RUMEN MOTILITY RESPONSES TO ADRENALINE AND NORADRENALINE AND ORGAN WEIGHTS OF WARM- AND COLD-ACCLIMATED SHEEP

The rumen motility responses to 2.5 h adrenaline (A) and noradrenaline (NA) intrajugular infusions at 0, 0.15, 0.30, 0.60 and 0.90 μg∙kg−1∙min−1 were studied in 10 shorn wethers which had been acclimated for 4–8 wk to either a warm (19–24 °C) or cold (8–13 °C) environment. Preinfusion rumen motility was not significantly influenced by acclimation temperature. Adrenaline infusions at the highest dose rates depressed motility in both warm-acclimated (WA) and cold-acclimated (CA) sheep and during the final 90 min of infusion the inhibition appeared greater in the WA sheep. Noradrenaline resulted in a depression of two contractions per hour in rumen motility of WA sheep but an increase of five contractions per hour in CA sheep. The results suggest that the CA sheep were resistent to the inhibitory effects of catecholamines on rumen motility. Six WA and six CA sheep were sacrificed and internal organs and gastrointestinal content weights were compared. Cold acclimation was associated with a significant increase in the weight of the liver, kidneys, reticulorumen and small intestine and of the content of the colon. Because CA sheep were fed 800 g more feed per day than WA sheep the effect of cold acclimation on organ and organ content weights cannot be separated from the effect that the increase in intake alone might have had. In experiments in which two groups of animals of different mean body weight are compared, individual organ weights of the animals can be expressed as absolute weights, as fractional weights, or as absolute weights statistically regressed onto constant body weights. The second, and commonest, mode of expression involves the assumption that the part is directly proportional to the whole, and this is shown to be unlikely for all organs except the muscle mass. Practical as well as theoretical justifications for the use of regressed weights (which utilize the actual slope of the line relating the organ weight to the whole) are given.The experimental data are from white rats kept for 4 weeks in a warm (30 °C.) or a cold (6 °C.) environment. It is shown that cold adaptation had no effect on brain, genitals, and lung weights, but that it reduced the growth of muscle, pelt, fat, skeleton, spleen, and thymus, and that it hypertrophied the liver, intestine, kidney, heart, and adrenals. Apparently cold acclimated rats are smaller than the controls mainly because they have a smaller muscle mass.

An analysis has been made of the organ weights of normal rabbits exposed to a constant illumination having none of the shorter ultraviolet rays and of other rabbits kept in darkness for periods of 2 to 12 weeks. The environment of constant illumination was associated with a well marked decrease in the relative weights of most organs, and in certain instances this occurred when the organ weights of the controls were becoming increasingly large. There was also an associated effect of stabilization of organ weight. The majority of the organs of rabbits caged in constant darkness also showed a tendency toward decreased and stabilized weights, but these effects were less pronounced than in the rabbits caged under conditions of constant illumination. A notable exception to this general result was the weight of the liver which was markedly increased. The results of this experiment support the conception that there is a relationship between light and the physical state of the animal organism which may be expressed in the concrete form implied by the trend or direction of organ weight.

The goal of enteral nutritional support for infants born preterm or small for gestational age (SGA) is to achieve normal growth and development. Yet, this is difficult to achieve because of intestinal immaturity. Our objective was to determine if birth weight, protein intake, and the growth promoters leucine (10 g/L) or calcium-ß-hydroxy-ß-methylbutryate (HMB; 1.1 g/L) would affect trajectories of intestinal growth and functions and weights of other organs. Preterm pigs were delivered at gestational day 105 (91% of term) and fed for 6 or 7 days isocaloric formulas that differed in protein content (50 g or 100 g protein/L), with and without the growth promoters leucine or HMB. For comparative purposes organ weights were measured within 12 h after delivery for six term pigs of low and six of average birth weights. The responses of intestinal growth and total intestinal brush border membrane carbohydrases to protein level and supplemental leucine were of greater magnitude for preterm pigs of lower birth weight. Forskolin stimulated chloride secretion in the proximal small intestine was lower for pigs fed the low protein milk replacers. Capacities of the entire small intestine to transport glucose (mmol/kg-day) were not responsive to protein level, leucine, or HMB, and did not differ between small and large pigs. Relative organ weights of the small and average weight term pigs were similar, but some differed from those of the preterm pigs suggesting preterm birth and the standards of care used for this study altered the trajectories of development for the intestine and other organs. Although leucine is an effective generalized growth promoter that enhances gut development of small preterm pigs, it does not mitigate compromised neurodevelopment. Our findings using preterm pigs as a relevant preclinical model indicate nutrition support strategies can influence development of some gastrointestinal tract characteristics and the growth of other organs.

In the present study, effects of ileal infusions of nutrients on motor patterns of the proximal small intestine and on gastric emptying were investigated in dogs. An acaloric meal was administered orally, and equicaloric loads of amino acids, oleate, and glucose were infused into the ileum at different doses (0.3, 0.6, and 0.9 kJ/min). The computerized analysis of motor patterns was focused on the differentiation between stationary and propagated contractions recorded by closely spaced extraluminal strain gauges. All three nutrients exerted inhibitory effects on gastric emptying and on contraction force and frequency of the proximal small intestine. Additionally, the propulsive motor pattern induced by the acaloric meal was modulated by reducing the number of contraction waves and their length of spread. All the effects were dose dependent. Among the three nutrients, glucose significantly changed motility at lower doses compared with amino acids and oleate. We conclude that in dogs the ileal brake mechanism is induced by all three nutrients and that it influences not only contraction force and frequency but also the motor patterns of the proximal small intestine.

An experiment was carried out to examine the effects of supplementing diets for weanling pigs with oligosaccharides and LAC on nutrient digestion and bacterial populations and metabolites in the small intestine. Twelve barrows, weaned at 28 d, were fitted with a simple T-cannula at the distal ileum. The BW of the pigs at weaning and at the conclusion of the experiment were 9.1 and 13.8 kg, respectively. The pigs were fed four diets based on barley, wheat and soybean meal according to a two-period change-over design. The diets were formulated to contain 18% CP. Transgalactosylated oligosaccharides (0.2%), GUO (0.2%) and LAC (1%, 4-O-β-D-galactopyranosyl-D-sorbitol) were included at the expense of corn starch. Supplementation with oligosaccharides or LAC had little effect on the apparent ileal digestibilities of AA and monosaccharides. Supplementation of diets with oligosaccharides or LAC did not affect (P > 0.05) the monosaccharide concentrations in ileal digesta except for galactose which was higher (P < 0.05) in digesta from pigs fed the diet supplemented with LAC. The daily ileal output of monosaccharides, pH, ammonia and VFA concentrations, bacterial populations in ileal digesta and incidence of diarrhea were not affected (P > 0.05). Therefore, the supplementation of diets for weanling pigs (9.1–13.8 kg) with oligosaccharides or LAC at these levels does not affect nutrient digestibilities and bacterial populations in the small intestine. Specific activity and kinetic constants of trypsin from the pyloric caeca of two strains of rainbow trout, Salmo gairdneri, were measured using α-N-benzoyl-DL-arginine-ρ-nitroaniline∙HCl No increase in activity was observed with cold acclimation, suggesting that cold acclimation induces no increase in trypsin concentration. The apparent Km for the substrate was independent of assay temperature over the physiological range in both strains, probably to maintain high rates of catalysis at higher temperatures when nutrient requirements are high. Strain A trout produced a trypsin with lower affinity on cold acclimation, but Strain B trout did not. The two strains differed in intestinal morphology as well as in the characteristics of their trypsins.

Measurements of soluble proportions in the rumen fluid and duodenal digesta and flow into the small intestine of zinc, manganese, copper and iron were carried out in defaunated and faunated sheep. There was no significant (P > 0.05) effect of the defaunation on any measurements. It was concluded that ciliate protozoa do not have a critical role in trace metal metabolism in the rumen of sheep.
