Bioactive metabolites in Codonopsis pilosula are of particular interest as an immunostimulant. Methyl jasmonate (MeJA) plays an important role in the elicitation of metabolite biosynthesis. Here, we explored the response of metabolites to MeJA elicitation in C. pilosula adventitious roots and multiple shoots. The results showed that the biomass, polysaccharide, and lobetyolin content of adventitious roots exhibited the highest increases with 100 µmol·L−1 MeJA at the 16th day of subculture, whereas the atractylenolide III (a terpenoid) content increased extremely with 50 µmol·L−1 MeJA treatment at the 7th day of subculture. In addition, the biomass and lobetyolin content significantly increased at the 4th day after treatment. Similarly, the polysaccharide and lobetyolin content increased in multiple shoots. Further identification of different metabolites responding to MeJA by 1H-NMR showed an extremely significant increase of the lobetyolinin level, which coincided with lobetyolin. Accordingly, the precursor, fatty acids, showed a highly significant decrease in their levels. Furthermore, a significant increase in β-d-fructose-butanol glycoside was detected, which was accompanied by a decrease in the sucrose level. Accordingly, the enzyme genes responsible for terpenoid and carbohydrate biosynthesis, CpUGPase, and CpPMK, were up regulated. In conclusion, MeJA promoted culture growth and accelerated bioactive metabolite accumulation by regulating the expression of the metabolite biosynthesis related genes, CpUGPase and CpPMK in C. pilosula.

The genus Philodendron Schott comprises the following three currently accepted subgenenera: P. subg. Philodendron, P. subg. Pteromischum and P. subg. Meconostigma; however, these lack a well-defined classification. In the present study, we examined anatomically samples of adventitious roots in species of the group, so as to establish aspects relevant for taxonomic purposes. The anatomical analyses emphasised the characteristics of the steles in cross-sections of the root samples from regions near the apex to the most mature zones. A species of a closely related genus Adelonema, namely A. crinipes, was included in the study to clarify the results. Our results indicated notable differences in the species of the subgenus Meconostigma, mainly in terms of the presence (and variations) of a lobed stele, whereas the cylindrical stele stood out among the common characteristics in P. subg. Philodendron, P. subg. Pteromischum and the related species A. crinipes. Moreover, the characteristics shared by P. subg. Philodendron and P. subg. Pteromischum corroborated the phylogenetic hypothesis that these two taxa were more closely related to one another than to P. subg. Meconostigma.

Five 2,5-dimethyl-1-pyrrolidinecarboxanilides were effective inhibitors of the Hill reaction. However, only thecisisomers were active; thetransisomers were totally inactive. Experiments were conducted using14C-5328 (cis-2,5-dimethyl-1-pyrrolidinecarboxanilide). A correlation existed between resistance of various plants to 5328 and their ability to metabolize it to water soluble metabolites. Velvetleaf (Abutilon theophrastiMedic.) and proso millet (Panicum miliaceumL.) seedlings were very susceptible to 5328 and were unable to metabolize it. Tall morningglory [Ipomoea purpurea(L.) Roth] seedlings were highly tolerant to 5328 and converted it completely to its metabolites. Corn (Zea maysL. ‘DeKalb variety XL-45′) seedlings which were slightly susceptible to 5328 injury were able to metabolize up to 90% of the parent compound. Corn foliage uptake of14C-5328 applied to the soil surface occurred through the adventitious roots.

Propagation by stem cuttings and in vitro culture of apical bud explants were studied on Fraxinus excelsior L. Stem cuttings from 4- to 7-year-old trees growing under natural conditions sprouted only when cuttings were taken from dormant material. Only 6% of those that had sprouted developed roots by the 7th month of culture. Similarly, only apical bud explants harvested during the dormant period sprouted in vitro. Up to 87% of these sprouts developed two to four branching adventitious roots after 5 months of culture. During the initial phase of in vitro culture, the Quoirin and Lepoivre medium and the woody plant medium favoured sprout lengthening. During the phase of multiplication, up to three sprouts per explant developed with the woody plant medium in the presence of a combination of high 6-benzylaminopurine (3.0–4.0 mg∙L−1) and low indole-3-butyric acid (0.01–0.03 mg∙L−1) concentrations. Rooting was obtained in a medium without any growth regulators. Microscopic analysis showed a direct connection between the vascular elements of adventitious roots and stem of plantlet. Chromosome number in root apices of ash plantlets and ash trees grown under natural conditions was 2n = 46. The objective of this study was to discover the diploid progenitors of 80-chromosome Glycine tabacina with adventitious roots (WAR) and no adventitious roots (NAR). Three synthetic amphiploids were obtained by somatic chromosome doubling. These were (i) (G. latifolia, 2n = 40, genome B1B1,) × (G. microphylla, 2n = 40, genome BB) = F1(2n = 40, genome BB1) – 0.1% colchicine treatment (CT) – 2n = 80, genome BBB1B1; (ii) (G. canescens, 2n = 40, genome AA) × G. microphylla, 2n = 40, genome BB) = F1 (2n = 40, genome AB) – (CT) – 2n = 80, genome AABB; (iii) (G. latifolia, 2n = 40, B1B1) × G. canescens, 2n = 40, AA) = F1 (2n = 40, genome AB1) – (CT) – 2n = 80, genome AAB1B1. The segmental allotetraploid BBB1B1 was morphologically similar to the 80-chromosome G. tabacina (WAR), but meiotic pairing data in F1 hybrids did not support the complete genomic affinity. Despite normal diploid-like meiosis in allotetraploids AABB and AAB1B1, AABB was completely fertile, while pod set in AAB1B1 was very sparse. Morphologically, allotetraploid AABB was indistinguishable from the 80-chromosome G. tabacina (NAR) but in their F1 hybrids, the range of univalents at metaphase I was wide (4–44). The allotetraploid AAB1B1 did not morphologically resemble the 80-chromosome G. tabacina (NAR). However, the F1 hybrid of AABB × AAB1B1 showed normal meiosis with an average chromosome association (range) of 1.7 I (0–4) + 39.2 II (38–40). Based on this information, we cannot correctly deduce the diploid progenitor species of the 80-chromosome G. tabacina (NAR). The lack of exact genome homology may be attributed to the geographical isolation, natural mutation, and growing environmental conditions since the inception of 80-chromosome G. tabacina. Thus, it is logical to suggest that the 80-chromosome G. tabacina (NAR) is a complex, probably synthesized from A genome (G. canescens, G. clandestina, G. argyrea, G. tomentella D4 isozyme group) and B genome (G. latifolia, G. microphylla, G. tabacina) species, and the 80-chromosome G. tabacina (WAR) complex was evolved through segmental allopolyploidy from the B genome species.Key words: Glycine spp., allopolyploidy, colchicine, genome, intra- and inter-specific hybridization, polyploid complex.

The aim of this work was to study the effects of soluble aluminium on the morphology and growth of the adventitious root system, aerial biomass and grain yield of maize (Zea mays). The analysis focuses on two hybrid cultivars (Al-sensitive HS7777 and Al-tolerant C525M). Experiments were carried out in the field and in a rhizotron in Curitiba, Paraná, Brazil. In the field, four levels of lime application were used: T0 = 0 t ha−1, T1 = 3.5 t ha−1, T2 = 7.0 t ha−1, and T3 = 10.5 t ha−1. Two levels were used in a rhizotron: T0 and T3. In the surface horizon (0–15 cm), the Al concentrations of the soil solution were: T0 = 15, T1 = 5.1, T2 = 4.4, and T3 = 3.1 μM. In the field, neither Al concentration in the soil solution nor cultivar affected the number of primary adventitious roots per internode or the total number of primary adventitious roots. However, root diameter, plant population and grain yield of the two cultivars confirmed the differences in Al tolerance between them. Al was observed to have an adverse effect on the grain yield from C525M, while low yields from HS7777, at all levels of Al, precluded any response to liming. In the rhizotron studies, Al concentration and cultivar affected the root branching and total root length. Cultivar C525M had more branches and total root length than HS7777, mainly at low concentrations of soil Al solution, leading to greater spatial colonization of the soil down to 0.9 m depth.

Abstract Mini-cutting is a technique with large applications in various crops, mainly due to the increase in the percentage and quality of adventitious roots, reducing time for the formation of clonal seedlings. The aim of this study was to evaluate IBA levels and substrates on the rooting of UENF/CALIMAN 02 hybrid papaya mini-cuttings. To perform the experiment, papaya mini-cuttings were taken from mother plants grown in pots in greenhouse, induced to produce shoots through pruning and growth regulator applications. Mini-cuttings were fixed in vermiculite or coconut fiver substrates placed in alveolate trays with 4.5x4.5x5.0 cm cells, and styrofoam trays were placed in plastic trays where different IBA levels were added in a modified Hoagland solution. After 45 days, rooted buds were transplanted to plastic pots of 600 mL of volume with soil, sand, well-cured bovine fertilizer, in the proportion of 3:1:1, remaining for 45 days. When they were taken from pots, roots were carefully washed, and the length of shoots, length of the largest root, dried mass of shoots and radicular system and root percentage were measured. The experiment was set up in a randomized complete block 5 x 2 factorial design, with 5 IBA levels: 0; 2.5; 5.0; 7.5 and 10 mg L-1, two substrates: vermiculite and coconut fiber, three replicates, with six plants per replicate. IBA levels of 5.0 mg L-1 and substrate vermiculite are the most adequate for the rooting of ‘UENF/CALIMAN 02’ papaya mini-cuttings in semi-hydroponic system in alveolate styrofoam trays with 4.5x4.5x5.0 cm cells.
