Hubbard has found that the photoisomerization of retinene was important for the regeneration of rhodopsin in vitro, and the object of the present investigation was to find whether this was also true for regeneration in the living human eye. In the Appendix is described a device which permits the rhodopsin density to be measured by analysing the light reflected from the fundus oculi in an ophthalmoscopic arrangement, the measurement taking about 5 seconds. Now if a blue and a yellow light viewed scotopically are adjusted in intensity so as to appear identical, they must bleach rhodopsin equally, but the blue will be more than 10 times as effective in isomerizing retinene. Therefore if retinene isomerization is important for rhodopsin regeneration, blue light should cause a more rapid regeneration after bleaching, and during bleaching the equilibrium level attained should be less profound. But, as the figures show, the course of bleaching and regeneration is identical for the matched yellow or blue bleaching lights, therefore isomerization of retinene is not important for rhodopsin regeneration in the living human eye.

The experiment was designed to evaluate the effect of growth regulators on leaf explant of Gerbera for callus induction. Various kinds of plant growth regulators such as 6-Benzylaminopurine (BAP), α-Naphthalene acetic acid (NAA), 2, 4-Dichlorophenoxyacetic acid (2, 4-D), Indole-3-acetic acid (IAA) were used to initiate cultures. These were added to Murashige and Skoog medium in different combinations and concentrations. Leaf explants cultured on MS medium supplemented with BAP+ 2, 4-D+ IAA in T4 treatment & BAP+ 2,4-D in T5 treatment showed the best results for callus induction. On the other hand callus was induced early in the combination of BA+ 2,4-D + IAA hormone in T5, T9 & T8 treatment respectively. The rate of callus induction was very low in BA + NAA combinations but it was much earlier.

Cucumber green mottle mosaic virus (CGMMV), a member of the genus Tobamovirus (family Virgaviridae), is an economically important virus that has detrimental effects on cucurbit crops worldwide. Understanding the interaction between host factors and CGMMV viral proteins will facilitate the design of new strategies for disease control. In this study, a yeast two-hybrid assay revealed that the CGMMV helicase (HEL) domain interacts with a Citrullus lanatus small heat shock protein (sHSP), and we verified this observation by performing in vitro GST pull-down and in vivo coimmunoprecipitation assays. Measurement of the levels of accumulated sHSP transcript revealed that sHSP is upregulated on initial CGMMV infection in both Nicotiana benthamiana and C. lanatus plants, although not in the systemically infected leaves. We also found that the subcellular localization of the sHSP was altered after CGMMV infection. To further validate the role of sHSP in CGMMV infection, we produced and assayed N. benthamiana transgenic plants with up- and down-regulated sHSP expression. Overexpression of sHSP inhibited viral RNA accumulation and retarded disease development, whereas sHSP silencing had no marked effect on CGMMV infection. Therefore, we postulate that the identified sHSP may be one of the factors modulating host defense mechanisms in response to CGMMV infection and that the HEL domain interaction may inhibit this sHSP function to promote viral infection.

ABSTRACT The aim of this study was to identify parents and promising hybrid combinations for the improvement of forage watermelon. Five parents were evaluated: BGCIA 996 (1), BGCIA 997 (2), BGCIA 998 (3), BGCIA 228 (4), Jojoba (5) and ten F1 hybrids, which were obtained from balanced diallel crosses. The experimental design was in a complete randomized block, with three replications. The morphoagronomic and bromatological traits were evaluated. The highlights were the progenitors BGCIA 997, BGCIA 998, BGCIA 228 and Jojoba for protein content, fruit yield, in vitro digestibility of dry matter and number of seeds, respectively. The hybrid 1x4 stood out for fruit length, seed number, and ethereal extract. The hybrid 2x3 stood out for rind and pulp thickness while the hybrids 3x4, 3x5 and 4x5 had exceptional digestibility, protein content and fruit yield, respectively. The analysis of the standard deviation of the SCA estimates of both SD (Sij- Sik) and SD (Sij- Skl) indicated that no hybrids were found that had SCA estimates twice that of SD (Sij- Sik) or SD (Sij- Skl), except for the hybrid 1x4 for the number of seeds per fruit. These results suggest that the parents were more promising than the hybrids. Similarly, the GCA estimates favor the intrapopulation method, which will promote greater efficiency in selection for genetic gains.

The first experiment involving different explants and concentrations of 2,4-D and kinetin showed highly significant differences for length and weight of callus formed except interaction of callus weight. Leaf explant appeared to be best of all for callus length and weight when 1.0 mg/L 2,4-D + 0.25 mg/L kinetin concentration was used. Similarly, different explants versus different concentrations of BAP/GA3/IAA showed significant differences for shoot length and leaf number per plantlet and also for root length. However, interaction term confirmed node and node/internode explants produced better results in shoot length and number of leaves per plantlet when concentrations 1 .0 mg/L BAP + 0.1 mg/L GA3 and 1.0 mg/L BAP + 0.2 mg/L GA3, 1.0 mg/L BAP + 0.4 mg/L GA3, respectively, were used. Similarly, internode explants produced better results for root length after 21 days plantlet-1 when concentration of 1.0 mg/L IAA + 0.25 mg/L GA3 was used. Shoot tip explants also produced better results in root length after 28 days plantlet-1 when concentrations 1.0 mg/L IAA + 0.25 mg/L GA3 were used. A standard protocol was established for rapid in vitro propagation of watermelon (Citrullus lanatus Thumb.) from nodal explants of field grown plant. Multiple shoot proliferation was achieved from nodal explants on MS medium supplemented with 1.0 mg/l BAP + 0.2 mg/l NAA within 30 days of inoculation. The elongation of shoots was obtained on the same medium. Highest percentage of root induction was achieved on MS medium supplement with 1.0 mg/l IBA within 25 days of culture. Well rooted plantlets were transferred to small pots and after proper acclimatization the plantlets were transplanted in the field condition, where 80% plantlets were survived and grew successfully.
