Fatty acid (FA) balance is strictly related to human health. The composition of fatty acids in lipid membranes seems to be influenced by diet. Shark liver oil (SLO) supplementation has been widely used recently in the prevention and treatment of human diseases. We analyzed the impact of short-term SLO supplementation on certain biochemical parameters and erythrocyte FA composition in a group of young healthy women. Our results showed that 6 weeks of SLO supplementation led to a significant decrease in C-reactive protein levels in sera and intracellular cholesterol levels in peripheral blood mononuclear cells. SLO supplementation caused a significant increase in the content of the polyunsaturated omega-3 FAs: docosahexaenoic acid, docosapentaenoic acid and α-linolenic acid. In the group of omega-6 FAs, we observed a significant elevation of arachidonic and dihomo-gamma-linoleic acid content. Due to these alterations, the omega-3 index increased significantly from 3.6% (before) to 4.2% (after supplementation). We also observed the impact of SLO supplementation on the membrane fluidity index. The ratio between saturated and unsaturated FAs decreased significantly from 13.1 to 9.9. In conclusion, our results show that even short-term SLO supplementation can improve human erythrocyte fatty acid composition and other parameters that may have health-promoting consequences.

Thirty 7-month-old crossbred lambs (Poll Dorset × Border Leicester × Merino) finished over 5 weeks on either low quality pasture or grain-with-hay on a farm in southern Victoria were assessed for carcass parameters, muscle fat composition and retail colour stability. Lambs on the grain diet had a mix of barley grain (80%) and lentils (20%) at 800 g/head.day (air-dry basis ~ad libitum) with cape weed (Arctotheca calendula) hay available at all times. Lambs under grazing had predominantly rye grass (Lolium perenne) and barley grass (Hordeum leporinum) available ad libitum. Carcass weight tended to be higher (P = 0.14) in grain-fed lambs than in grass-fed lambs, but fatness indicated by GR (total muscle + fat tissue thickness at 11 cm from midline) did not differ between feeding systems. Fatty acid composition was determined in the loin from the forequarter (M. longissimus thoracis) and lumbar (M. longissimus lumborum) regions and from the leg region (M. semimembranosus). This showed that grain-finished lamb had higher muscle fat (P < 0.001) and omega-6 fatty acid (P < 0.001) content. Alpha-linolenic acid, eicosapentaenoic acid (EPA) or total omega-3 fat did not differ (P > 0.05) between feeding groups. Saturated fatty acids were greater (P < 0.01) in both loin sampling sites than the leg. The levels of EPA, docosahexaenoic acid and docosapentaenoic acid were lower (P < 0.01) in the forequarter (9%) or lumbar (11%) sites than the leg sampling site. The distribution pattern of fatty acids across the three sampling sites did not differ between feed types. Retail colour stability determined over 4 days of display (only performed in muscle from the lumbar site), evaluated by the redness (HunterLab a*-value) and metmyoglobin formation (reflectance ratio at 630 : 580-nm wavelengths) was superior for grass-fed lamb compared with short-term grain-finished lamb. The results demonstrate that the health claimable omega-3 fat mainly EPA or total omega-3 fat content in lamb was not altered by short-term grain finishing compared with lamb finished under pasture grazing conditions.

Dietary intake of 18: 2n-6 and 18: 3n-3 may affect endogenous production and incorporation of n-3 long-chain PUFA (LCPUFA) from fish oils (FO). This double-blinded controlled 2 × 2-factorial 8-week intervention investigates the effects of high and low 18: 2n-6 intake in combination with FO-supplementation on tissue fatty acid composition. Healthy young men (n 64) were randomized to capsules with FO or olive oil (control) (4·4 (2·0–5·6) ml/d) and to either sunflower oil and margarine (S/B) or rapeseed oil and a butter spread (R/K) to provide a high or a low 18: 2n-6 intake. Diet was measured by 4-d weighed dietary records at baseline, during and 8 weeks after the intervention and tissue incorporation as fatty acid composition of peripheral blood mononuclear cells (PBMC). The fat intervention gave a mean difference in the 18: 2n-6 intake of 7·3 g/d (95 % CI 4·6, 10·0) and a similar 18: 3n-3 intake in the groups. The R/K groups had a 0·2 % fatty acid (FA%) (95 % CI 0·0, 0·4, P = 0·02) higher content of 22: 5n-3 in the PBMC, a tendency of slightly higher 20: 5n-3 (P = 0·06), but no more 22: 6n-3 (P = 0·83) than the S/B groups. FO effectively raised the PBMC content of all n-3 LCPUFA (P < 0·001). The fat intervention did not markedly influence the effect of FO; the mean PBMC content of n-3 LCPUFA was 10·3 (sem 0·3) FA% in the FO+S/B group and 10·6 (sem 0·2) FA% in the FO+R/K group. In conclusion, increasing the 18: 2n-6 intake did not have any pronounced effect on incorporation of n-3 LCPUFA in PBMC, either alone or with simultaneous FO supplementation.

Measurement of fatty acids in biological fluids and cell membranes including leucocytes from multiple sclerosis patients is inconsistent. The objective of the present study was to investigate the fatty acid composition within the different membrane phospholipid fractions in peripheral blood mononuclear cells in multiple sclerosis patients, and correlate with severity of neurological outcome as measured by the Kurtzke Expanded Disability Status Scale and Functional System Scores. The fatty acid composition of phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, sphingomyelin and phosphatidylinositol phospholipids in the peripheral blood mononuclear cells of twenty-six multiple sclerosis and twenty-five control subjects were measured by GC, and C-reactive protein was measured in all subjects. The elongation product of 20 : 4n-6, 22 : 4n-6, was significantly decreased in membrane phosphatidylethanolamine and phosphatidylserine in multiple sclerosis patients (P = 0·01 and P = 0·03 respectively), and correlated inversely with severity of disease and C-reactive protein. Also an inverse correlation was observed between the C-reactive protein and membrane phosphatidylcholine and phosphatidylserine 20 : 4n-6. Cultural and ethnic differences, as well as dietary variability, especially in a diseased state have been implicated in the differences observed in the fatty acid composition in peripheral blood mononuclear cell membranes of patients with multiple sclerosis. The present results suggest that the disease state may in part explain the reported inconsistencies in fatty acid levels in multiple sclerosis patients.

Once-abundant aerial insectivores, such as the Tree Swallow (Tachycineta bicolor), have declined steadily in the past several decades, making it imperative to understand all aspects of their ecology. Aerial insectivores forage on a mixture of aquatic and terrestrial insects that differ in fatty acid composition, specifically long-chain omega-3 polyunsaturated fatty acid (LCPUFA) content. Aquatic insects contain high levels of both LCPUFA and their precursor omega-3 PUFA, alpha-linolenic acid (ALA), whereas terrestrial insects contain much lower levels of both. We manipulated both the quantity and quality of food for Tree Swallow chicks in a full factorial design. Diets were either high-LCPUFA or low in LCPUFA but high in ALA, allowing us to separate the effects of direct LCPUFA in diet from the ability of Tree Swallows to convert their precursor, ALA, into LCPUFA. We found that fatty acid composition was more important for Tree Swallow chick performance than food quantity. On high-LCPUFA diets, chicks grew faster, were in better condition, and had greater immunocompetence and lower basal metabolic rates compared with chicks on both low LCPUFA diets. Increasing the quantity of high-LCPUFA diets resulted in improvements to all metrics of performance while increasing the quantity of low-LCPUFA diets only resulted in greater immunocompetence and lower metabolic rates. Chicks preferentially retained LCPUFA in brain and muscle when both food quantity and LCPUFA were limited. Our work suggests that fatty acid composition is an important dimension of aerial insectivore nutritional ecology and reinforces the importance of high-quality aquatic habitat for these declining birds.
