Bulbs are the main vegetative reproductive organs of Fritillaria meleagris L. In nature, as well as in vitro, they become dormant and require low temperatures for further growth during the next vegetative period. In the present study, using 10 μM of gibberellic acid (GA3), or gibberellin biosynthesis (GA) inhibitors—ancymidol (A) and paclobutrazol (P)—the dynamic changes in soluble sugars, fructose and glucose content, fresh weight and sprouting capacity were investigated. F. meleagris bulbs were cultured on medium with GA3 and GA inhibitors for 1, 2 and 5 weeks at two different temperatures (24 and 7 °C). GA3 improved bulb fresh weight, as well as sprouting percentage at both tested temperatures, compared to the control. The highest fresh weight increase (57.7%) and sprouting rate (29.02%) were achieved when bulbs were grown at 24 °C for 5 weeks. In addition, soluble sugar content was the highest in bulbs grown for 5 weeks on medium supplemented with GA3. The main sugar in fritillary bulbs was glucose, while fructose content was lower. The sensitivity of bulbs to GA inhibitors differed and significantly affected sugar content in bulbs. To our knowledge, this is the first study of the sugar composition in F. meleagris bulbs during breaking of the bulb’s dormancy and its sprouting.

Broccoli (Brassica oleracea L. Botrytis group `Green Duke') seeds were cultured photoautotrophically (without sugar) or photomixotrophically (with sugar) in vitro for 3 weeks at 23 °C and150 μmol·m-2·s-1 photosynthetic photon flux (PPF). In vitro seedlings were stored for 0, 4, 8, or 12 weeks at 5 °C in darkness or under 5 μmol·m-2·s-1 of white (400–800 nm), blue (400–500 nm), or red (600–700 nm) light. Photosynthetic ability and soluble sugar contents were determined after removal from storage. Photomixotrophic seedlings contained approximately five times more soluble sugars than did photoautotrophic seedlings. Dark storage reduced soluble sugars in both photoautotrophic and photomixotrophic plants, but photosynthetic ability was maintained for up to 8 weeks in the latter whereas it decreased in the former. Illumination in storage increased leaf soluble sgars in both photoautotrophic and photomixotrophic seedlings. Soluble sugars in stems decreased during storage regardless of illumination, but remained higher in illuminated seedlings. Red light was more effective in increasing or maintaining leaf and stem soluble sugars than was white or blue light. Regardless of media composition or illumination, storage for more tan 8 weeks resulted in dramatic losses in quality and recovery, as well as photosynthetic ability. Seedlings stored for 12 weeks comletely lost their photosynthetic ability regardless of media composition or illumination. The results suggest that carbohydrate, supplied in the media or through illumination, is essential for maintenance of photosynthetic ability during low-temperature storage for up to 4 or 8 weeks.

Plenty of studies have demonstrated that DIF has an effect on the fruit growth. To study the effects of day and night temperature differences on the strawberry sugar quality, an experiment using climate chambers was conducted. Five different differences between the day and night temperatures (DIF) were set, which were 6 °C (28 °C/22 °C, day/night temperature), 8 °C (29 °C/21 °C), 10 °C (30 °C/20 °C), 12 °C (31 °C/19 °C), 14 °C (32 °C/18 °C). The results showed the following indices peaked with a DIF of 12 °C, including the photosynthesis rate, glucose content, fructose content, sucrose content, soluble sugar content and sugar metabolic enzyme activity. The respiration rate increased with the DIF during the day and decreased with the DIF at night. The root dry weight peaked at a DIF of 10 °C, the stolon dry weight peaked at a DIF of 8 °C and the leaf dry weight peaked at a DIF of 6 °C; however, the fruit dry weight reached maximum values at a DIF of 12 °C. The Grey correlation analysis showed that the most important factor in our experiment affecting the fructose content was the sucrose phosphate synthase; however, for the sucrose, glucose, and soluble sugars, the most important factor was the photosynthesis. We found that a DIF of 12 °C (31 °C/19 °C, day/night temperature) was the most suitable for strawberry growth, especially for the sugar content accumulation.

The decay of seeds is irreversible and at best can only be delayed by applying techniques that reduce the velocity of the metabolic reactions involved. There is little information on the biochemistry of tropical forest tree seeds related to their storability. It was investigated the influence of the composition of lipids and soluble sugars of two storage compartments, the cotyledons and the embryonic axis, of Melanoxylon brauna Schot. (Leguminosae- Caesalpinioideae), a hardwood known as black brauna, seeds stored at 20 ºC for 0, 3, 6, 9 and 12 months (natural ageing) and for 0, 24, 48, 72 and 96 hours at 40 ºC (accelerated ageing). The levels of fatty acids and monosaccharides varied differentially in each of the embryo storage compartments. Changes in oligosaccharide levels were similar for both types of ageing, diminishing in both compartments. Ageing can be attributed to the significant decrease of oligosaccharides and the increase of glucose in both types of ageing and both embryo compartments.

The present study was aimed to evaluate biochemical and phytotochemical of dry leaves of Stevia rebaudiana. Biochemical analysis indicated that Stevia leaves are a good source of carbohydrate and reducing sugar. Different extraction methods were used to prepare four different dry extracts (Extract A, B, C, D). Total soluble sugars and reducing sugars were analyzed for these four dry extracts and commercial Stevia powder. The highest amount of total soluble sugar (477 mg sugar g-1 dry extract) was obtained from extract C and higher amount of reducing sugar (82 mg g-1 dry extract) was obtained from extract D among the extracts. But commercial Stevia powder showed higher total soluble sugar content (754 mg g-1 dry powder) and highest amount of reducing sugar (98 mg g-1 dry extract) than all the extracted dry samples. The extraction process of dry extract C was feasible for the extraction of total soluble sugar. For the phytochemical screening, crude extract was tested for the presence of different chemical groups and presence of alkaloids, phenolic compounds, steroids, tannins, flavonoids, cardiac glycosides and saponins that were identified. The highest amount of total phenolic compounds (92 mg) was recorded from methanolic extract of extraction B. The lowest amount of total phenolic compounds (36 mg) was recorded in ethanolic extract of extraction A. So, Methanol proved as best solvent to extract increased quantity of total phenolic compounds than other solvents.Asian J. Med. Biol. Res. March 2016, 2(1): 121-130

AbstractLarge differences in seed desiccation sensitivity have been observed previously among ten coffee species (Coffea arabica, C. brevipes, C. canephora, C. eugenioides, C. humilis, C. liberica, C. pocsii, C. pseudo-zanguebariae, C. sessiliflora and C.stenophylla). Of these species,C. libericaandC. humiliswere the most sensitive to desiccation andC. pseudozanguebariaethe most tolerant. A study was carried out using the same seed lots to investigate if these differences in desiccation tolerance could be correlated with differences in soluble sugar content. Soluble sugars were extracted from dry seeds and analysed using high performance liquid chromatography. The seed monosaccharide (glucose and fructose) content was very low (1.5 to 2 mg g-1dry weight [dw]) in all species studied. The sucrose content ranged from 33 mg g-1dw inC. libericaseeds to 89 mg g-1dw in seeds ofC. pocsii. Raffinose was detected in the seeds of only five species (C.arabica, C.brevipes, C.humilis, C.sessiliflora, C.stenophylla), among which only three species (C.arabica, C.sessilifloraandC.brevipes) also contained stachyose. Both raffinose and stachyose were present in very low quantities (0.3–1.4 mg g-1dw and 0.1–0.7 mg g-1dw, respectively). Verbascose was never detected. No significant relationship was found between seed desiccation sensitivity and: (i) the sugar content; (ii) the presence/absence of oligosaccharides; and (iii) the oligosaccharide:sucrose ratio.

Abstract Two years experiment was carried out to compare the nutritional composition in bulbs of ten onion cultivars grown in the field conditions. Pyruvic acid, ascorbic acid and soluble sugars were determined in the edible part of onion bulbs. In most cultivars the content of pyruvate as the pungency index was ranged between 3-7 μmol g-1 fresh weight (medium pungency), however, three of them exceeded 7 μmol g-1 fresh weight (high pungency). A higher level of pyruvate corresponded in most cases with higher sugar content but was not correlated with that of ascorbic acid.

SUMMARYThe influence of different drying conditions on the chemical composition, physical properties,in vitroorganic matter degradability and fermentation kinetics of forages was investigated using young and old grass (Lolium perenne) samples (harvested on 15 June and 9 July 1992 at Lelystad, The Netherlands) and young and old maize (Zea mayscv. Scana) stem samples (harvested on 19 August and 30 September 1991 at Lelystad). The samples were either freeze-dried with a maximum sample temperature of 10 °C, dried in a vacuum at 20 °C or air-dried at 30, 50, 70 and 105 °C. The different drying methods had little effect on ash, acid detergent fibre (ADF), acid detergent lignin (ADL), crude fibre and crude protein (CP) contents andin vitrodegradation of the forage samples. However, some effects were found for sugars and phenolic acids. The neutral detergent fibre (NDF) content in protein-rich samples and the fermentation kinetics in rumen fluid differed significantly according to drying method. In samples dried other than by freeze-drying, proteins were bound to the NDF content and in some cases an effect on the amount of soluble sugars was also seen. Physical properties of the samples were determined by differential scanning calorimetry (DSC) and differences were found between freeze-dried materials and those dried at 70 °C. The influence of age on the maize samples was very pronounced, whereas it had little effect on the characteristics of the grass samples, with the exception of a decreased CP content and an increased sugar content after acid hydrolysis.

Rose is the most economically important ornamental plant. However, cold stress seriously affects the survival and regrowth of garden roses in northern regions. Cold acclimation was studied using two genotypes (Rosa wichurana and R. hybrida ‘Yesterday’) selected from a rose breeding program. During the winter season (November to April), the cold hardiness of stems, soluble sugar content, and expression of dehydrins and the related key genes in the soluble sugar metabolism were analyzed. ‘Yesterday’ is more cold-hardy and acclimated faster, reaching its maximum cold hardiness in December. R. wichurana is relatively less cold-hardy, only reaching its maximum cold hardiness in January after prolonged exposure to freezing temperatures. Dehydrin transcripts accumulated significantly during November–January in both genotypes. Soluble sugars are highly involved in cold acclimation, with sucrose and oligosaccharides significantly correlated with cold hardiness. Sucrose occupied the highest proportion of total soluble sugars in both genotypes. During November–January, downregulation of RhSUS was found in both genotypes, while upregulation of RhSPS was observed in ‘Yesterday’ and upregulation of RhINV2 was found in R. wichurana. Oligosaccharides accumulated from November to February and decreased to a significantly low level in April. RhRS6 had a significant upregulation in December in R. wichurana. This study provides insight into the cold acclimation mechanism of roses by combining transcription patterns with metabolite quantification.

This experiment was undertaken to characterize the physiological changes taking place during the petal senescence of Hibiscus syriacus. Five distinctive developmental stages were chronologically suggested. Flower bud dry weight increased almost linearly from Stage I to Stage IV at a rate of ≈15 mg/day. Fresh weight and fresh/dry weight ratio increased much more rapidly between Stage III and Stage IV than during the early stage of development. It showed that petal expansion was partially due to an increased water uptake. The highest osmolality (411 mmol) was found in the fully open flowers. During the subsequent senescence and collapse of the flower, from Stage IV to Stage V, there were a rapid loss of fresh and dry weight and the fall of fresh/dry weight ratio, corresponding to the wilting that characterizes early senescence. A rise in cell sap osmolality coincided with the increase in soluble sugar content and fresh/dry weight ratio, and with the expansion of Hibiscus syriacus petal. Therefore, buds at Stage III, where they are under physiological maturity, might be appropriate to harvest. Hibiscus syriacus flowers showed a small but respiratory peak at Stage IV. The maximum rate of respiration was obtained with fully open flowers (Stage IV), whereas ethylene production remained extremely low until the petals started to open. Ethylene production, ACC synthase, and ACC content increased as the fresh weight of the flowers started to decline. At Stage V, there were a loss of petal fresh weight and a considerable increase in ethylene production (9 nL/g per h). The results of the present study have shown that petal tissue at Stage IV, presenescent stage, was characterized by the increase of soluble sugar and fresh weight, which might be expected to lead to petal expansion and limit turgidity. ABA and the stomata on petal might promote the disorganization.

AbstractStrawberry trees (Arbutus unedoL.) are naturally grown in particular Black Sea and Mediterranean regions of Turkey with great diversity due to continuous seed propagation for centuries. The trees differ in terms of most of the horticultural characteristics. We investigated the phenolic compounds and the biochemical and pomological characteristics of the fruits of eight strawberry tree selections naturally grown in the western part of Turkey. Significant differences were found among the genotypes in terms of their phenolic compounds and their biochemical and pomological characteristics. Among soluble sugars, fructose (11.63 g 100 g−1) was the dominant sugar, followed by glucose (6.10 g 100 g−1) and sucrose (1.44 g 100 g−1) for all the genotypes. Positive correlation was found between fruit weight and soluble sugar content. Malic acid was the major organic acid (0.67-2.33 g 100 g−1), and the second major organic acid in strawberry tree fruits was citric acid (0.25-0.87 g 100 g−1). Vitamin C content was an average of 56.22 g 100 g−1for the eight genotypes. Among phenolic compounds, gallic acid was dominant (1.62-7.29 mg 100 g−1), followed by chlorogenic acid (1.23-3.14 mg 100 g−1), on an average basis.
