There is an opinion in professional literature that edge-strands in β-sheet are critical to the processes of amyloid transformation. Propagation of fibrillar forms mainly takes place on the basis of β-sheet type interactions. In many proteins, the edge strands represent only a partially matched form to the β-sheet. Therefore, the edge-strand takes slightly distorted forms. The assessment of the level of arrangement can be carried out based on studying the secondary structure as well as the structure of the hydrophobic core. For this purpose, a fuzzy oil drop model was used to determine the contribution of each fragment with a specific secondary structure to the construction of the system being the effect of a certain synergy, which results in the construction of a hydrophobic core. Studying the participation of β-sheets edge fragments in the hydrophobic core construction is the subject of the current analysis. Statuses of these edge fragments in β-sheets in ferredoxin-like folds are treated as factors that disturb the symmetry of the system.

<p>Prion is a protein smaller than virus and it infects host in the absence of nucleic acid. The secondary structure of protein folds incorrectly from α-helices to β-sheets through breaking and re-formation of hydrogen bond. Structural analogy of α-helix and DNA double helix and comparing differences between α-helix and β-sheet show prion's infectivity and propagation. Aggregates of dimers and polymers generate β-amyloid fibril in Alzheimer's disease.</p>

AbstractThe fuzzy oil drop model was applied to characterize the hydrophobic core structure in plant carboxylesterase. The characteristics revealed the status of β-sheets in the central part of the molecule as discordant as opposed to the expected hydrophobicity distribution. Particularly, the β-strands and helices in close proximity to the enzymatic residues recognized as discordant with respect to the ideal hydrophobicity distribution of hydrophobic core are of high importance. It is assumed that this local irregularity is the form of coding the specificity of enzymes. The protein under consideration appears to be the next example proving this assumption.

The roles of hydrophobic bonding and charge electrostatics in the stabilization of human β2-microglobulin have been probed by variations in solution conditions and monitored by circular dichroism in the near and far UV regions. Sodium perchlorate initially gives a decrease in intensity of the positive 234 nm peak in the near UV followed by a shift of this peak to negative ellipticity at high perchlorate concentration. These 234 nm changes indicate a new environment for a tyrosyl chromophore(s). A conformational rearrangement of the β-sheet sandwich must occur since all six tyrosines of human β2-microglobulin are located in the two β-sheets of this sandwich. A slight decrease in intensity for the 200 nm positive peak in the far UV indicates a less close packing of the β-sheets at high perchlorate. In other experiments, enthalpy and entropy values have been calculated from thermal unfolding studies at 50 and 180 mM NaCl for pH values 6.0 and 8.0. Larger enthalpy values are obtained at higher NaCl concentration consistent with salt shielding of predominantly unfavorable charge interactions. These enthalpy differences are relatively large suggesting that charge electrostatics are energetically significant in stabilization of human β2-microglobulin.

Abstract The T-knot scaffold, a disulphide-reinforced structural motif shared by several proteins with very different biological functions, has been defined as ‘a stretch of the protein chain which comprises two strands of a β-sheet and three loops, knotted by two disulphides into the shape of the letter T’. In this communication we show that the presence of a central β-sheet is not a required structural feature for proteins sharing the T-knot topology. Moreover, superposition of the three-dimensional structures of representative members of the T-knot family highlights a common and structurally well-defined core, formed by the two knotted disulphides, substituting for a larger residue-based hydrophobic core. These results suggest that folding and stability of the T-knot scaffold mainly depend on the geometry of the two knotted disulphides and on the loop length, and that the secondary structure elements are not a prerequisite for motif formation. Accordingly, a redefinition of the T-knot motif is proposed.

AbstractSequence-to-structure relation is one of the major objects of the analysis of protein folding problem. The pair of two small proteins (domains) of similar structure (β-hairpin/α-helix/β-hairpin) generated by the chains of similar length (about 60 amino acids) with very low sequence similarity (15%) is the object of the comparable analysis of 3D structure. The criterion for similarity estimation is the status of polypeptide chain with respect to the hydrophobic core structure. The fuzzy oil drop model is applied to reveal the differentiated status of fragments of the well-defined secondary structure. This analysis allows the interpretation of the structure in other than the geometric form as it is made based on secondary structure classification. The two compared highly similar proteins appear to be different with respect to the hydrophobic core structure.

Purpose Academia’s overwhelming reliance on non-tenure track, or contingent, faculty is a well-known fact. While the status and working conditions of contingent classroom faculty have been well studied and documented, the corresponding trend in academic libraries has not been explored as deeply. As this paper reviews the limited LIS literature on the subject, the purpose of this paper is to provide administrators and managers with a deeper understanding of the benefits and drawbacks of contingent appointments. It also offers strategies for fostering a workplace culture that recognizes contingent librarians’ contributions and promotes their professional growth. Design/methodology/approach An overview of scholarly and professional literature on contingent librarianship, this paper is based on published research studies and academic articles; given the prominence of anecdotal and personal writing on the subject, columns and first-person essays from trade publications, as well as library-related blogs and job search sites, are also included. Findings Contingent librarians have been a steady presence in academic libraries for the last few decades. The trend is continuing. There are specific practices that can be applied to effectively manage contingent librarians. Originality/value The paper offers academic library administrators and managers a concise yet comprehensive overview of the issues related to contingent appointments.

Purpose. The purpose of the work is to analyze the application of the second and third parts of Article 3692 of the Criminal Code of Ukraine to officials in the context, that defines them by the Criminal Code of Ukraine in the note to Article 364 of the Criminal Code of Ukraine. Methodology. The methodology includes a comprehensive analysis and synthesis of the available scientific and theoretical material and the formulation of relevant conclusions and recommendations. In the course of the study, the following methods of scientific knowledge were used: terminological, logical-semantic, system-structural, logical-normative. Results: in the course of research the cause of criminalization of such act as "abuse of power" is considered, the subject of the specified criminal act which has the features of "an official" in the context, that defines it by the note to Article 364 of the Criminal Code of Ukraine is analyzed and the main ways of committing criminal acts, that are provided for in this article of the Criminal Code of Ukraine are identified. Originality. The study found, that one of the key conditions for the opportunity to influence officials, that are authorized to perform government or local self-government functions, is the position held by the official and the related opportunities. Therefore, taking into account the opinion of the scientists, that the subject of crimes, that are provided for by the second and third parts of Article 3692 is special, the peculiarities of which is the cumulative feature, that denotes, that such person is not endowed with the status of an official, well-founded need to specify the criminal legislation of Ukraine in terms of the application the second and third parts of Article 3692 of the Criminal code of Ukraine concerning officials in the context, that defines them by the criminal legislation of Ukraine in the note to Article 364 of the Criminal Code of Ukraine. Practical significance. The research results can be used in lawmaking in the improvement of anti-corruption legislation.
