Several methods for isolating adrenocorticotrophin from small quantities of porcine and bovine pituitary tissue are compared. Initial extraction of the hormone by an acid–acetone technique was simpler and more efficient than one employing acetic acid extraction and ether precipitation. Subsequent purification procedures utilizing adsorption of the peptide on to oxycellulose realized the highest yields. CM-cellulose-column chromatography followed by Sephadex-gel filtration were suitable final steps for obtaining highly purified adrenocorticotrophin. The purity of the hormone was demonstrated by determining its amino acid composition, C-terminal analysis, polyacrylamide-gel electrophoresis, chymotrypsin digestion and paper electrophoresis and by radioimmunoassay and bioassay. Adrenocorticotrophin was found to be rapidly destroyed in intact and especially in homogenized glands kept at room temperature. At 4° the rate of destruction was less rapid and at −20° losses were minimal.

In this paper, an automatic nucleic acid extraction instrument is described. It consists of mechanical components, motor controlling module, temperature controlling module and computer. Also, a prototype has been designed and constructed. The maximum sample throughput of this prototype is 96, the sample volume is 300-1000ul, the maximum sample handling time is about 60min, the sample temperature controlling range is from room temperature to 95oC, and the accuracy of temperature controlling achieved is about ± 2oC.

Attention was paid in the present paper to the fact that the precision of the values obtained by different methods for the total organic phosphorus in soil cannot be very high. Even the variation caused by the treatment of the extracts and connected with the colorimetric estimation of phosphate in the solution makes it impossible to report the results more accurately than by 10—20 ppm organic P, at least if routine analyses are in question. Although the somewhat modified methods of Dean, Wrenshall and Dyer,. Pearson, and Mehta et al. yielded equal results for the organic phosphorus content of the respective mineral soils and of most of the peat soils analyzed, the treatment with cold alkali in the Wrenshall and Dyer procedure apparently failed to extract the organic phosphorus from two peat samples as quantitatively as the treatment with hot alkali in the other methods. On the basis of this observations a new modification of the method of Wrenshall and Dyer was proposed. It consists of an extraction of 1-g sample with 25 ml of 4 N sulphuric acid at room temperature for 18 hours, followed by washing with water and two successive extractions with 100 ml of 0.5 N sodium hydroxide, the first of them for 18 hours at room temperature, the second for 4 hours at 90°C. This method was found to extract from 40 peat soil samples on the average about 97 per cent of the total phosphorus dissolved by the Kjeldahl digestion and about 9 per cent more organic phosphorus than the method of Pearson. Experiments concerning the ignition and acid extraction procedures indicated that the method of Ghani was not suitable for the determination of organic phosphorus in the twelve samples analysed. The extraction with sulphuric acid showed no marked differences between the increase in the soluble phosphorus due to the ignition when the ratio of extraction was varied from 1:40 to 1:200, and the extractant from 0.2 N acid to 5 N acid. The results obtained for 40 peat samples by ignition for one hour at 600°C and extraction of the ignited and untreated samples with 0.2 N sulphuric acid in a ratio of 1:100 for half an hour were on the average 8 per cent higher than those given by the proposed acid-alkali extraction. The total organic phosphorus content of soil may probably be somewhat higher than the figure yielded by the acid-alkali extraction and slightly lower than the value obtained by the ignition method. For the present, the most reliable result seems to be found in the average of the data given by these two methods.

A tightly packed regularly coiled DNA ring is assumed to be present inside the polyoma virion. Applying the “one-step” technique of nucleic acid extraction from virions while being adsorbed to a protein monolayer, we studied the configurational changes of the DNA.Intact polyoma virions were mixed with DFP-trypsin (100μg/ml) and spread onto a subphase of a mixture of urea and sodium perchlorate (6M and 2M, or 3M and 1M respectively), at pH 8.2 and room temperature. After spreading the film, aliquots were transferred to grids at different periods of time, and the DNA stained by acetonic uranyl acetate.The length distribution of the DNA molecules measured from electron micrographs is shown in Fig. 1. The number of twists present at different times after spreading were also counted. At 12 minutes, with 3M urea and 1M sodium perchlorate in the subphase, the DNA was found to be 85% supertwisted (Fig. 2); 12% was opened to rings, and 3% existed as linear filaments.

Environmental contextIn the environment, iron exists mainly as FeII and FeIII and plays an important role in biogeochemical processes. The FeII and FeIII content is often quantified by hydrochloric acid extraction and the acid is thought to prevent FeII oxidation by oxygen. However, we found that with increasing HCl concentration and temperature, oxidation of FeII by oxygen is accelerated. Therefore, in order to obtain reliable results extractions should be performed with dilute HCl or in the absence of oxygen. AbstractHCl is commonly used to stabilise FeII under oxic conditions and is often included in Fe extractions. Although FeII oxidation by molecular O2 in HCl is described in the field of hydrometallurgy, this phenomenon has not been systematically studied in environmentally relevant systems. The extent of FeII oxidation by O2 during extraction of soils and magnetite by HCl and in HCl/FeCl2 solutions was therefore quantified. FeII was stable in 1 M HCl at room temperature for several days, whereas in 6 M HCl at 70°C, 90% of the FeII was oxidised within 24 h. In the absence of O2, no FeII oxidation occurred. Experiments at low pH with increasing H+ or Cl– concentration alone and geochemical modelling suggested that the formation of complexes of FeII and HCl may be responsible for the observed FeII oxidation. The use of strictly anoxic conditions for Fe extraction by HCl to obtain reliable Fe redox speciation data is therefore recommended.
