As part of our ongoing research on phytoestrogens, we investigated the phytochemical profile and estrogen-like activities of eight extracts from the aerial parts of four Genista species of Greek flora using estrogen-responsive cell lines. Ethyl acetate and methanolic extracts of G. acanthoclada, G. depressa,G. hassertiana, and G. millii were obtained with accelerated solvent extraction and their phytochemical profiles were compared using ultra-high-performance liquid chromatography-high-resolution mass spectrometry (uHPLC-HRMS). Fourteen isoflavonoids, previously isolated from G. halacsyi, were used as reference standards for their identification in the extracts. Thirteen isoflavonoids were detected in both extracts of G. acanthoclada and G. hassertiana, while fewer and far fewer were detected in G. millii and G. depressa, respectively. The ethyl acetate extracts of G. hassertiana and G. acanthoclada displayed 2.45- and 1.79-fold higher, respectively, estrogen-like agonist activity in Ishikawa cells compared to MCF-7 cells at pharmacologically relevant concentrations. Both these extracts, but not that of G. depressa, contained mono- and di-O-β-d-glucosides of genistein as well as the aglycone, all three of which are known to display full estrogen-like activity at lower-than-micromolar concentrations. The possibility of using preparations rich in G. hassertiana and/or G. acanthoclada extracts as a potentially safer substitute for low-dose vaginal estrogen for menopausal symptoms is discussed.

Spondias species have been used in traditional medicine for different human ailments. In this study, the effect of different solvents (ethyl acetate, methanol, and water) and extraction methods (infusion, maceration, and Soxhlet extraction) on the enzyme inhibitory activity against acetylcholinesterase, butyrylcholinesterase, tyrosinase, α-amylase, α-glucosidase, and antioxidant properties of S. mombin and S. dulcis leaves and stem bark were evaluated. Ultra-high-performance liquid chromatography–high resolution mass spectrometry (UHPLC-HRMS) yield in the identification and/or annotation of 98 compounds showing that the main secondary metabolites of the plant are gallic and ellagic acids and their derivatives, ellagitannins, hydroxybenzoic, hydroxycinnamic, acylquinic acids and flavonols, flavanones, and flavanonols. The leaves infusion of both Spondias species showed highest inhibition against acetylcholinesterase (AChE) (10.10 and 10.45 mg galantamine equivalent (GALAE)/g, for S. dulcis and S. mombin, respectively). The ethyl acetate extracts of the stem bark of S. mombin and S. dulcis actively inhibited α-glucosidase. Methanolic extracts of the leaves and stem bark exhibited highest tyrosinase inhibitory action. Antioxidant activity and higher levels of phenolics were observed for the methanolic extracts of Spondias. The results suggested that the Spondias species could be considered as natural phyto-therapeutic agents in medicinal and cosmeceutical applications.

The aim of present study was to assess phytochemical profile and antibacterial activities of some selected bacterial species. All the bacteria species tested showed higher inhibition zone varying from 20mm-27mm with 100mg/ml of ethyl acetate, methanol, Hexane and chloroform respectively. Moreover, the phytochemical profile revealed the presence of steroids, alkaloids, flavonoids and tannin in the extracts. Thus, result of this study showed that Garlic bulb extract could be a good candidate for production of synthetic antibacterial drug for the treatment of infections caused by the organisms used in this study.

During the present study, an endeavor has been made to produce a simple, rapid, and simultaneous method for determination of phenolic compounds by using high-performance liquid chromatography in aerial parts of Leptadenia pyrotechnica (Forssk.) Decne. collected from the Indian Thar Desert. The optimized process was used for the quantification of ten phenolic compounds. The chromatographic separation was accomplished on an Atlantis T3 column at 25°C with isocratic elution. A mixture of acetonitrile and water was used as the mobile phase at a flow rate of 0.8 mL/min. The linear regression examination data for the calibration plots displayed a good linear relationship with r2 > 0.999 in the concentration range of 2–20 µL. In the methanolic extracts of the whole plant of L. pyrotechnica, the content of caffeic acid (3.3%) was reported to be the highest concentration.

Foliage of jujube (Ziziphus jujuba Mill.) as a byproduct of agriculture, is a traditional nutraceutical material in China. Previous studies have shown that it is a rich resource of polyphenols. However, information on its complete phenolic profile and the difference between cultivars is still limited. This study investigated and compared phytochemical profiles of leaves of 66 Chinese jujube cultivars. Forty-two compounds, including 22 flavonols, two flavanols, one flavanone, 13 derivatives of phenolic acids, three simple acids, and one unknown hexoside were identified/tentatively identified using high-performance liquid chromatography (HPLC) coupled with high-resolution mass spectrometry. Eight major flavonols were quantified by HPLC coupled with an ultraviolet (UV) detector. The contents of total flavonoids ranged from 2.6–25.1 mg/g dry weight (DW). Differences between cultivars were analyzed by hierarchical cluster analysis (HCA) and principal component analysis (PCA). This study presents a systematic study on the phenolic compounds in Chinese jujube leaves of different cultivars.

In this work, we report the phenolic composition and bioactivity of the aerial parts of three species of Sarcocapnos (S. enneaphylla, S. pulcherrima, and S. saetabensis) to study their potential as sources of bioactive compounds to revalorize them and contribute to the conservation of these plant species. Samples were collected in different locations in the province of Jaén (southeast of Spain), and qualitative and quantitative analyses of phenolic compounds were performed by high-performance liquid chromatography with diode array and mass spectrometry detection. S. enneaphylla presented the highest concentration of phenolic compounds (58 mg/g DE). The most abundant compound in S. enneaphylla and S. saetabensis was rutin (35 mg/g DE and 11.7 mg/g DE, respectively), whereas isorhamnetin-O-rutinoside was dominant in S. pulcherrima (11.5 mg/g DE). Several assays were performed to evaluate the potential bioactivity of the three species of Sarcocapnos. These assays included antioxidant and radical scavenging (ABTS and DPPH), reducing power (CUPRAC and FRAP), phosphomolybdenum and metal chelating, and enzyme inhibitory activity (acetylcholinesterase, amylase, butyrylcholinesterase, glucosidase, and tyrosinase). In general, all methanolic extracts presented the highest phenolic and flavonoid contents, as well as the highest radical scavenging, antioxidant, and enzyme inhibitory properties. This relationship between phenolics and bioactivity was confirmed by multivariate analysis.

AbstractAn ultra-high-performance liquid chromatography – high-resolution mass spectrometry profiling method was used for a comprehensive study of flavonoid and saponin-rich fractions from the aerial parts of wild spinach (Chenopodium bonus-henricus L.). Thirty-six compounds, respectively, 22 saponins of eight sapogenins (phytolaccagenin, bayogenin, medicagenic acid, 2β-hydroxygypsogenin, 2β-hydroxyoleanoic acid, 2-hydroxy-30-nor-gypsogenin, 2-hydroxyakebonic acid, and akebonic acid) together with 12 flavonoid glycosides of 6-methoxykaempferol, isorhamnetin, patuletin, spinacetin as well as two ecdysteroids (20-hydroxyecdysone and polypodine B) were detected. The occurrence of sapogenins 2-hydroxy-30-nor-gypsogenin, 2-hydroxyakebonic acid, and akebonic acid in the Chenopodium genus is reported here for the first time. The flavonoid and saponin-rich fractions showed in vitro hepatoprotective and antioxidant activity comparable to those of flavonoid complex silymarin (60 μg/mL) in a model of metabolic bioactivation, induced by CCl4. All tested fractions, compared to silymarin, significantly reduced the cellular damage caused by CCl4 in rat hepatocytes, preserved cell viability and GSH level, decreased LDH leakage, and reduced lipid damage. The results showed that saponin-rich fractions F3A and F3B possessed better hepatoprotective activity than flavonoid-rich fractions (F2A and F2B). The most active was fraction F3B and this is probably due to the synergism between the saponins and some acylated flavonol glycosides found there.

In the current study, Achillea santolinoides and Achillea aleppica aeral parts and root were extracted with ethyl acetate, methanol, and water. Detailed phytochemical profiles were obtained using UHPLC-MS, yielding the identification of hydroxybenzoic and hydroxycinnamic acids, phenolic acid glycosides and sugar esters, acylquinic acids, O-glycosyl flavones and flavonols, and flavonoid aglycons, among others. The antioxidant properties and enzyme inhibitory activities of the extracts were assayed with in vitro tests. The phenolic content of the water extracts was significantly higher as compared to the ethyl acetate and methanol ones. A. aleppica aerial parts methanol extract possessed highest flavonoid content (49.18 mg rutin equivalent/g). Antioxidant properties assessment revealed that the methanol extract of A. santolinoides roots actively scavenged DPPH (54.11 mg TE/g) and ABTS radicals (112.53 mg TE/g) and possessed highest reducing potential (183.55 and 129.92 mg TE/g, for CUPRAC and FRAP, respectively). The ethyl acetate extracts of aerial parts and roots of both species showed highest inhibition against BuCHE (6.07–6.76 mg GALAE/g). The ethyl acetate extract of A.santolinoides aerial part showed highest inhibition against tyrosinase (73.00 mg KAE/g). These results showed that the tested Achillea species might represent novel phytotherapeutic avenues for the management of Alzheimer’s disease and epidermal hyperpigmentation conditions, which are both associated with oxidative stress. This paper could shed light into future potential industrial applications using the tested Achillea species.

Abstract “Nanicão Corupá” (Musa acuminata) comes from Southern Brazil. The tropical climate in the region provides unique characteristics, including a sweeter flavor. This difference resulted in a Geographical Indication Recognition and Designation of Origin, recognized by the National Institute of Industrial Property (INPI) in Brazil. Considering that “Nanicão Corupá” has some peculiarities related to the climate and there are no studies evaluating this banana cultivars, the purpose of the present study was to investigate the qualitative and quantitative phenolic composition of the aerial parts of “Nanicão Corupá” by HPLC-ESI-MS/MS in comparison to 46 commercial standards of phenolic compounds. Aerial parts (flower, leaves, fruit and stem) of “Nanicão Corupá” were collected and macerated in methanolic extracts, which were partitioned with solvents of different polarities (dichloromethane and ethyl acetate). The HPLC-ESI-MS/MS analysis was performed using the sample pre-treatment, chromatographic and mass spectrometer parameters. Results demonstrated that a total of 11 phenolic compounds were identified in the analyzed samples. The majority of compounds was identified in the ethyl acetate fraction (BFEF) of banana flowers: rutin (36.06 ± 0.23) and isoquercetin (28.83 ± 5). The compounds isoquercetin, naringerin and myricitrin were identified for the first time in the Musa genus.

AbstractAchillea moschata Wulfen, which grows in the Alps, is extensively used by local people for its medicinal properties. Two studied samples were collected, at the flowering stage, in Val Mustair (Switzerland) and Valchiavenna (Italy), respectively. The aerial parts were defatted with petroleum ether (PET) and successively extracted with dichloromethane (DCM) and methanol (MeOH). High-performance liquid chromatography and electrospray ionization-tandem mass spectrometry analyses of the methanolic extracts evidenced that flavonoids were the predominant compounds compared to phenolic acids in both samples (89.5 vs. 33.0 μg/mg DW in A. moschata Valchiavenna and 82.5 vs. 40.0 μg/mg DW in A. moschata Val Mustair). Among flavonoid derivatives, luteolin and apigenin were the predominant aglycones, free and glycosilated. The A. moschata Valchiavenna extract was characterized by apigenin as the main compound (60.4 μg/mg DW), while A. moschata Val Mustair was characterized by its derivative apigenin 7-O-glucoside (44.7 μg/mg DW). The antioxidant activity of all the obtained extracts was tested by the DPPH (2,2-diphenyl-picryl hydrazyl) and ABTS (2,21-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) methods, which showed their increasing scavenger capacity in relation to extract polarity (PET extract < DCM extract < MeOH extract). The extracts were also investigated against three Gram-positive (Bacillus cereus, Enterococcus faecalis and Staphylococcus aureus) and three Gram-negative (Escherichia coli, Proteus mirabilis and Pseudomonas aeruginosa) bacterial species using the disc diffusion assay. DMC and PET were the most active extracts (inhibition diameter: ≥12 mm).

The present study was aimed to validate the phenolic, flavonoids, alkaloids and tannins profile of Aspleniuma ethiopicum (Burm. f.) Becherer methanolic extracts using HPTLC (High-performance thin-layer chromatography). The chromatographic separation was performed using the standard method. The separation methodology was optimized and phytoconstituents of A. aethiopicum methanolic extracts were separated. The methanolic extract of A. aethiopicum showed several bands identified as known phenolic compounds. The obtained flavonoids profiles were identified. HPTLC alkaloids profile of A. aethiopicum revealed 11 types of alkaloids. The tannin profile of methanolic extracts of A. aethiopicum demonstrated 11 different types of tannins. The observed profiles will be used as phytomarker to identify the chemical constituents of A. aethiopicum methanolic extracts. These profiles will act as a fingerprint of A. aethiopicum and help us to distinguish from its adulterants. The observed profile will help us to identify the crude drugs and improve the therapeutic potentials of A. aethiopicum.
