This study determined the antimicrobial efficiency of light-activated disinfection (LAD) and photodynamic therapy (PDT) on polymer-infiltrated ceramic network (PICN) material contaminated with three periodontal bacteria and explored if PDT and LAD cause PICN surface alterations. Sixty PICN discs were contaminated with Tannerella forsythia, Porphyromonas gingivalis, and Treponema denticola and randomly divided into five groups (n = 12 samples/each) according to the treatment groups: Group PDT—PDT (630 ± 10 nm diode laser) with methylene blue; Group DL—808 nm diode laser in contact mode without photosensitizer; Group MB–methylene blue without light application; Group CHX—0.12% chlorhexidine digluconate solution and; Group NT—no treatment. Each disc was then placed in tubes containing phosphate buffered saline (PBS) and vortexed for 30 s to remove the remaining bacteria from the discs. A total of 10× serial dilutions were performed followed by plating of 30 μL of suspension on Brucella agar plates. The colony forming units (CFU) were calculated after 72 h. PICN discs with the attached biofilms were used for confocal microscopy investigation for live/dead bacterial viability. A random single sample from each group was selected to study the bacterial adherence and topographical alterations on PICN discs under scanning electron microscope (SEM). The PDT group showed higher reduction for each bacterial species and total counts of bacteria assessed followed by the DL group (p < 0.05). When compared with MB group, the two laser groups were significantly superior (p < 0.05). The MB group did not show significant differences for any bacteria when compared to NT. The bacteria with the CHX group and DL groups appeared dead with few areas of surviving green stained bacteria. The PDT group showed the highest dead cell count (p < 0.05). PDT and DL groups indicate no significant changes on the surface compared to the sterile PICN discs on visual assessment. Photodynamic therapy produced superior periodontal bacteria reduction over the surface of PICN surface. PDT group showed higher reduction for each bacterial species and total counts of bacteria assessed followed by the DL group. Both PDT and DL treatment strategies are effective without producing surface alterations on PICN.

Abstract Background Photodynamic therapy (PDT) is a new option for oral lichen planus (OLP) management; however, there are different opinions on the efficacy of PDT for OLP. The aim of this study was to comprehensively assess the efficacy of PDT in the treatment of OLP and compare PDT with steroid therapy. Methods A systematic review and meta-analysis were conducted to assess the curative effect of PDT. Five electronic databases were searched, PubMed, Web of Science, the Cochrane Library, Embase, and EBSCO up to 1 December, 2019. Random and fixed effects models for pooled estimates calculation were used and the Meta package of R was applied. Results Pooled estimates revealed that, after PDT, the lesion size decreased by 1.53 cm2 (95% confidence interval (CI): 0.71–2.35) after PDT and the partial response (PR) was 0.77 (95% CI: 0.65–0.85). The visual analogue scale (VAS) score decreased by 3.82 (95% CI: 2.80–4.85) and the Thongprasom sign score decreased by 1.33 (95% CI: 0.56–2.10) after PDT. Subgroup analyses revealed that the 5-aminolevulinic acid (5-ALA) was more effective than methylene blue (MB), with a PR of 0.87 (95% CI: 0.80–0.91). The topical use of 5-ALA yielded a better response than gargling methylene blue. In terms of VAS, the diode laser showed a better clinical PR in the treatment of OLP. In terms of changes in lesion size, the efficacy of the semiconductor laser was higher than that of the diode laser. PDT had a similar efficacy to topical corticosteroids, as shown by pooled estimates of five randomised controlled trials with 139 lesions. Conclusion This systematic review indicates that PDT is an effective treatment modality for the management of OLP. PDT is as effective as topical corticosteroid in the treatment of OLP and could be used for cases resistant to steroids or when steroids are contraindicated.

Introduction: Enterococcus faecalis is a common bacterial species in resistant and recurrent infections. It has an active proton pump which prevents its easy inhibition leading to endodontic treatment failure. LASER and photodynamic therapy have been employed lately in improving antimicrobial chemotherapy of localized infections. Aim: To compare disinfection efficacy of 940nm Diode LASER & Photodynamic therapy(PDT) to Conventional method using 3% Sodium hypochlorite in canals of endodontically treated primary teeth. Method: Single sitting Pulpectomy was performed on 60 mandibular first primary molars, divided into 3 groups, disinfected using mentioned modalities. Disinfection potential was compared collecting samples at 3 stages using paper points, cultured and incubated for 24hrs to check change in Colony Forming Units of the bacteria. Results: Data analysis with SPSS version 20 software using Kruskal-Wallis and Mann Whitney-U test. Conclusion: The three groups showed effective disinfection as revealed by the reduction in the colony forming units of the bacteria at baseline and after intervention. However, diode laser when used in conjunction with sodium hypochlorite as a disinfecting modality was found to be the most effective for the eradication of Enterococcus faecalis from infected root canals.

Besides, PDT is as effective as topical corticosteroid in treating OLP and could be used for cases resistant to steroids or when steroids are contraindicated.

AIM: To assess the antibacterial competence of 650 nm diode laser, Methylene Blue (MB) and Silver Nano-Particles (Ag NPs) on Streptococcus mutans in biofilm-induced caries models.MATERIAL AND METHODS: One hundred eighty specimens were prepared and equally divided into 6 groups. One group was untreated (control), and the others were subjected to either MB, laser, Ag NPs, the combination of MB and Laser or MB, laser and Ag NPs.RESULTS: Comparison of the log10 mean Colony Forming Units per millilitre (CFU/ml) values of each of the treated 5 groups and the control group was found statistically significant (P-value < 0.05).The combination of MB, laser and Ag NPs recorded the greatest reduction (95.28%). MB alone represented the least capable (74.09%). The efficiency differences among the Ag NPs treated group; the Laser treated group and the combined MB/Laser treated group were found statistically insignificant.CONCLUSION: The combination of MB, 650 nm diode laser and Ag NPs may be among the highly effective modern antimicrobial therapeutics in dentistry.

Introduction: Photodynamic therapy (PDT) has emerged as a powerful tool for conventional endodontic treatment, capable of eradicating microorganisms present in endodontic infections. Despite this, the use of PDT in the surgical endodontic treatment is not well-known. Case Report: This paper describes the case of a patient with asymptomatic apical periodontitis in a mandibular incisor submitted to surgical endodontic treatment associated with PDT. After conventional procedures, photosensitizer methylene blue (0.01%) was applied to the retrograde cavity for 3 minutes, followed by irradiation with diode laser (100 mW and 660 nm) for 3 minutes. Six months after the procedure, the patient was asymptomatic, and the radiographic examination showed healthy periradicular tissues. Conclusion: The association of PDT with surgical endodontic treatment was effective, suggesting that this therapy may provide additional benefits to patients when compared to the conventional surgical technique.
