.031 to 7.687 g wet weight. Barnacles and hydroids were the first colonisers and were recorded on tags after only 47 days. Bivalves, polychaetes and algae succeeded within 60 days. The last two taxa to be recorded were ascidians and sponges. There was no significant relationship between either the biomass of sessile organisms (P = 0.17) or the number of taxa (P = 0.59) found growing on tags and their time at liberty. Calcium carbonate plugs, ranging in weight from 0.16 to 9.18 g, were found at the site of the tag insertion wound on 27.3% of recaptures. The present study highlights the extent of biofouling growth on tags and its potential to bias results from tagging studies.

Amphipoda are an important component of fish food. They serve also as bioindicators, intermediate hosts for parasitic organisms, and utilizers of primary and secondary production. In this work, dynamics of biodiversity and abundance of Amphipoda from the fouling community of mussel farm collectors in the open coastal zone near Karantinnaya Bay (Sevastopol, Black Sea) have been studied over 16 months. Eighteen species in 72 613 specimens of Amphipoda have been identified. Four species of Amphipoda dominate in the community in different seasons. Highly synchronous dynamics of number of Amphipoda and total wet weight of sessile organisms in the community was found (with correlation coefficient 0.95). Amphipoda have been systematized according to their biotopes, and the sizes of the Amphipoda populations have been evaluated.

3H-galactose (gal) and 3H-glucose (glu) were compared to determine which compound was preferable for pulse labeling newly formed hepatic glycogen. Control fed rats were used to achieve substantial and consistent levels of hepatic glycogen and to stimulate glycogen synthesis.Rats fed once daily for 4 hr achieved hepatic glycogen levels > 3% wet weight liver prior to injection by tail vein of a tracer dose of 3H-gal or 3H-glu. The rats were sacrificed 15-120 min later and liver was prepared by routine techniques for light (LM) and electron microscopic (EM) radioautography (RAG) and biochemical analysis.

ABSTRACT The response of the pigeon crop-sac to systemically acting prolactin (injected subcutaneously) was evaluated by measuring the wet weight of the responsive lateral lobes of the organ and by determining the dry weight of a 4 cm diameter disc of mucosal epithelium taken from one hemicrop. Of several different injection schedules tested, administration of prolactin in four daily injections was found to yield optimal responses. When compared with a graded series of prolactin doses, measurement of the mucosal dry weight proved to be a better method of response quantification than determination of the crop-sac wet weight with respect to both assay sensitivity and precision. The submucosal tissue of the crop-sac was estimated to constitute about 64 % of the total dry weight of the unstimulated organ and it was found to be relatively unresponsive to prolactin stimulation in comparison with the mucosa. The lipid content of the mucosal epithelium was determined using unstimulated crop-sacs or tissues which showed varying degrees of prolactin-induced proliferation. The fat content of the mucosal epithelial cells increased only slightly more rapidly than the dry weight or the defatted dry weight of the mucosa. Suggestions are made for the further improvement of the systemic crop-sac assay for prolactin.

Sperm storage tubules from the utero-vaginal junction of chickens, quails and turkeys were analysed for calcium and zinc using X-ray microanalysis of ultra-rapidly frozen tissue in a scanning electron microscope. This technique enabled the tubular fluid surrounding the stored spermatozoa and the intracellular content of the cells of the sperm storage tubules to be analysed separately and, by using standards with known concentrations, their elemental concentrations were estimated. The mean (+/- SEM) concentration of calcium in the tubular fluid from chickens, quails and turkeys was 17 +/- 3, 19 +/- 3 and 17 +/- 4 mmol kg(-1) wet weight, respectively. The intracellular calcium concentration of the cells of the tubules did not differ significantly from these values and was also similar in the mucosal epithelial cells of the utero-vaginal junction. Zinc was localized in the cells of turkey sperm storage tubules and tubular fluid, but at low concentrations. No zinc could be detected in corresponding structures from chickens and quails. The concentration of calcium in the tubular fluid is within the range known to inhibit the motility of spermatozoa, supporting this function for calcium during storage. Zinc is known to depress turkey sperm metabolism and it may also be involved in inducing quiescence of spermatozoa during storage in this species.
