The present study evaluated the effect of vitamin C on alteration in thyroid hormones induced by short-term acute heat stress. Eight male lambs were divided into two groups of 4 animals each. Both groups were placed in an environment with hyper-acute heat stress based on the temperature – humidity index (THI). Groups I and II were injected intramuscularly normal saline and vitamin C (20 mg/kg), respectively, for the first five consecutive days of the experiment. All lambs were fed ad libitum. Blood samples were collected from both groups on days one, two, four, six and eight. Thyroxine and free thyroxine numerically increased (91.03 vs. 70.78 nmol L-1, P=0.080 and 29.8 vs. 24.8 pmol L-1, P=0.080; respectively) in heat stressed lambs supplemented with vitamin C compared to control group. Respiration rates and heart rates were elevated until day five of the experiment and then decreased. Mechanism for increasing the levels of thyroxine and free thyroxine by vitamin C is not well known. However, it may occur in part because of vitamin C antioxidant properties. The present study revealed that vitamin C might ameliorate the adverse effect of heat stress in lambs.

The present study evaluated the ameliorative effect of vitamin C on alteration in thyroid hormones induced by low-dose subchronic coadministration of chlorpyrifos (CPF) and lead (Pb). Forty Wistar rats were divided into 4 groups of 10 animals each. Groups I and II were administered soya oil (2 mL/kg) and vitamin C (100 mg/kg), respectively. Group III was coadministered CPF (4.25 mg/kg ~1/20th LD50) and Pb (250 mg/kg ~1/20th LD50), respectively. Group IV was pretreated with vitamin C (100 mg/kg) and then coadministered with CPF (4.25 mg/kg) and Pb (250 mg/kg), 30 min later. The regimens were administered by gavage for a period of 9 weeks. The marginal decrease in serum triiodothyronine and thyroxine and the significant increase in the concentrations of thyroid stimulating hormone and malonaldehyde in the group coadministered with CPF and Pb were ameliorated by vitamin C partly due to its antioxidant properties.

AbstractThyroid hormone reference intervals are crucial for diagnosing and monitoring thyroid dysfunction during early pregnancy, and the dynamic change trend of thyroid hormones during pregnancy can assist clinicians to assess the thyroid function of pregnant women. This study aims to establish early pregnancy related thyroid hormones models and reference intervals for pregnant women. We established two derived databases: derived database* and derived database#. Reference individuals in database* were used to establish gestational age-specific reference intervals for thyroid hormones and early pregnancy related thyroid hormones models for pregnant women. Individuals in database# were apparently healthy non-pregnant women. The thyroid hormones levels of individuals in database# were compared with that of individuals in database* using nonparametric methods and the comparative confidence interval method. The differences in thyroid stimulating hormone and free thyroxine between early pregnant and non-pregnant women were statistically significant (p<0.0001). The reference intervals of thyroid stimulating hormone, free thyroxine and free triiodothyronine for early pregnant women were 0.052–3.393 μIU/ml, 1.01–1.54 ng/dl, and 2.51–3.66 pg/ml, respectively. Results concerning thyroid stimulating hormone and free thyroxine reference intervals of early pregnancy are comparable with those from other studies using the same detection platform. Early pregnancy related thyroid hormones models showed various change patterns with gestational age for thyroid hormones. Early pregnancy related thyroid hormones models and reference intervals for pregnant women were established, so as to provide accurate and reliable reference basis for the diagnosing and monitoring of maternal thyroid disfunction in early pregnancy.

This study aims to characterize the influence of short-term heat stress (HS; 4 day) in early lactating Holstein dairy cows, in terms of triggering blood metabolomics and parameters, milk yield and composition, and milk microRNA expression. Eight cows (milk yield = 30 ± 1.5 kg/day, parity = 1.09 ± 0.05) were homogeneously housed in environmentally controlled chambers, assigned into two groups with respect to the temperature humidity index (THI) at two distinct levels: approximately ~71 (low-temperature, low-humidity; LTLH) and ~86 (high-temperature, high-humidity; HTHH). Average feed intake (FI) dropped about 10 kg in the HTHH group, compared with the LTLH group (p = 0.001), whereas water intake was only numerically higher (p = 0.183) in the HTHH group than in the LTLH group. Physiological parameters, including rectal temperature (p = 0.001) and heart rate (p = 0.038), were significantly higher in the HTHH group than in the LTLH group. Plasma cortisol and haptoglobin were higher (p < 0.05) in the HTHH group, compared to the LTLH group. Milk yield, milk fat yield, 3.5% fat-corrected milk (FCM), and energy-corrected milk (ECM) were lower (p < 0.05) in the HTHH group than in the LTLH group. Higher relative expression of milk miRNA-216 was observed in the HTHH group (p < 0.05). Valine, isoleucine, methionine, phenylalanine, tyrosine, tryptophan, lactic acid, 3-phenylpropionic acid, 1,5-anhydro-D-sorbitol, myo-inositol, and urea were decreased (p < 0.05). These results suggest that early lactating cows are more vulnerable to short-term (4 day) high THI levels—that is, HTHH conditions—compared with LTLH, considering the enormous negative effects observed in measured blood metabolomics and parameters, milk yield and compositions, and milk miRNA-216 expression.

Abstract Phytogenic feed additives that contain anti-inflammatory and antioxidant properties may have potential to reduce inflammation and oxidation observed during bouts of heat stress. The trial objective was to determine the effects of a phytogenic feed additive on markers of stress in heifers subjected to diurnal heat stress (dHS). Eighteen Holstein and four Jersey heifers (5–6 months of age; d1 BW=205±9 kg) were assigned to two dietary treatments (n = 11) based on breed and age; 1) a basal TMR diet (CTL), 2) CTL supplemented with 0.25 g/head/d of a proprietary blend of phytogenics (PFA; BIOMIN America, Inc., Overland Park, KS). Heifers were housed individually and fed their respective treatments for 14d prior to dHS (d-14-1), subjected dHS (d1-7), and followed through a recovery period (d8-11). Weights were collected at d1 (baseline), d7 (end of dHS), and d11 (end of recovery). The temperature set point was assigned at 33°C from 0900 to 2100 h and allowed to equilibrate with outside temperature during night (24°C) for a total of 7d dHS. Total DMI (tDMI) and DMI from 0900 to 2100 h (hsDMI) were recorded daily. Serum was collected at d1, 2, 7 and 11 to determine effect of treatment on markers of oxidation, inflammation, and stress. Treatment effects were analyzed using PROC MIXED in SAS with treatment and time as fixed effects and heifer as a random effect. Temperature humidity index during dHS was greater during the day (84.5) versus night (73.3). There were no treatment differences (P&gt;0.05) in d 0–11 on ADG, ADFI, or GF between treatments. However, PFA numerically increased hsDMI, reduced (P&lt; 0.05) haptoglobin and tended to reduce (P&lt; 0.10) lipopolysaccharide binding protein compared with CTL. Overall, dietary PFA may be a potential strategy to mitigate the stresses that heifers experience during dHS.

The systolic blood pressures of two groups of rats that were exposed to cold (5 °C) for 4 weeks were elevated significantly above that of warm-acclimated controls maintained at 24 °C. At this time these groups were given the antithyroid drug aminotriazole in their food at 0.3 g/kg. At the same time, one group was given 15.8 μg thyroxine (T4)/kg body mass per day, while the second received 31.6. The doses were chosen as replacement (15.8 μg/kg) and twice replacement (31.8 μg/kg) for the rats. The results of the study revealed that both groups receiving aminotriazole and T4 had reductions in blood pressure within 1 week of initiation of treatment. Blood pressures reached control level after 5 weeks. Cardiac hypertrophy accompanying cold-induced hypertension was reduced with the lower dose of T4 and prevented with the higher dose. Serum concentrations of T4 and triiodothyronine (T3) in the two treated groups were reduced, while serum thyroid-stimulating hormone concentration and thyroid mass were increased above that of the warm-acclimated control group. This suggests that the rats were hypothyroid relative to the warm-acclimated control group. However, the treated rats grew at the same rate as nontreated, cold-exposed controls and had similar food and water intakes, a similar dipsogenic response to acute administration of isoproterenol, and similar colonic temperatures. These measurements suggest that the rats were not functionally hypothyroid. Nevertheless, the results suggest that a paradigm in which the secretory ability of the thyroid gland is blocked, and T4 is returned at a constant, albeit suboptimal, level, reduced blood pressure and cardiac hypertrophy in cold-exposed rats. Hence, the increased turnover of thyroid hormones that characteristically accompanies exposure to cold plays a role in these changes. This study was conducted to determine the effects of dietary vitamin C (L-ascorbic acid) and vitamin E (a-tocopherol acetate) on lipid peroxidation status measured as MDA and serum triiodothyronine (T3), thyroxine (T4), thyroid-stimulating hormone (TSH), adrenocorticotropic hormone (ACTH), as well as some other serum metabolite and mineral concentrations of Japanese quails reared under heat stress (34º C). One hundred-eighty 10-day-old Japanese quails were randomly assigned to six treatment groups, three replicates of 10 birds each. Using a 2 × 3 factorial design, the birds received two levels of vitamin C (100 and 200 mg/kg of diet) or three levels of vitamin E (125, 250, or 500 mg/kg of diet). Greater dietary vitamin E and vitamin C resulted in a greater serum T3, T4, and TSH (p=0.001), but lower ACTH (p=0.001) concentrations. Serum concentrations of T4 and TSH increased to a greater extent by increasing dietary vitamin C when greater vitamin E levels were fed (interaction, p=0.001). Serum glucose, urea, triglycerides, and cholesterol concentrations decreased (p=0.001), while protein and albumin concentrations increased (p =0.001) when both dietary vitamin C and vitamin E were increased. Serum activities of SGOT and SGPT were not influenced by dietary vitamin C or vitamin E (p>0.43). However, serum activity of AP increased (p=0.001) by increasing both dietary vitamin C and vitamin E. Increasing both dietary vitamin C and vitamin E caused an increase in serum concentrations of Ca, P, K (p=0.001), Fe, and Zn (p=0.01) but a decrease in serum concentrations of Na (p=0.001) and Cu (p=0.01). Interactions between vitamin C and vitamin E were detected for Ca, P, Na, and K (p =0.001). Greater dietary vitamin C and vitamin E resulted in a greater serum and liver vitamin E, C, and A (p_0.05), but lower MDA (p=0.001) concentrations. Results of the present study conclude that supplementing a combination of dietary vitamin C (200 mg) and vitamin E (250–500 mg) offers a good management practice to reduce heat stress-related decreases in performance of Japanese quails.

Purpose:Prolonged physical exertion and environmental heat stress may elicit postexercise depression of immune cell function, increasing upper respiratory tract infection (URTI) susceptibility. We investigated the effects of acute and short-term vitamin C (VC) compared with placebo (PL) supplementation on URTI susceptibility, salivary immunoglobulin A (s-IgA), and cortisol responses in healthy individuals following prolonged exercise-heat stress.Methods:Twelve participants were randomized into the VC or PL group in a double-blind design. For 12 days, participants consumed 3 × 500 mg tablets of VC or PL per day, with testing completed at baseline, then following acute (1 d) and short-term (8 d) supplementation. Participants performed 120.1 ± 49.6 min of cycling at 54 ± 6% VO2max in a hot (34.8 ± 1.0°C and 13 ± 3% relative humidity) environment, with saliva samples collected at pre-, post-, and 72 h postexercise. Health logs specifying URTI symptoms were completed for 7 days postexercise.Results:A 2 × 3 × 3 mixed ANOVA with a post hoc Bonferroni correction factor revealed a significant linear trend in postexercise cortisol attenuation in the VC group, 21.7 ± 15.1 nmol/L (mean ± SD) at baseline, to 13.5 ± 10.0 at acute, to 7.6 ± 4.2 after short term (P = .032). No differences were detected in ratio of s-IgA to protein or URTI symptoms between groups.Conclusions:These data suggest that vitamin C supplementation can decrease postexercise cortisol in individuals performing exercise similar to that of a half-marathon or marathon in hot conditions. However, no changes in s-IgA and URTI were evident, possibly due to previous moderate training and reduced physical and psychological stress compared with athletes participating in ultramarathons.

ABSTRACTObjective: Apart from its well-known deleterious dental and skeletal effects, fluoride excess can have toxic effects on many other tissues. Fluoride, when in excess, is known to interfere with thyroid gland function. Fluoride-induced thyroid disturbances similar to those observed in iodine deficiency state in spite of adequate iodine intake have been documented. Similar thyroid disturbances in individuals with dental fluorosis have not been well studied in populations with endemic fluorosis. This work was undertaken to study the effects of fluoride-induced thyroid disturbances in individuals with dental fluorosis.Methods: The study group included 65 subjects with dental fluorosis from endemic fluorosis populations. An additional control group was comprised of 10 subjects without dental fluorosis. The drinking water fluoride levels of the study populations were analyzed. Serum free FT3, FT4, and TSH levels of both groups were assessed.Results: All subjects with dental fluorosis had serum levels of thyroid hormones (FT3, FT4, and TSH) within the normal range, with the exception of 1 individual, who had elevated levels of TSH. Statistical significance was found when FT3 and TSH values were compared with different Dean’s index groups by a 1-way ANOVA test: FT3 (F = 3.4572; P=.0377) and TSH (F = 3.2649 and P=.0449).Conclusions: Findings of this study did not show any significant alterations in the levels of the thyroid hormones FT3, FT4, and TSH in subjects with dental fluorosis. Our observations suggest that thyroid hormone levels were not altered in subjects with dental fluorosis. Hence, future studies of this kind, along with more detailed investigations are needed. Abstract Background: Accurate measurement of free thyroid hormones is important for managing thyroid disorders. Ultrafiltration liquid chromatography tandem mass spectrometry (LC-MS/MS) can reliably measure the concentrations of small molecules, including thyroid hormones. Our study was designed to compare free thyroid hormone measurements performed with immunoassay and LC-MS/MS. Methods: We studied the performance of LC-MS/MS in 4 different populations comprising pediatric patients, euthyroid adults, and healthy nonpregnant and pregnant women. The samples obtained from each population numbered 38, 200, 28, and 128, respectively. Free thyroxine, free triiodothyronine, and thyroid-stimulating hormone (TSH) concentrations were documented. Results: LC-MS/MS measurement of free thyroid hormones provided better correlation with log-transformed serum TSH in each population and also the populations combined. The correlations between free thyroxine measured by LC-MS/MS and log TSH in the pediatric outpatients and healthy adults were −0.90 and −0.77, respectively. The correlations for immunoassay were −0.82 and −0.48. The correlations between free triiodothyronine measured by LC-MS/MS and TSH for both pediatric and healthy adult populations were −0.72 and −0.68, respectively. Conclusions: Free thyroid hormone concentrations measured by LC-MS/MS correlate to a greater degree with log TSH values compared to concentrations measured by immunoassay. This correlation was maintained across the patient populations we studied and may reflect the accuracy and specificity of LC-MS/MS. The superior ability of LC-MS/MS to enable documentation of the well-known thyroid hormone–TSH relationship supports the use of this measurement technique in a variety of clinical situations.
