A method of chemical peeling which provides a fast and accurate color reading of barley aleurone at the flint state of maturity is presented. The grains are steeped for two to five hours in 50% sulfuric acid to peel the investments outside of the aleurone. The acid does not reach the aleurone, because of an impermeability barrier either between the aleurone envelope and nucellar epidermis or in the aleurone envelope itself. Consequently, the chemical peeling technique has made the classification of barley aleurone color segregants precise. Because it does not impair seed viability, the technique has potential application in studies on aleurone physiology and biochemistry.
