The effect of dietary level of soybean oil and palm oil on apparent ileal amino acid digestibility and postprandial flow patterns of chromic oxide and amino acids in pigs

A study was conducted to evaluate the apparent ileal digestibilities of amino acids in pigs (initial weight 78 kg) fed diets supplemented with either soybean or palm oil, at either 100 or 200 g kg–1 diet. A control diet with no added fat was also evaluated. All diets contained 5 g chromic oxide kg–1. The pigs were surgically fitted with a simple T cannula at the distal ileum, and were allowed to adapt to each experimental diet for 5 d. This was followed by 1 d of continuous collection of ileal digesta between feedings, and a second day of continuous collection separated into six 2-h postprandial time blocks to determine postprandial flow patterns of chromic oxide and amino acids. The apparent ileal leucine and arginine digestibilities were improved (P < 0.05) by 3.8 and 3.6%, respectively, with 200 g soybean oil kg–1 diet. Linear improvements (P < 0.05) were found in the apparent ileal digestibilities of serine, histidine, arginine, tyrosine, and leucine when 100 and 200 g soybean oil kg–1 diet were supplemented to the control diet, and for serine, histidine, arginine, and phenylalanine with the addition of 100 and 200 g palm oil kg–1 diet. Quadratic responses (P < 0.05) were found in the apparent ileal digestibilities of isoleucine and leucine with the addition of 100 and 200 g palm oil kg–1 diet. The postprandial flow patterns of chromic oxide and amino acids were very stable despite the different dietary treatments. Also, amino acid digestibility coefficients determined at different postprandial sampling times had low variability. In conclusion, the addition of high levels of soybean and palm oil to a semipurified swine diet increased the apparent ileal digestibilities of some amino acids. Also, the two fat sources affected some apparent ileal amino acid digestibility coefficients differently. Key words: Pigs, ileal, amino acid digestibility, fat, postprandial flow patterns

Abstract Three experiments were conducted to determine the effect of three fermented amino acids (AA) with their respective biomass compared to crystalline AA on the growth performance of swine and poultry. In experiment 1, 315 barrows (DNA 200 × 400, initially 11.3 ± 0.69 kg) were allotted to 1 of 4 dietary treatments with 5 pigs per pen and 15 or 16 pens per treatment. Dietary treatments included a negative control (16% standardized ileal digestible [SID] Tryptophan:lysine [Trp:Lys] ratio), positive control (21% SID Trp:Lys ratio from crystalline Trp), or diets containing Trp with biomass to provide 21 or 23.5% SID Trp:Lys ratios, respectively. Pigs fed the positive control or low Trp with biomass diet had increased (P &lt; 0.05) ADG compared to pigs fed the negative control diet, with pigs fed the high Trp with biomass diet intermediate. Pigs fed the low Trp with biomass diet had increased (P &lt; 0.05) G:F compared to the negative control diet, with others intermediate. In experiment 2, 1,320 1-d-old male broilers (Cobb 500, initially 45.2 g) were allotted to one of four dietary treatments with 33 birds per pen and 10 pens per treatment. Dietary treatments included a negative control (58/58% Threonine:lysine [Thr:Lys] ratio), positive control (65/66% Thr:Lys ratio from crystalline Thr), or diets containing Thr with biomass to provide 65/66 or 69/70% Thr:Lys ratios in starter and grower diets, respectively. Broilers fed the positive control or Thr with biomass diets had increased (P &lt; 0.05) ADG compared to broilers fed the negative control diet. Broilers fed the positive control or the low Thr with biomass diet had increased (P &lt; 0.05) G:F compared to the negative control and high Thr with biomass treatments. In experiment 3, 2,100 one-day-old male broilers (Cobb 500, initially 39.4 g) were allotted to one of four dietary treatments with 35 birds per pen and 15 pens per treatment. Dietary treatments included a negative control (59/63% Valine:lysine [Val:Lys] ratio), positive control (75/76% Val:Lys ratio from crystalline Val), or diets containing Val with biomass to provide 75/76 or 84/83% Val:Lys ratios in starter and grower diets, respectively. Broilers fed the positive control or Val with biomass diets had increased (P &lt; 0.05) ADG, ADFI, and G:F compared to those fed the negative control diet. In conclusion, Trp, Thr, or Val with their respective biomass appear to be equally bioavailable and a suitable alternative to crystalline AA in swine and poultry diets.

Abstract Background Recent accumulating evidence suggests that amino acids have crucial roles in the maintenance of intestinal homeostasis. In inflammatory bowel disease (IBD), amino acid metabolism is changed in both host and the gut microbiota. Among amino acids, L-serine plays a central role in several metabolic processes that are essential for the growth and survival of both mammalian and bacterial cells. However, the role of L-serine in intestinal homeostasis and IBD remains incompletely understood. In this study, we investigated the effect of dietary L-serine on intestinal inflammation in a murine model of colitis. Methods Specific pathogen-free (SPF) mice were fed either a control diet (amino acid-based diet) or an L-serine-deficient diet (SDD). Colitis was induced by the treatment of dextran sodium sulfate (DSS). The gut microbiome was analyzed by 16S rRNA sequencing. We also evaluate the effect of dietary L-serine in germ-free mice and gnotobiotic mice that were colonized by a consortium of non-mucolytic bacterial strains or the consortium plus mucolytic bacterial strains. Results We found that the SDD exacerbated experimental colitis in SPF mice. However, the severity of colitis in SDD-fed mice was comparable to control diet-fed mice in germ-free condition, suggesting that the gut microbiota is required for exacerbation of colitis caused by the restriction of dietary L-serine. The gut microbiome analysis revealed that dietary L-serine restriction fosters the blooms of a mucus-degrading bacterium Akkermansia muciniphila and adherent-invasive Escherichia coli in the inflamed gut. Consistent with the expansion of mucolytic bacteria, SDD-fed mice showed a loss of the intestinal mucus layer. Dysfunction of the mucus barrier resulted in increased intestinal permeability, thereby leading to bacterial translocation to the intestinal mucosa, which subsequently increased the severity of colitis. The increased intestinal permeability and subsequent bacterial translocation were observed in SDD-fed gnotobiotic mice that colonized by mucolytic bacteria. In contrast, dietary L-serine restriction did not alter intestinal barrier integrity in gnotobiotic mice that colonized only by non-mucolytic bacteria. Conclusion Our results suggest that dietary L-serine regulates the integrity of the intestinal mucus barrier during inflammation by limiting the expansion of mucus degrading bacteria.

Abstract The objective was to determine the influence of amino acid (AA) supplementation during the adaptation period on the ileal digestibility of crude protein and AA in corn and soybean meal (SBM). Six barrows with an initial body weight of 30.9 ± 2.6 kg fitted with a T-cannula in the distal ileum were assigned to a 6 × 6 Latin square design with 6 dietary treatments and 6 periods. Two experimental diets contained corn or SBM as the sole source of AA and an N-free diet was additionally prepared. For AA supplementation groups, an AA mixture consisted of Gly, Lys, Met, Thr, Trp, Ile, Val, His, and Phe was added to the corn diet and the N-free diet at the expense of cornstarch, and an AA mixture of Lys, Met, and Thr was added to the SBM diet. All diets contained 0.5% of chromic oxide. The 6 experimental diets were fed to the pigs for 4 and half days, and the 3 diets containing AA mixture were switched to the respective diets without AA mixture during the following 2 and half days. Ileal digesta were collected during the last 2 days. The addition of AA mixture during the adaptation period caused increased apparent ileal digestibility of Arg and Trp in corn (P &lt; 0.05), but did not affect that in SBM. The addition of AA mixture during the adaptation period caused increased apparent ileal digestibility of Pro and Gly regardless of feed ingredient (P &lt; 0.05), but did not affect that of other AA. All AA except Pro in corn and SBM were unaffected by the addition of AA mixture during the adaptation period. In conclusion, the addition of amino acid during the adaptation period does not affect the standardized ileal digestibility of indispensable amino acids in feed ingredients.

The regulation of glycerol permeability in the gastrointestinal tract is crucial to control fat deposition, lipolysis and gluconeogenesis. Knowing that the amino acid glutamine is a physiological regulator of gluconeogenesis, whereas cystine promotes adiposity, herein we investigated the effects of dietary supplementation with glutamine and cystine on the serum biochemical parameters of piglets fed on amino acid-enriched diets, as well as on the transcriptional profile of membrane water and glycerol channels aquaporins (AQPs) in the ileum portion of the small intestine and its impact on intestinal permeability. Twenty male piglets with an initial body weight of 8.8 ± 0.89 kg were allocated to four dietary treatments (n = 5) and received, during a four week-period, a basal diet without supplementation (control) or supplemented with 8 kg/ton of glutamine (Gln), cystine (Cys) or the combination of the two amino acids in equal proportions (Gln + Cys). Most biochemical parameters were found improved in piglets fed Gln and Cys diet. mRNA levels of AQP3 were found predominant over the others. Both amino acids, individually or combined, were responsible for a consistent downregulation of AQP1, AQP7 and AQP10, without impacting on water permeability. Conversely, Cys enriched diet upregulated AQP3 enhancing basolateral membranes glycerol permeability and downregulating glycerol kinase (GK) of intestinal cells. Altogether, our data reveal that amino acids dietary supplementation can modulate intestinal AQPs expression and unveil AQP3 as a promising target for adipogenesis regulation.

To evaluate phenolic compounds and whether the combination of a tanniferous fruit and soybean oil could improve the performance, meat characteristics, and fatty acid (FA) profile in lambs, an experiment was conducted over 40 days with twenty creole male lambs (23.71±3.46 kg). The lambs were allotted in a completely randomised design, with factorial arrangement 2 × 2, with the following dietary treatments: (1) control diet, (2) 2%Caesalpinia coriariaground fruit dry matter (DM), (3) 2% soybean oil DM, and (4) 2%Caesalpinia coriariafruit plus 2% soybean oil. The concentration of condensed tannins (CT) inCaesalpinia coriaceawas 21.71 g/kg DM. No interactions were detected (P>0.05) among soybean oil andCaesalpinia coriaria, and there were no differences in daily gain, intake, and feed conversion. Soybean oil reduced (P<0.05) DM digestibility (68.05 versus 59.56%). In fat from thelongissimus thoracis et lumborum(LTL) muscle, only linoleic acid presented differences (P<0.05) between treatments. The combination ofCaesalpinia coriaceafruit and soybean oil did not improve lamb performance at the included levels.

Abstract Geese are extremely adept in utilizing plant-derived roughage within their diet. However, the intestinal microbiome of geese remains limited, especially the dietary effect on microbial diversity. Cassava foliage was widely used in animal feed, but little information is available for geese. In this study, the geese were fed with control diet (CK), experimental diet supplemented with 5% cassava foliage (CF5) or 10% (CF10) for 42 days, respectively. The cecal samples were collected after animals were killed. High-throughput sequencing technology was used to investigate the microbial diversity in the caecum of geese with different dietary supplements. Taxonomic analysis indicated that the predominant phyla were distinct with different dietary treatments. The phyla Firmicutes (51.4%), Bacteroidetes (29.55%) and Proteobacteria (7.90%) were dominant in the CK group, but Bacteroidetes (65.19% and 67.29%,) Firmicutes (18.01% and 17.39%), Proteobacteria (8.72% and 10.18%), Synergistete (2.51% and 1.76%) and Spirochaetes (2.60% and 1.46%) were dominant in CF5 and CF10 groups. The abundance of Firmicutes was negatively correlated with the supplementation of cassava foliage. However, the abundance of Bacteroidetes and Proteobacteria were positively correlated with the supplementation of cassava foliage. Our study also revealed that the microbial communities were significantly different at genus levels. Genes related to nutrient and energy metabolism, immunity and signal transduction pathways were primarily enriched by the microbiome.

Abstract This study was conducted to determine the digestible energy (DE), metabolizable energy (ME), and ileal digestibility of amino acids (AA) of fish protein hydrolysate (FPH), and to evaluate the effects of FPH on the performance of piglets. In Exp. 1, the available energy content of FPH was determined by difference methods: 12 barrows with an initial body weight (BW) of 40.0±2.1kg were randomly assigned into two dietary treatments with six replicates per treatment, the treatments contained a corn basal diet and an experimental diet in which 20% corn was replaced by FPH. The experiment was lasted for 12 days, with 7 days adaptation period followed by 5 days total collection of feces and urine. In Exp. 2, the standardized ileal digestibility (SID) of AA in FPH by the N-free method was determined, 12 barrows (initial BW of 35.3±1.8 kg) with ileal T-cannulas were randomly allotted into two treatments with six replicates per treatment, the dietary treatments consisted of an experimental diet and a N-free diet. The experiment was lasted for 7 days, with 5 days adaptation period followed by 2 days collection of ileal digesta. In Exp. 3, the effect of FPH on the performance of piglets was conducted. 192 piglets (initial BW of 8.10±1.8kg) were randomly allotted into four treatments with six replicates per treatment. The treatments were a control diet with fish meal (FM) and part or all FM was replaced by FPH, got diets containing 2%, 3% or 5% FPH. The experiment lasted for 28 days. Results showed that: In Exp. 1, the DE and ME of FPH were 21.12MJ/kg and 20.28MJ/kg. In Exp. 2, the SID of Lys, Met, Thr and Trp were 79.99%, 87.17%, 68.29%, and 71.53% respectively. In Exp. 3, addition of 3% FPH increased nutrient digestibility and volatile fatty acid content in feces. Addition of 5% FPH increased the average daily feed intake (ADFI), significantly increased (P&lt;0.05) the activities of total antioxidant capacity (T-AOC), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and immunoglobulin A (IgA), while significantly decreased (P&lt;0.05) the average daily gain (ADG). In conclusion, FPH had a high value of digestible energy and ileal digestible essential amino acids, could improve nutrient digestibility, immunity, antioxidant capacity, and intestinal health of piglets, while adverse to nitrogen deposition in piglets.

AbstractThe optimum pattern and limiting order of some essential amino acids for growing-furring blue foxes were assessed from nitrogen (N) retention responses. Total tract digestibility and N balance trials were carried out on 24 weaned blue fox males in an 8 ✕ 5 cyclic change-over experiment. Eight experimental diets were prepared by removing proportionately about 0·4 of each of the amino acids studied – methionine + cystine, lysine, threonine, tryptophan and histidine – successively from the amino acid control diet. The main source of protein in the amino acid control diet was casein and an amino acid mixture was added to bring the calculated crude protein (CP) content up to the level of 170 g/kg dry matter (DM). Low-protein (CP 95·7 g/kg DM) and high-protein (CP 166·6 g/kg DM) diets, the protein proportion of which was casein protein, served as negative and positive control diets, respectively. The reduction in N retention when one amino acid in turn was deleted from the amino acid control diet was calculated, and a regression analysis was made between N retention and relative amino acid intake. Data on the animals’ intake of each limiting amino acid and those on the amino acid control diet were used. The optimum amino acid pattern, expressed relative to lysine = 100, proved to be: methionine + cystine 77, threonine 64, histidine 55 and tryptophan 22. The first-limiting amino acids were methionine + cystine. Blue fox responses (N retention, weight gain) to deletion of methionine + cystine from the diet were very severe and exceeded those to deletion of any other amino acid. Moreover, removing methionine + cystine from the diet significantly impaired the apparent digestibility of organic matter, reducing it to a level even lower than that of the low-protein diet. After methionine + cystine, the next-limiting amino acid in casein-based diets was threonine, followed by histidine and tryptophan. The results show the importance of verifying the sufficiency of dietary methionine + cystine in the practical feeding of blue foxes.

The effect of two levels of fishmeal substitution (50 (FM1) and 150 (FM2) g/kg) of a grass silage control diet (C) on the rumen digestion of organic matter and nitrogen, and the small intestinal disappearance of amino acids was examined in young growing cattle each equipped with simple PVC cannulas in the dorsal sac of the reticulo-rumen, the proximal duodenum and the terminal ileum. The silage was a primary growth of perennial ryegrass (Lnlium pevenne) (+formic acid) with a total N content of 22 g/kg dry matter (DM) (diet C). Fishmeal substitution increased this to 26 (diet FM1) and 34 (diet FM2) g/kg DM. On diets C and FM1, approximately 0.71 of digestible organic matter intake was apparently digested in the rumen, but this was significantly (P < 0.05) reduced on diet FM2 (0.60). Whilst duodenal flows of non-ammonia N and total amino acids were significantly (P < 001) increased at the highest level of fishmeal inclusion only, the synthesis of microbial N was significantly (P < 0.001) reduced by fishmeal inclusion, and feed N degradability declined progressively in response to increased fishmeal. Both levels of fishmeal addition caused a significant (P< 0.05) reduction in the fractional outflow rate of water from the rumen, and on the highest level of fishmeal significant (P< 0.05) increases in rumen ammonia concentration and rumen propionate molar proportions were observed. The net effect of the highest level of fishmeal substitution was to increase amino acid absorption from the small intestine by 0.47 compared with the control diet (P< 0.05), but due to an elevated ileal flow of amino acid no such effect was detected at the lowest level of fishmeal substitution. Composition of the absorbed amino acid fraction was relatively unaffected by the treatments imposed, despite large changes in the composition of the duodenal protein. The apparent non-linearity of response to fishmeal substitution is discussed and the amino acid supply findings are compared with the protein retention findings obtained in an earlier study by Gill et al. (1987). By two methods of calculation it was estimated that the amino acid N fraction disappearing from the small intestine was utilized with an efficiency of between 0.51 and 0.53 and no apparent effects due to diet or level of amino acid supply were detected.

Abstract Objectives We previously reported that the grape pomace had a protective role against colorectal cancer development. This study is to assess the role of fecal metabolites in mediating the observed beneficial effects of grape pomace. Methods Nine-week-old female mice were fed a control diet (CON) or CON with 5% grape pomace for 2 weeks when mice were subjected to azoxymethane (AOM)/dextran sulfate sodium (DSS) for colorectal cancer (CRC) induction. Fecal samples collected at the end of dietary treatment were used to profile the fecal metabolomic changes in response to grape pomace supplementation using gas chromatography-mass spectrometry (GC-MS) based metabolomic analysis. Results Dietary grape pomace supplementation resulted in significant changes of a total of 29 compounds, which included bile acids, amino acids, fatty acids, phenols/flavonoids, glycerolipids, carbohydrates, organic acids, and others. These differential compounds were enriched in several pathways including alpha-linolenic acid and linoleic acid metabolism, phenylacetate metabolism, malate-aspartate shuttle, tyrosine metabolism, taurine, and hypotaurine metabolism, and others. The major changes of metabolites in feces include increased deoxycholic acid and decreased L-Aspartic acid, L-Proline, and Valine amino acid contents, all of which have strong associations with intestine inflammation and carcinogenesis. Urease hydrolyzes urea to produce ammonia, which increases amino acid concentrations in the gut lumen. Consistently, the urease activity of fecal samples was significantly reduced in the grape pomace supplemented group compared to the control group. Conclusions The increase of fecal deoxycholic acid and the reduction of urease and amino acids might be key factors contributing to the beneficial effects of grape pomace supplementation against CRC development.
