Microorganisms are regarded as a sustainable source of biologically active molecules. Among them, Trichoderma spp. have been an attractive source of biological compounds. However, the study of marine-derived Trichoderma has developed slowly because of the difficulty in isolating the fungi. In our study, 30 strains of marine-derived Trichoderma were identified through the translation elongation factor 1-alpha (EF1α) sequences, and their biological activities, such as antioxidant activity by ABTS and DPPH assays, antifungal activity against Asteromyces cruciatus and Lindra thalassiae, and tyrosinase inhibition activity, were investigated. As a result, the 30 marine Trichoderma species were classified into 21 taxa, including three new species candidates. Three strains of T. asperellum showed the highest ABTS radical scavenging activity and antifungal activity. T. bissettii SFC20170821-M05 and T. guizhouense SFC20180619-M23 showed notable DPPH radical scavenging activity and tyrosinase inhibition activity, respectively. This study showed the potential of marine-derived Trichoderma as a source of bioactive compounds.

Introduction: Kacangma (Leonurus sibiricus), turmeric (Curcuma longa) and black pepper (Piper nigrum) are the oldest and most important spice crops used for culinary purposes in Malaysia for centuries. Although many biological activities including antioxidant properties (AP) have been reported, there are no confirmed reports on the activities of this cocktail, nutri-Pepper enhancer. Purpose: The study evaluated the AP of the Nutri-Pepper enhancer in vitro. Methodology: The AP were evaluated using in vitro antioxidant assays where the total polyphenolic content (TPC), DPPH radical scavenging activity, tyrosinase inhibition activity, xanthine oxidase superoxide scavenging activity (XOD), and oxygen radical absorbance capacity (ORAC) were measured.  Results: Based on the antioxidant activity range, extract of Nutri pepper enhancer was high in tyrosinase inhibition activity, 87.16%, moderate in xanthine oxidase superoxide scavenging activity (XOD) 69.72% and low in 1,2-diphenyl-2- picrylhydrazyl radical scavenging activity (DPPH) 46.78%. On the other hand, the extract showed high ORAC and total phenolic content with the value of 236,00 µmol TE/100g and 282 mg/100 GAE. Conclusion: This finding proven that the consumption of this product can reduce or even eliminate the harmful effects on humans from contaminants in foods and from the environment; this is even more important for people living in polluted cities.

Aerva javanica (Burm. f) Juss. ex Schult. (Family: Amaranthaceae) family is one of the traditional medicinal plant growing in the United Arab Emirates. Apart from studies related to some medicinal properties, phytochemical, GC MS compound characterization and biological activities still to be investigated. An experiment was conducted to determine the possible bioactive components with their chemical structures and elucidation of phytochemicals from the aerial parts of the plant. The macro and micro-mineral constituents and antioxidant activities were also evaluated. Aerial parts of A. javanica were extracted sequentially with hexane, chloroform, ethyl acetate, acetone, methanol by cold percolation method. Free radical scavenging and antioxidant properties of methanolic extract were evaluated by using in vitro antioxidant assays such as hydroxyl radical scavenging activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, superoxide radical scavenging activity and ABTS radical scavenging activity. Primary phytochemical and micro-macro element was tested using standard protocol. The chemical characterization was done with the help of Gas Chromatography-Mass Spectrometry (GC–MS), and the mass spectra of the total compounds in the extract were matched with the National Institute of Standard and Technology (NIST) library. Mineral constituents were identified and estimated by ICP-OES. Ninety-nine metabolites were obtained by GC-MS anslysis; indole was found to be major components followed by 2-Chlorallyl diethyldithiocarbamate (CDEC), Carbaril, Bis(2-ethylhexyl) phthalate, Quinoline, 4H-Cyclopenta[def]phenanthrene,2-[Bis(2-chloroethylamino)]-tetrahydro-2H-1,3,2-oxazaphosphorine-2-oxide, Phenobarbital, 1H-Indole, 2-methyl-, 2,3,7,8-Tetrachlorodibenzo-p-dioxin Disulfide, diphenyl. The presence of various bioactive compounds in the extract validates the traditional medicinal uses of this plant.

A series of 20 new biologically active derivatives of 2-{4, 5-(substituted diphenyl)-4H-1,2,4-triazol-3-ylthio}acetyl chloride has been synthesized, with the aim to investigate antimicrobial, free radical scavenging activity. All the synthesized compounds were characterized by spectroscopic data and elemental analysis. The final compounds were tested for antibacterial activity against Gram-positive bacteria: Staphylococcus aureus MTCC 3160, Bacillus subtilis MTCC 441; Gram-negative bacteria: Escherichia coli MTCC 443, and, for antifungal activity, against Candida albicans MTCC 227 and Aspergillus niger MTCC 281, taking ciprofloxacin as antibacterial and fluconazole as antifungal standard drugs. Compound 7a6 was found to be the most effective antibacterial (MIC = 3.12 µg/ml), and compounds 7a2 and 7d1 (MIC = 3.12 and 6.25 µg/ml) had the most effective antifungal effects on the selected strains, as compared to the standard drugs. The results of antioxidant studies revealed that compound 7b1 was found to be most active antioxidant, with 40.4±0.687 µg/ml, and compounds 7b3, 7d7, and 7d4 also showed promising free radical scavenging activity, as compared with the standard drug ascorbic acid.

A series of 1,3,5-triazine derivatives, incorporating aminobenzenesulfonamide, aminoalcohol/phenol, piperazine, chalcone, or stilbene structural motifs, were evaluated as potential antioxidants. The compounds were prepared by using step by step nucleophilic substitution of chlorine atoms in starting 2,4,6-trichloro-1,3,5-triazine. Reactions were catalyzed by Cu(I)-supported on a weakly acidic resin. The radical scavenging activity was determined in terms of %inhibition activity and IC50, using the ABTS method. Trolox and ascorbic acid (ASA) were used as standards. In the lowest used concentration 1 × 10−4 M, the %inhibition activity at time 0 min was comparable with both standards at least for 10 compounds. After 60 min compounds 1, 2, 9 and 25 showed nearly twice %inhibition (73.44–87.09%) in comparison with standards (Trolox = 41.49%; ASA = 31.07%). Values of IC50 correlated with %inhibition activity. For compounds 1, 2, 9 and 25 values of IC50 in time 60 min (17.16–27.78 μM) were 5 times lower than IC50 of both standards (Trolox = 178.33 μM; ASA = 147.47 μM). Based on these results, the presented 1,3,5-triazine derivatives and their analogs have a high potential in the treatment of illnesses caused or related to oxidative stress.

Distillation wastewater, by-products from steam and water distillation as well as raw material used as control of flower heads of Helichrysum italicum were comparative analyzed for content of the biologically active compounds by GC/MS. Acetone exudates, methanol extracts and ethyl acetate fractions obtained after alkaline hydrolyze of the studied materials were received. The three types of extraction products as well as the distillation wastewater were examined for free radical scavenging activity by DPPH assay. Phenol, fatty- and organic acids, sterols, triterpenes, sugars and sugar alcohols were identified. Succinic acid and myo-inositol were identified as main components of distillation wastewater. Hydroxycinnamic acid, caffeic acid and 4(p)-hydroxybenzoic acid were dominant compounds of the ethyl acetate fractions. Triterpenes and fatty acids, sterols and flavonoids are among the main biologically active substances in the methanolic extracts and acetone exudates. The ethyl acetate fractions were found to possess the highest free radical scavenging activity (IC50 < 50 µg/mL). Significant differences in the activity between wastes and raw materials were not found. The results showed that the waste products after distillation of H. italicum contain important biologically active substances and the extracts with high antioxidant activity can be obtained from them.

A new class of inulin derivatives possessing 1,2,3-triazolium charged units by associating “click reaction” with efficient 1,2,3-triazole quaternization were designed and synthesized. The synthesized inulin derivatives possess excellent free radical-scavenging ability.

Free radicals can rapidly and irreversibly oxidize various structures, including unsaturated fatty acids in vegetable oils, which affect the sensory properties. Spectrophotometry is the most widely used method for the determination of free radical scavenging activity (RSA) using 2,2-diphenyl-1-picrylhydrazyl (DPPH). Barrier to the further use of classical analytical methods to analyze biologically active compounds in foodstuffs is that equipment requires high cost and has limited mobility. One of solutions is to replace classical methods, such as spectroscopy, with smartphonebased colorimetry. Huawei P30 Lite smartphone was used for colorimetric detection. The free radical scavenging activity (RSA) in vegetable oil was detected using an application ‘Color Picker’, with image matching algorithm for red, green, and blue (RGB) model. RSA was expressed as percentage and measured by the DPPH method. The aim of the study was to determinate the total free radical scavenging activity with smartphone-based colorimetry. For the data comparison and accuracy spectrophotometer as analytical optical instrument was used. Eleven vegetable oils: sea buckthorn, sunflower, rice, macadamia nut, hemp, corn, grape, linseed, rapeseed, olive and milk thistle oils were selected for analysis. The best results with no significant differences (p>0.05) compared to smartphone-based colorimetry from spectrophotometry were determined using RG values. The poor results were detected by using B value (p<0.05) and were not suitable for determination of RSA. Smartphone-based colorimetry can be used in the determination of the RSA in vegetable oils.

Coffee is a popular beverage all over the world, but spent coffee grounds (SCGs) constituting almost 75% of original beans are usually considered waste and disposed off. The present study analyzed the functionalities of SCG with a view of its reuse in the cosmetic industry. The SCG extraction was carried out by the hydrothermal method. The resultant extracts were tested for its antioxidant capacity, tyrosinase inhibition, and moisturizing ability. LC–MS/MS results showed two major components in SCG extracts, namely, trigonelline and caffeine. Also, the SCG contained total flavonoid contents of 29 ± 4.5 mg quercetin equivalents (QE)/g SCG and total phenolic contents of 9.44 ± 0.90 mg gallic acid equivalents (GAE)/g SCG. Regarding functionality analysis, SCG extracts exhibited reduction capacity of 8.18 ± 0.39 mg vitamin C equivalent (VCE)/g SCG, DPPH free-radical scavenging activity (IC50) of 3.11 mg SCG/mL, ABTS free-radical scavenging activity (IC50) of 13.61 mg SCG/mL, and tyrosinase inhibition capacity (IC50) of 2.23 mg SCG/mL. Moreover, the volatilization rate of the extract solution (37 mg SCG/mL) reduced by 15.9%. These results demonstrate the utility of recycling of SCG and illustrate its potential application in the development of skin care products.

Values of EC50 correlated with %inhibition activity. Based on these results, the presented 1,3,5-triazine analogues have a high potential in the treatment of illnesses caused or related to oxidative stress.
