A procedure that provides information on the suitability of barley for malting and, brewing purposes is described. Three parameters are measured: mash filtration rate, wort viscosity and wort-reducing sugar content, as maltose. Maltose is determined by an auto-analyzer method and this is a satisfactory alternative to the specific gravity procedure for the estimation of percent extract. The addition of these criteria to those already in use enables more discriminatory selection at early generations of barley-breeding programs. The technique requires only one sample extraction and 18 g of laboratory malt.

Although most of our tree-breeding programs are concerned with coniferous forest species, we must not neglect the valuable hardwoods exploited in our region. The need of these programs for hardwoods is undeniable and they should be based, as much as possible, on genecological studies.One such study illustrates the wide variation in genetic adaptation of Juglans nigra and indicates its practical implications. Further, these results were obtained from a short-term study of 17 provenances for variations in frost-hardiness at a single location.Intra-specific variation is being investigated in Juglans cinerea, a more common species in our region than the one previously mentioned and one which is also in strong demand for furniture-making and wood sculpture. The resulting data will permit the selection of the best seed zones. Such a breeding program could eventually support far more than small, local industries.Another project involving Acer saccharum describes the variation in sugar-content of sap in three maple stands sampled each spring from 1965 to 1969. Two stands were very homogeneous compared to the third. The magnitude of the differences maintained throughout the study period led to the selection of nine genotypes whose sugar contents varied from 4.6% to 9.7%, indicating maximum genetic gains on the order of 150%.Continued research in vegetative propagation of Acer saccharum is needed for the production of clonal populations for breeding programs.

Identification and deployment of disease resistance genes are key objectives of Australian barley breeding programs. Two doubled haploid (DH) populations derived from Tallon × Kaputar (TK) and VB9524 × ND11231 (VN) crosses were used to identify markers for net type net blotch (NTNB) (Pyrenophora teres f. teres). The maps included 263 and 250 markers for TK and VN populations, respectively. The TK population was screened with 5 pathotypes and the VN population with 1 pathotype of NTNB as seedlings in the glasshouse. In addition, the TK population was subjected to natural infection in the field at Hermitage Research Station, Qld. Analyses of the markers were performed using the software packages MapManager and Qgene. One region on chromosome 6H was strongly associated with resistance to NTNB in both populations (R2 = 83% for TK and 66% for VN). In the TK population, 2 more quantitative trait loci (QTLs) were identified on chromosomes 2H and 3H, with R2 values of 30% and 31%, respectively. These associations were consistent over all pathotypes studied during the seedling stage. The same QTL on chromosome 6H was also found to be highly significantly associated (R2 = 65%) with the adult plant (field) response in the TK population. There are several very closely linked markers showing strong associations in these regions. Association of the 4 markers on chromosome 6H QTL with resistance to the NTNB has been validated in 2 other DH populations derived from barley crosses Pompadour × Stirling and WPG8412 × Stirling. These markers present an opportunity for marker assisted selection of lines resistant to NTNB in barley breeding programs.

<p>Rumput laut dengan kandungan karbohidrat dan lipid yang tinggi dianggap menjadi sumber energi terbarukan generasi ketiga. Penelitian ini bertujuan menentukan kadar gula pereduksi optimum, mengetahui tekstur permukaan serbuk sebelum dan sesudah hidrolisis serta menentukan kadar etanol hasil fermentasi. Kandungan karbohidrat rumput laut <em>Ulva reticulata</em> dapat dikonversi menjadi gula heksosa dan pentosa (glukosa, arabinosa, ramnosa, dan xilosa) melalui hidrolisis asam. Campuran gula optimum hasil proses hidrolisis kemudian dikonversi menjadi etanol menggunakan ragi <em>Saccharomyces cerevisiae</em>. Fermentasi dilakukan dengan konsentrasi inokulum 10% (v/v) selama 5 hari pada temperatur 30 °C dan pH 4,5. Analisis tekstur permukaan sampel dilakukan dengan <em>Scanning Electron Microscopy</em> (SEM). Analisis gula pereduksi dilakukan dengan metode DNS (Dinitro salisilat). Analisis etanol dilakukan dengan uji kualitatif dan kuantitatif menggunakan metode berat jenis dan kromatografi gas. Hasil penelitian menunjukkan bahwa kadar gula pereduksi meningkat seiring meningkatnya suhu hidrolisis pada suhu      75 °C ke 150 °C (2,3 – 23,7 g/L) dan mengalami penurunan kadar pada suhu 175 °C menjadi 17,1 g/L. Hasil analisis dengan variasi konsentrasi terhadap waktu 30, 40, 50, dan 60 menit terlihat bahwa kadar gula pereduksi meningkat seiring meningkatnya waktu hidrolisis dari 30 menit sampai 50 menit yakni 23,7 – 33,4 g/L dan mengalami penurunan pada waktu 60 menit yakni 19,2 g/L. Kadar gula pereduksi optimum sebesar 33,4 g/L  pada suhu 150 °C dengan konsentrasi asam 2 % pada waktu hidrolisis 50 menit. Tekstur permukaan serbuk sebelum dan sesudah hidrolisis mengalami perubahan yang signifikan. Hasil uji kualitatif etanol hasil fermentasi dibuktikan dengan adanya perubahan warna dari jingga menjadi biru. Hasil uji kadar etanol dengan metode berat jenis yakni sebesar 1% dan metode kromatografi gas sebesar 5,02%.</p><p> </p><p><strong>Bioethanol Production as Renewable Energy from <em>Ulva Reticulata</em> Seaweed from Timor Island. </strong>Seaweed with carbohydrate and lipid content is considered to be the third generation of renewable energy sources. The carbohydrate content of <em>Ulva reticulata</em> seaweed can be converted into hexose and pentose sugars (glucose, arabinose, ramnose, and xylose) through acid hydrolysis. The optimum sugar mixture resulting from the hydrolysis process is then converted to ethanol using <em>Saccharomyces cerevisiae</em> yeast. Fermentation was carried out with an inoculum concentration of 10% (v/v) for five days at a temperature of 30 °C and a pH of 4.5. Analysis of the surface texture of the sample was carried out by <em>Scanning Electron Microscopy</em> (SEM). Reducing sugar analysis was performed using the DNS (Dinitrosalicylate) method. Ethanol analysis was carried out by qualitative and quantitative tests using specific gravity and gas chromatography methods. The results showed that the reducing sugar content increased with increasing hydrolysis temperature at 75 °C to 150 °C (2.3 – 23.7 g/L) and decreased levels at 175 °C to 17.1 g/L. The results of the analysis with various concentrations of 30, 40, 50, and 60 minutes showed that reducing sugar levels increased with increasing hydrolysis time from 30 minutes to 50 minutes, namely 23.7 – 33.4 g/L and decreased at 60 minutes, namely 19.2 g/L. The optimum reducing sugar content was 33.4 g/L at 150 °C with an acid concentration of 2% at 50 minutes of hydrolysis. Powder surface texture before and after hydrolysis experienced significant changes. The qualitative test results of fermented ethanol are evidenced by a change in color from orange to blue. The results of the ethanol content test using the specific gravity method were 1%, and that using the gas chromatography method was 5.02%.</p>

When stored at temperatures less than 10 °C, tubers of all cultivated potatoes exhibit cold-induced sweetening (CIS) during which starch degrades to sucrose, glucose, and fructose. Upon frying at high temperatures, the reducing sugars (Fru, Glu) interact with free amino acids via the non-enzymatic Maillard reaction to form dark-colored chips that are unacceptable to consumers. In addition, scientists recently discovered that the toxic chemical acrylamide is also produced during frying. Although storage at warmer temperatures reverses CIS and circumvents dark chip production, the probability of storage loss due to shrinkage and disease increases. Wild Solanum species form the backbone of many potato-breeding programs. In this study, we evaluated 36 different plant introductions (PI) including 20 different species, grown in Madison and Rhinelander, Wis., to identify germplasm resistant to CIS for genetic analysis. After storage for 2–3 months at 4 °C, tuber sugar and amino acid content were analyzed via HPLC and slices were fried to determine chip color. Sugar and chipping data support previous research indicating CIS resistance in S. okadae, S. raphanifolium, and S. phujera. Interestingly, some germplasm selections with high reducing sugar content produced light-colored chips, indicating exceptions to the typical correlation between reducing sugar content and chip color. Genetic bases to these exceptions are under evaluation.

Lignocellulose wastes are the most abundant residues on the surface of the earth. This project studies the possibility of ethanol production from a forestry waste. Wood wastes from Gmelina arborea were treated with dillute sulfuric acid to break down the lignin component. Fermentation for ethanol production was done using baker’s yeast (Saccharomyces cerevisiae ATCC 204508/S288c) for 120 hours using submerged fermentation, and the pH, reducing sugar, specific gravity and lignin content were determined using standard techniques. Ethanol concentration and yield were measured via vinometer and ethanol standard curve techniques. From the results, the highest pH was obtained at 72 hours of the fermentation period. The reducing sugar content and specific gravity decreased over the fermentation time . The acid-pretreated wood wastes gave a maximum ethanol concentration of 3.84 % and a yield of 7.60 ml/g as measured from the vinometer and ethanol standard curve methods at 72 and 96 hours of fermentation, respectively. About 13.6% v/v of ethanol was recovered from the distillation process employed to separate the components of the product generated after fermentation. The observations in this research reveal the possibility of producing ethanol from G. arborea wood wastes and under optimized culture conditions. Gravel, bone, carbonized seeds, and charcoal recovered from a 2,000 year old cereal pit were successfully separated by chemical flotation. Specific gravities of each material were determined experimentally, and then for each pair of materials to be separated (e.g., bone from gravel, seeds from charcoal), a solution with a specific gravity intermediate between those of the two materials was used for flotation. Optimal sample sizes and possible sources of error are discussed, as are criteria for selection of solutions. A procedure for evaluating the effect of solutions and flotation practices on test seeds is also presented.

Program pemuliaan ikan mas varietas Punten dilakukan dengan seleksi individu terhadap karakter bobot ikan. Pembentukan populasi dasar untuk kegiatan seleksi dilakukan dengan memijahkan secara massal induk ikan mas yang terdiri atas 20 induk betina dan 21 induk jantan yang dikoleksi dari daerah Punten, Kepanjen (delapan betina dan enam jantan), Kediri (tujuh betina dan 12 jantan), Sragen (27 betina dan 10 jantan), dan Blitar (15 betina dan 11 jantan). Larva umur 10 hari dipelihara selama empat bulan. Selanjutnya dilakukan penjarangan sebesar 50% dan benih dipelihara selama 14 bulan untuk dilakukan seleksi dengan panduan hasil sampling 250 ekor individu setiap populasi. Seleksi terhadap calon induk dilakukan saat umur 18 bulan pada populasi jantan dan betina secara terpisah dengan memilih berdasarkan 10% bobot ikan yang terbaik. Calon induk yang terseleksi kemudian dipelihara hingga matang gonad, kemudian dipilih sebanyak 150 pasang dan dipijahkan secara massal. Didapatkan respons positif dari hasil seleksi berdasarkan bobot ikan, yaitu 49,89 g atau 3,66% (populasi ikan jantan) dan 168,47 g atau 11,43% (populasi ikan betina). Nilai heritabilitas untuk bobot ikan adalah 0,238 (jantan) dan 0,505 (betina).Punten carp breeding programs were carried out by individual selection for body weight trait. The base population for selection activities were conducted by mass breeding of parent consisted of 20 female and 21 male collected from area Punten, eight female and six male (Kepanjen), seven female and 12 male (Kediri), 27 female and 10 male (Sragen), 15 female and 11 male (Blitar). Larvae 10 days old reared for four moths. Then after spacing out 50% of total harvest, the offspring reared for 14 months for selection activity based on the sampling of 250 individual each population. Selection of broodstock candidates performed since 18 months age on male and female populations separately by selecting based on 10% of fish with best body weight. Candidates selected broodstocks were then maintained until mature. In oder to produce the next generation 150 pairs were sets and held for mass spawning. The results revealed that selection response were positive, 49.89 g (3.66%) for male and 168.47 (11.43%) for female. Heritability for body weight is 0.238 (male) and 0.505 (female).

Ayurveda is the healing medicine. Dosha, Dhatu, Mala and Agni play important role in maintaining health of a person, but without equilibrium of Atma, Indriya and Manas they are helpless. Manas is one entity which is the controller of health. There are several herbs which have direct impact on Manas, among them Mandukaparni is one. Swarasa of Mandukaparni is highly effective, but it is not easily available for children. This study is intended to make Syrup form of Mandukaparni and evaluate its pharmacognostical parameters. According to the methodology refractive index, total solids, specific gravity, reducing and non Reducing sugar and HPTLC parameters were assessed. The results were found to be genuine fulfilling the standard protocol. This study is under taken to evaluate the pharmacognostic properties of Mandukaparni syrup.
